Polyphosphate and associated enzymes as global regulators of stress response and virulence in Campylobacter jejuni

Campylobacter jejuni(C. jejuni),a Gram-negative microaerophilic bacterium,is a predominant cause of bacterial foodborne gastroenteritis in humans worldwide. Despite its importance as a major foodborne pathogen,our understanding of the molecular mechanisms underlying C. jejuni stress survival and pathogenesis is limited. Inorganic polyphosphate(poly P) has been shown to play significant roles in bacterial resistance to stress and virulence in many pathogenic bacteria. C. jejuni contains the complete repertoire of enzymes required for poly P metabolism. Recent work in our laboratory and others have demonstrated that poly P controls a plethora of C. jejuni properties that impact its ability to survive in the environment as well as to colonize/infect mammalian hosts. This review article summarizes the current literature on the role of poly P in C. jejuni stress survival and virulence and discusses on how poly P-related enzymes can be exploited for therapeutic/prevention purposes. Additionally,the review article identifies potential areas for future investigation that would enhance our understanding of the role of poly P in C. jejuni and other bacteria,which ultimately would facilitate design of effective therapeutic/preventive strategies to reduce not only the burden of C. jejuni-caused foodborne infections but also of other bacterial infections in humans.

西瓜嗜酸菌（Acidovorax citrulli）是引起瓜类细菌性果斑病的重要种传细菌,对瓜类作物生产造成巨大经济损失,种子处理是预防和控制果斑病的关键环节。本研究利用高通量小分子化合物筛选技术,对4个小分子化合物库共计4 952个小分子化合物进行靶标菌的生长抑制初步筛选。初次筛选小分子化合物浓度为100μmol/L,以***野生型菌株Xu 3-14为靶标,共筛选出具有致死和完全抑制效果的小分子化合物127个（选中率为2.5%）。二次筛选发现部分化合物在对番茄细菌性溃疡病菌（Clavibacter miciganensis ***）和十字花科黑腐病菌（Xanthomonas campestris ***）保持抑制作用的同时,对植物有益菌如荧光假单胞菌（Pseudomonas fluorescens）和枯草芽孢杆菌（Bacillus subtilis）的生长无显著影响。50%以上的化合物对拟南芥和西瓜没有表现植物毒性。综合分析二次筛选中小分子化合物的特异性、敏感性和植物毒性的评估结果,以及小分子化合物对***种子到种苗传带抑制率的实验室测试结果,选出了9个化合物作为种子处理药剂对人工接菌的西瓜、甜瓜种子进行田间测试。最终选出5个显著抑制***的小分子化合物,防病效果最高可达到95%。本研究筛选出的5种小分子化合物为***特异性杀菌剂的开发提供了参考。

A Review on Pharmacological and Analytical Aspects of Naringenin

Flavonoids are a widely distributed group of phytochemicals having benzo-pyrone nucleus, and more than 4,000 different flavonoids have been described and categorized into flavonols, flavones, flavanones, isoflavones, catechins and anthocyanidins. Flavonoids occurs naturally in fruits, vegetables, nuts, and beverages such as coffee, tea, and red wine, as well as in medical herbs. Flavonoids are responsible for the different colors of plant parts and are important constituents of the human diet. Flavanoids have different pharmacological activities, such as antioxidant, anti-allergic, antibacterial, anti-inflammatory, antimutagenic and anticancer activity. Naringenin belongs to the flavanones and is mainly found in fruits（grapefruit and oranges） and vegetables. Pharmacologically, it has anticancer, antimutagenic, anti-inflammatory, antioxidant, antiproliferative and antiatherogenic activities. Naringenin is used for the treatments of osteoporosis, cancer and cardiovascular diseases, and showed lipid-lowering and insulin-like properties. In the present review, detailed pharmacological and analytical aspects of naringenin have been presented, which revealed the impressive pharmacological profile and the possible usefulness in the treatment of different types of diseases in the future. The information provided in this communication will act as an important source for development of effective medicines for the treatment of various disorders.

