Metabolic basis of solute carrier transporters in treatment of type 2 diabetes mellitus

Solute carriers(SLCs) constitute the largest superfamily of membrane transporter *** transporters, present in various SLC families, play a vital role in energy metabolism by facilitating the transport of diverse substances, including glucose, fatty acids, amino acids, nucleotides, and *** actively participate in the regulation of glucose metabolism at various steps, such as glucose uptake(e.g., SLC2A4/GLUT4), glucose reabsorption(e.g., SLC5A2/SGLT2), thermogenesis(e.g., SLC25A7/UCP-1), and ATP production(e.g., SLC25A4/ANT1 and SLC25A5/ANT2). The activities of these transporters contribute to the pathogenesis of type 2 diabetes mellitus(T2DM). Notably, SLC5A2 has emerged as a valid drug target for T2DM due to its role in renal glucose reabsorption, leading to groundbreaking advancements in diabetes drug discovery. Alongside SLC5A2, multiple families of SLC transporters involved in the regulation of glucose homeostasis hold potential applications for T2DM therapy. SLCs also impact drug metabolism of diabetic medicines through gene polymorphisms, such as rosiglitazone(SLCO1B1/OATP1B1) and metformin(SLC22A1-3/OCT1-3 and SLC47A1, 2/MATE1, 2). By consolidating insights into the biological activities and clinical relevance of SLC transporters in T2DM, this review offers a comprehensive update on their roles in controlling glucose metabolism as potential drug targets.

Effectiveness of ivermectin mass drug administration in the control of soil-transmitted helminth infections in endemic populations: a systematic review and meta-analysis

Background Current soil-transmitted helminth(STH)control guidelines endorse the use of albendazole or meben-dazole for school-based targeted preventive chemotherapy(PC),yet their reduced efficacy against Strongyloides sterc-oralis and Trichuris trichiura presents significant *** evidence indicates that community-wide PC[or mass drug administration(MDA)]using ivermectin,commonly used in other neglected tropical disease(NTD)control programs,may play an important role in controlling these *** conducted a systematic review and meta-analysis to evaluate the effectiveness of ivermectin PC in reducing STH prevalence in endemic *** We searched Pubmed,EMBASE,and Web of Science on February 14,2023,for studies that investigated the effectiveness of ivermectin PC,either alone or in combination with other anthelmintic drugs,on STH infec-tions,and provided a measure of STH prevalence before and after *** calculated pooled prevalence reductions for each STH using random-effects *** protocol is available on PROSPERO(registration number CRD42023401219).Results A total of 21 were eligible for the systematic review,of which 15 were eligible for *** studies delivered ivermectin through *** pooled prevalence reduction of *** following MDA with ivermec-tin alone was 84.49%(95%CI:54.96-94.66)across five studies and 81.37%(95% CI:61.62-90.96)across seven studies with or without *** prevalence reduction of *** was 49.93%(95%CI:18.23-69.34)across five studies with ivermectin alone,and 89.40%(95%CI:73.66-95.73)across three studies with the addition of *** was high heterogeneity for all syntheses(I^(2)>65%).Conclusions This study underscores the key role of ivermectin-based MDA in addressing limitations in current global STH guidelines in terms of limited efficacy against *** and *** on these findings,revising inter-national STH guidelines to include i

Current strategies for drug discovery research mainly focus on molecular targets,in which structure biology plays a central role,such as the detailed mechanism of a drug-target *** principle of this approach is the presumed molecular mechanism or genetic background of the disease being *** this approach has brought progress in understanding the mode of action for some drugs,most prevalent illnesses-especially chronic diseases-are multifactorial and present with a group of symptoms,as is often the case in cardiovascular diseases,metabolic disorders,cancers,and so *** newly emerging term“multi-morbidity”(i.e.,multiple co-occurring diseases)describes a grand challenge for this strategy,and successful drug intervention for multimorbidity should be multifaceted[1].So,our research team has shifted its attention to a new direction that views the disease as a whole instead of a single-molecule abnormality,and focusing on drugs with multiple targets.

