Dietary-suppression of hepatic lipogenic enzyme expression in intact male transgenic mice

AIM:To study,in intact male transgenic mice,the effects of three diets based on olive oil and olive oil diet supplemented with lovastatin and orlistat on hepatic lipogenic enzymes expression,considered markers of cell ***:Forty ApcMin/+mice were randomly divided into 4 groups and fed for 10 wk:olive oil(OO)group,n=10 animals received a diet with olive oil 12%;olive oil plus lovastatin(LOVA)group,n=10 animals received the same diet with olive oil supplemented with lovastatin 5 mg/kg;olive oil plus orlistat(OR)group,n=10 animals fed the diet with olive oil supplemented with orlistat 50 mg/kg and SD group,n=10 animals fed a standard *** activity of lipogenic enzymes and their gene expression were evaluated by radiomet-ric and real-time reverse transcription-polymerase chain reaction assay,***:After 10 wk of dietary treatment,the body weight was no different among animal groups(21.3±3.1 g for standard group,22.1±3.6 g for OO group,22.0±3.2 g for LOVA group and 20.7±3.4 g for OR group,data expressed as mean±SD),observing a generalized well-being in all *** the dietary managed treated groups presented significantly reduced hepatic levels of fatty acid synthase,farnesyl pyrophosphate synthase and 3-hydroxyl-3-methyl-glutaryl CoA reductase activity and gene expression when compared with the mice fed the standard *** evaluate cell proliferation in the liver of treated mice,the levels of cyclin E mRNA have been measured,demonstrating a significant reduction of cyclin E gene expression in all treated *** of reduced hepatic cell proliferation was present overall in OO group ***:We confirm the role of lipogenic enzymes as markers of cell proliferation,suggesting that appropriate dietary management alone or with drugs can be a feasible approach to counteract hepatic cell proliferation in mice.

Wilson's disease: Prospective developments towards new therapies

Wilson's disease(WD) is an autosomal recessive disorder of copper metabolism, caused by mutations in the ATP7 B gene. A clear demand for novel WD treatment strategies has emerged. Although therapies using zinc salts and copper chelators can effectively cure WD, these drugs exhibit limitations in a substantial pool of WD patients who develop intolerance and/or severe side effects. Several lines of research have indicated intriguing potential for novel strategies and targets for development of new therapies. Here, we review these new approaches, which comprise correction of ATP7 B mutants and discovery of new compounds that circumvent ATP7B-deficiency, as well as cell and gene therapies. We also discuss whether and when these new therapeutic strategies will be translated into clinical use, according to the key requirements for clinical trials that remain to be met. Finally, we discuss the hope for the current rapidly developing research on molecular mechanisms underlying WD pathogenesis and for the related potential therapeutic targets to provide a solid foundation for the next generation of WD therapies that may lead to an effective, tolerable and safe cure.

Obese children with fatty liver: Between reality and disease mongering

Following the current epidemic of obesity, the worldwide prevalence of nonalcoholic fatty liver disease(NAFLD)has increased with potential serious health implications. While it is established that in adults NAFLD can progress to end-stage liver disease in many cases, the risk of progression during childhood is less well defined. Since most obese children are not adherent to lifestyle modifications and hypocaloric diets, there is a growing number of studies on pharmacological interventions with the risk of disease mongering, the practice of widening the boundaries of illness in order to expand the markets for treatment. Here, we propose a critical appraisal of the best available evidence about long-term course of pediatric NAFLD and efficacy of treatments other than hypocaloric diet and physical exercise. As a result, the number of NAFLD children with a poor outcome is small in spite of the alarming tones used in some papers; large-scale longitudinal studies with longterm follow-up of pediatric NAFLD patients are lacking; the studies on ancillary pharmacological interventions have been performed in few patients with inconclusive and conflicting results.

Simulating the fate of indigenous antibiotic resistant bacteria in a mild slope wastewater polluted stream

The fate of indigenous surface-water and wastewater antibiotic resistant bacteria in a mild slope stream simulated through a hydraulic channel was investigated in outdoor *** effect of（i） natural（dark） decay,（ii） sunlight,（iii） cloudy cover,（iv） adsorption to the sediment,（v） hydraulic conditions,（vi） discharge of urban wastewater treatment plant（UWTP）effluent and（vii） bacterial species（presumptive Escherichia coli and enterococci） was ***-life time（T1/2） of E. coli under sunlight was in the range 6.48–27.7 min（initial bacterial concentration of 10^5 CFU/mL） depending on hydraulic and sunlight conditions. E. coli inactivation was quite similar in sunny and cloudy day experiments in the early 2 hr, despite of the light intensity gradient was in the range of 15–59 W/m^2; but subsequently the inactivation rate decreased in the cloudy day experiment（T1/2= 23.0 min） compared to sunny day（T1/2= 17.4 min）. The adsorption of bacterial cells to the sediment（biofilm） increased in the first hour and then was quite stable for the remaining experimental time. Finally, when the discharge of an UWTP effluent in the stream was simulated, the proportion of indigenous antibiotic resistant E. coli and enterococci was found to increase as the exposure time increased, thus showing a higher resistance to solar inactivation compared to the respective total populations.

