Analysis of 12 variants in the development of gastric and colorectal cancers

AIM To evaluate the relation between 12 polymorphisms and the development of gastric cancer(Gc) and colorectal cancer(cRc).METHODS In this study,we included 125 individuals with Gc diagnosis,66 individuals with cRc diagnosis and 475 cancer-free individuals. All participants resided in the North region of Brazil and authorized the use of their samples. The 12 polymorphisms(in cASP8,cYP2 E1,cYP19 A1,IL1 A,IL4,MDM2,NFKB1,PAR1,TP53,TYMS,UGT1 A1 and XRcc1 genes) were genotyped in a single PcR for each individual,followed by fragment analysis. To avoid misinterpretation due to population substructure,we applied a previously developed set of 61 ancestryinformative markers that can also be genotyped by multiplex PcR. The statistical analyses were performed in Structure v.2.3.4,R environment and SPSS *** After statistical analyses with the control of confounding factors,such as genetic ancestry,three markers(rs79071878 in IL4,rs3730485 in MDM2 and rs28362491 in NFKB1) were positively associated with the development of Gc. One of these markers(rs28362491) and the marker in the UGT1 A1 gene(rs8175347) were positively associated with the development of cRc. Therefore,we investigated whether the joint presence of the deleterious alleles of each marker could affect the development of cancer and we obtained positive results in all analyses. carriers of the combination of alleles RP1 + DEL(rs79071878 and rs28361491,respectively) are at 10-times greater risk of developing Gc than carriers of other combinations. Similarly,carriers of the combination of DEL + RARE(rs283628 and rs8175347) are at about 12-times greater risk of developing cRc than carriers of other *** These findings are important for the comprehension of gastric and cRc development,particularly in highly admixed populations,such as the Brazilian population.

The term serpinopathies was introduced to describe a family of diseases caused by point mutations in serine protease inhibitors,or *** inhibit their cognate protease by an irreversible suicide mechanism starting with the attack of the active site serine on the reactive center loop of the inhibitor,followed by formation of a covalent complex between both proteins,insertion of the reactive center loop of the serpin into its own beta-sheet A,and culminating in distortion of the active site of the serine protease and thus irreversible *** inhibitory mechanism,reminiscent of the movement of a mousetrap,requires a structural flexibility that proves to be unfavorable when the folding of the serpin is altered by mutations responsible for conformational rearrangements,allowing an intermolecular domain exchange characterized by the insertion of the c-terminal domain of a molecule into a second one,thus forming a *** of this insertion reaction leads to the formation of serpin polymers that accumulate within the endoplasmic reticulum(ER)of the cell and consequently reduces the secretion of the wild-type serpin(Greene et al.,2016).

Growth mechanism of octahedron-like BaMoO_4 microcrystals processed in microwave-hydrothermal:Experimental observations and computational modeling

Octahedron-like BaMoO4 microcrystals were synthesized by the co-precipitation method at room temperature and processed in microwave-hydrothermal at 413 K for different times （from 30 min to 5 h）. These microcrystals were analyzed by X-ray diffraction （XRD）, field-emission gun scanning electron microscopy （FEG-SEM） and transmission electron microscopy （TEM）. XRD patterns showed that this material presents a tetragonal structure without the presence of deleterious phases. FEG-SEM and TEM micrographs revealed that the BaMoO4 microcrystals present an octahedron-like morphology with agglomerate nature and polydisperse particle size distribution. These micrographs also indicated that the microcrystals grow along the [0 01 ] direction. The observed crystallographic planes in these structures were modeled computationally and a crystal growth model was proposed in order to explain the morphological changes as a function of processing time.

Since its discovery about 50 years ago,the ubiquitin-proteasome system(UPS)has not stopped to surprise us for its level of sophistication and complexity in achieving substrate-specific protein degradation(Vierstra,2009).At the heart of this system lies the concerted activity of three major classes of enzymes:ubiquitin activators(E1),ubiquitin conjugators(E2),and ubiquitin ligases(or E3).E3s,together with selected E2s,are responsible for promoting the formation of a ubiquitin chain on a specific protein substrate.