Self-actuating protection mechanisms for safer lithium-ion batteries

Safety issue is still a problem nowadays for the large-scale application of lithium-ion batteries(LIBs)in electric vehicles and energy storage *** unsafe behaviors of LIBs arise from the thermal run-away,which is intrinsically triggered by the overcharging and *** improve the safety of LIBs,various protection strategies based on self-actuating reaction control mechanisms(SRCMs)have been proposed,including redox shuttle,polymerizable monomer additive,potential-sensitive separator,thermal shutdown separator,positive-temperature-coefficient electrode,thermally polymerizable addi-tive,and reversible thermal phase transition *** build-in protection mechanisms,these meth-ods can sensitively detect either the temperature change inside battery or the potential change of the electrode,and spontaneously shut down the electrode reaction at risky conditions,thus preventing the battery from going into thermal *** their advantages in enhancing the intrinsic safety of LIBs,this paper overviews the research progresses of SRCMs after a brief introduction of thermal runaway mechanism and limitations of conventional thermal runaway mitigating *** importantly,the current states and issues,key challenges,and future developing trends of SRCTs are also discussed and outlined from the viewpoint of practical application,aiming at providing insights and guidance for developing more effective SRCMs for LIBs.

Perpendicular-Cutdepth:Perpendicular Direction Depth Cutting Data Augmentation Method

Depth estimation is an important task in computer *** data at scale for monocular depth estimation is challenging,as this task requires simultaneously capturing RGB images and depth ***,data augmentation is crucial for this *** data augmentationmethods often employ pixel-wise transformations,whichmay inadvertently disrupt edge *** this paper,we propose a data augmentationmethod formonocular depth estimation,which we refer to as the Perpendicular-Cutdepth *** method involves cutting realworld depth maps along perpendicular directions and pasting them onto input images,thereby diversifying the data without compromising edge *** validate the effectiveness of the algorithm,we compared it with existing convolutional neural network(CNN)against the current mainstream data augmentation ***,to verify the algorithm’s applicability to Transformer networks,we designed an encoder-decoder network structure based on Transformer to assess the generalization of our proposed *** results demonstrate that,in the field of monocular depth estimation,our proposed method,Perpendicular-Cutdepth,outperforms traditional data *** the indoor dataset NYU,our method increases accuracy from0.900 to 0.907 and reduces the error rate from0.357 to *** the outdoor dataset KITTI,our method improves accuracy from 0.9638 to 0.9642 and decreases the error rate from 0.060 to 0.0598.

Bridged polycyclic lactams are important structural units in organic functional materials,natural products,and pharmaceuticals.A flexible and efficient anion cascade reaction was developed for the preparation of bridged polycyclic lactams from readily available malonamides and 1,4‑*** highly substituted bridged polycyclic lactams were synthesized in good to excellent yields by tandem nucleophilic sequences in the presence of t BuOK in commercially available EtOH solvent at 60℃.Notably,the simple reactions can be run on a gram ***,bis-Michael addition reaction and hemiaminalization reactions are involved in the tandem transformation.

Understanding Surface Water-Groundwater Conversion Relationship and Associated Hydrogeochemistry Evolution in the Upper Reaches of Luan River Basin, North China

Luan River is the main water source in Beijing-Tianjin-Hebei region,northern China,where the groundwater system is vulnerable and pollution issue is *** is significant for regional groundwater resources protection to identify the hydrogeochemistry evolution and affecting factors along flow direction occurred in the upper reaches,especially the surface water-groundwater(SWGW)conversion *** this study,recharge,conversion and geochemistry evolution of SW and GW were elucidated based on physical-hydrochemical indicators and stable isotopes in 36 GW samples and 20 SW samples,which were collected in July 2019 and July *** factor analysis was further utilized to determine the main factors responsible for regional hydrogeochemical *** indicate that GW recharged SW in plateau area,and SW and GW recharged each other in typical Alpine valley *** hydrochemical types are HCO3-Ca·Mg and HCO3-Ca,and the hydrochemical evolution is dominated by weathering of silicate and carbonate *** cation exchange adsorption has minor impact on groundwater *** rise of SO42-and NO3-contents in groundwater is related to industrial and agricultural *** main controlling factors of SW hydrochemical components included recharge from groundwater,industrial and mining activities,explaining 90.04%of data ***,water-rock interaction,agricultural and domestic sewage are responsible for GW quality,accounting for 83.38%.