Embodied Simulation and a Few Remarks on the Philosophy of Language in Italy

The debate on the state of the art of philosophy in Italy shows many differences from the three main currents of contemporary philosophy, namely deconstructionism, critical theory, and analytic philosophy. On one hand, the philosophical stance towards language, and on the other hand, the strong separation between science and philosophy are the distinctive features of the Italian thought. In particular, philosophy of language shows a fragmented framework in which current researches are an example for future studies on language and human nature. The basic idea behind the Philosophy of Language in Italy is that language has a biological and social nature. The aim of this paper is to give an account of the range of ＂bio＂ through a dialogue between philosophy and neuroscience. I will review the Italian research on mirror neurons with particular attention to the developments in embodied simulation proposed by Vittorio Gallese. I will underline the philosophical aspects of this neuroscientific theory, providing some remarks on intersubjectivity, intentions, and language. Finally, I will suggest future joint studies in neuroscience and the philosophy of language.

Understanding IT Artefacts with Language

Every day we experience relationships with artefacts, which describe material objects made by humans in order to reach a goal and exploit the human feature to plan ahead. Artefacts bring together cognitive evolution and technical enhancement. Although artefacts are conceived as technical, we are now facing a relationship with Information Technology （IT） artefacts. IT artefacts include both hardware and software, as a two-sided entity. The definition of IT artefact corresponds with the Saussurean linguistic sign： a two-sided entity constituted by the signifier （hardware） and the signified （software）. I claim that IT artefacts share this ontological trait with the linguistic sign. I will show that IT artefacts are the result of design and planning, while language--which is an institution--is not the fabrication of one human but linked to collective human activity.

Insights From the Philosophical Reception of Michael Polanyi in Italy

Is there a national philosophy?It is hard to answer the question definitively,beyond mainstream philosophical currents and a supposed particular *** an invisible boundary,today,academic research is also defined by its international and interdisciplinary *** it is possible to argue about certain specificities,the interaction between different national traditions and distant scientific areas can be fruitful(and these parameters are also required in the case of research funding).One can take an example of the reception of Michael Polanyi’s epistemology in Italy,starting from a brief reference to the Italian tradition and its renewed interest thanks to the Italian ***,in this paper,I will try to consider Polanyi’s epistemology through his relations with Italian intellectuals and the dissemination of his writings in *** the end,I will verify the potential contribution of Polanyian personal epistemology in three philosophical and social issues addressed in Italy.

Pediatric chronic liver diseases encompass a wide spectrum of hepatopathies with a relevant health medical and financial burden(1,2).In addition to inherited disorders(e.g.,Alagille syndrome,metabolic disorders,etc.),there are numerous chronic liver conditions including metabolic dysfunction-associated steatotic liver disease(MASLD)and autoimmune liver diseases affecting morbidity,quality of life,and life expectancy of these young patients(3,4).To complicate matters,the exact prevalence of childhood chronic liver diseases is still unknown since their asymptomatic course especially in the early stages but increasing rates are reported worldwide(1,5,6).Moreover,management of these chronic conditions represents a great challenge for clinicians as it requires a lifelong multi-disciplinary approach(1).Worthy of note,advances in the knowledge of pathophysiological mechanisms underlying liver diseases greatly improved their natural history with increased survival rates into adulthood(1,3).Given that,adult hepatologists are increasingly dealing with most of these conditions(3).

Understanding Reputational Crisis： Evidence From the European Banking Sector

This paper presents a comparative qualitative analysis of reputational crisis of four European banks, and explores how in recent years these companies have faced the manifestation of reputational risk. To achieve this, the research follows three related steps：（1） to carry out a review of the literature on reputational risk in the banking sector aimed to identify the relationships between causes, effects, stakeholders, and key qualitative-quantitative variables involved during the reputational crisis of a bank; （2） to propose a conceptual framework for management of reputational risk （and reputational crisis） in banking; （3） to test this framework with the results of an empirical analysis, carried out through the observation of key variables of some reputational crisis of intemational banks. The main results show that：（1） the banks are not yet prepared to accurately manage a reputational crisis or to prevent them; （2） the reputational crisis is determined by several internal and external factors; （3） the conduct of the managers and the corporate communication are very important to overcome a reputational crisis. Finally, this research provides indications that will help banks to better manage their corporate reputation and prevent reputational crisis.

BACKGROUND Carbapenem-resistant Enterobacteriaceae has become a significant public health concern as hospital outbreaks are now being frequently reported and these organisms are becoming difficult to treat with the available *** SUMMARY An outbreak of VIM-producing Serratia marcescens occurred over a period of 11 wk(August,1 to October,18)in patients admitted to the adult polyvalent intensive care unit of the University of Campania“Luigi Vanvitelli”located in *** episodes occurred in three patients(two patients infected,and one patient colonized).All the strains revealed the production of *** After three decades of carbapenem antibiotics use,the emergence of carbapenemresistance in Enterobacteriaceae has become a significant concern and a stricter control to preserve its clinical application is *** is,to our knowledge,the first outbreak of VIM-producing Serratia marcescens in *** policies must be implemented to avoid future outbreaks.