A GFP-based bacterial biosensor with chromosomally integrated sensing cassette for quantitative detection of Hg(Ⅱ) in environment

A mercury biosensor was constructed by integrating biosensor genetic elements into E. coli JM109 chromosome in a single copy number, using the attP/attB recombination mechanism of λ phage. The genetic elements used include a regulatory protein gene （merR） along with operator/promoter （O/P） derived from the mercury resistance operon from pDU1358 plasmid of Serratia marcescens. The expression of reporter gene gfp is also controlled by merR/O/P. Integration of the construct into the chromosome was done to increase the stability and precision of the biosensor. This biosensor could detect Hg（Ⅱ） ions in the concentration range of 100–1700 nmol/L, and manifest the result as the expression of GFP. The GFP expression was significantly different （P 0.05） for each concentration of inducing Hg（Ⅱ） ions in the detection range, which reduces the chances of misinterpretation of results. A model using regression method was also derived for the quantification of the concentration of Hg（Ⅱ） in water samples.

Existence of Market Equilibria for Grid Computing

Grid computing has emerged as an effective mechanism for allocating globally available surplus computational capacity to applications whose requirements exceed local capacity. It is often viewed as a commodity exchange with additional grid computing specific constraints that may arise due to requirements on multiple resources (e.g., disk space) in addition to computing power. These constraints are related to complementarity and substitution effects among resources, and significantly alter the assumptions typically used for demonstrating the existence of market equilibrium. However, prior work in grid computing has simply assumed that market equilibria exist. Our work fills this gap by studying the existence of market equilibrium under the grid computing environment. To do so, we first establish an economic framework that incorporates the grid computing specific constraints into a commodity market. We next derive some intuitive necessary conditions based on the computing requirements of individual agents. We finally establish the existence of regular markets as a competitive equilibrium, given that these necessary conditions are met and that the agents’ utility functions satisfy some minimal requirements. In the process, we also show existence of competitive equilibrium for the special case of grid computing as a pure exchange economy.

基于高分辨率多道二维地震资料、重力异常数据及工业钻孔岩性信息，研究印度西南大陆边缘Laccadive洋脊和Laccadive盆地基底构造特征。地貌和基底特征地震资料解释结果表明，研究区基底表现为块断型地垒／地堑样式，存在平行于海岸线（南北走向）的基底高地，边缘高地上几乎未发生任何沉积物沉积作用，边缘高地周围区域却发育厚层沉积物，局部厚度可高达2～3S（双程旅行时），边缘高地周围的沉积序列通常突然尖灭于陡倾正断层的下盘。Laccadive洋脊之上覆盖薄层沉积物，洋脊上存在多个脊峰（沿洋脊伸长方向展布），有时可能露出海底，此种基底脊峰的宽度通常为数千米，表现为明显的重力异常高值。洋脊基底高度断裂，洋脊中央已演化成类似裂谷的洼地。

Beneficial Effects of <i>Fumaria indica</i>on Chronic Stress-Induced Neurobehavioral and Biochemical Perturbations in Rats

Objective: To evaluate the anti-stress activity of standardized extract of Fumaria indica (FI) through validated beha-vioral models of rodents followed by estimation of biochemical changes associated with chronic stress. Methods: Fifty percent ethanolic extract of FI used in this study was standardized on its contents of fumaric acid and its conjugates (0.45% and 0.35% respectively). Stressed Charles Foster rats received unpredictable foot shocks (2 mA, 1 hr, 14 days) through electric grid. FI was given orally as 0.3% carboxymethyl cellulose (CMC) suspension in 100, 200 and 400 mg/kg doses. For comparison, Panax ginseng (PG) extract (100 mg/kg, p.o.) was used as standard adaptogen. Incidence of gastric ulceration, changes in weight of adrenal and spleen, behavioral depression, cognitive dysfunction test and suppression of sexual behavior in male rats were used as validated behavioral models. Plasma corticosterone, brain levels of lipid peroxides (LPO), superoxide dismutase (SOD), catalase (CAT) and reduced glutathione (GSH), and expression of cytokines IL-1β, TNF-α and IL-10 in circulating white blood cells (WBC) were quantified for ascertaining biochemical changes accompanying stress. Results: As compared to vehicle treated stressed rats, the FI and PG treated rats showed fewer incidence of gastric ulceration, reversal of changes in weight of adrenal gland and spleen, reversal of behavioral depression, better performance in passive and active avoidance tests for cognitive function and increased sexual activity. FI and PG significantly decreased chronic unpredictable stress induced elevation of corticosterone, and both extracts normalized also the abnormal oxidative status of the brain observed in stressed rats. FI treatment also suppressed the elevated level of IL-1β, TNF-α and IL-10 in stressed animals. Conclusions: FI could be another adaptogenic herb and fumaric acid and its conjugates are possibly involved in observed bioactivity of its extract.