Predictive factors associated with hepatitis c antiviral therapy response

Hepatitis c virus(HcV) infection may lead to significant liver injury, and viral, environmental, host, immunologic and genetic factors may contribute to the differences in the disease expression and treatment response. In the early 2000 s, dual therapy using a combinationof pegylated interferon plus ribavirin(PR) became the standard of care for HcV treatment. In this PR era, predictive factors of therapy response related to virus and host have been identified. In 2010/2011, therapeutic regimens for HcV genotype 1 patients were modified, and the addition of NS3/4a protease inhibitors(boceprevir or telaprevir) to dual therapy increased the effectiveness and chances of sustained virologic response(SVR). Nevertheless, the first-generation triple therapy is associated with many adverse events, some of which are serious and associated with death, particularly in cirrhotic patients. This led to the need to identify viral and host predictive factors that might influence the SVR rate to triple therapy and avoid unnecessary exposure to these drugs. Over the past four years, hepatitis c treatment has been rapidly changing with the development of new therapies and other developments. currently, with the more recent generations of pangenotipic antiviral therapies, there have been higher sustained virologic rates, and prognostic factors may not have the same importance and strength as before. Nonetheless, some variables may still be consistent with the low rates of non-response with regimens that include sofosbuvir, daclatasvir and ledipasvir. In this manuscript, we review the predictive factors of therapy response across the different treatment regimens over the last decade including the new antiviral drugs.

Rietveld refinement and optical properties of SrWO_4:Eu^(3+) powders prepared by the non-hydrolytic sol-gel method

We investigated the effect of annealing time on the structure and optical properties of Sr WO4：Eu^3＋ powders prepared by the non-hydrolytic sol-gel method and heat treated at 800 ℃ for 2,4,8 and 16 *** and differential thermal analyses revealed that Sr WO4：Eu^3＋ powders were obtained at about 800 ℃.X-ray diffraction patterns and Rietveld refinement data confirmed that all powders had a scheelite-type tetragonal ***-Raman and Fourier transform infrared spectra indicated structural order at short range and anti-symmetric stretching vibrations of O-W-O bonds associated with tetrahedral[WO4]*** properties were investigated by ultraviolet-visible（UV-vis）diffuse reflectance,and photoluminescence（PL）data which provided the evolution of quantum efficiency（η）and lifetime（τ）.UV-vis spectroscopy evidenced intermediate energy levels within the band gap of Sr WO4：Eu^3＋ *** properties validated that the Eu^3＋ electric-dipole（^5D0→^7F2）transition in PL emission spectra was dominant which proved that Eu^3＋ ions were positioned in a site without an inversion center.[（^5D0→^7F2）/（^5D0→^7F1）]band ratios showed that Eu^3＋ ions were located in a low symmetry *** PL emission,ηandτproved the dependence on the annealing time in the behavior of Sr WO4：Eu3＋powders with a higher relative emission PL intensity as well as higherηandτvalues related to other samples when heat treated at 800℃ for 8 h.

can Biochar Protect Labile Organic Matter Against Mineralization in Soil?

Biochar could help to stabilize soil organic （SOM） matter, thus sequestering carbon （c） into the soil. The aim of this work was to determine an easy method i） to estimate the effects of the addition of biochar and nutrients on the organic matter （SOM） mineralization in an artificial soil, proposed by the Organization for Economic co-operation and Development （OEcD）, amended with glucose and ii） to measure the amount of labile organic matter （glucose） that can be sorbed and thus be partially protected in the same soil, amended or not amended with biochar. A factorial experiment was designed to check the effects of three single factors （biochar, nutrients, and glucose） and their interactions on whole SOM mineralization. Soil samples were inoculated with a microbial inoculum and preincubated to ensure that their biological activities were not limited by a small amount of microbial biomass, and then they were incubated in the dark at 21 ~c for 619 d. Periodical measurements of c mineralized to carbon dioxide （cO2） were carried out throughout the 619-d incubation to allow the mineralization of both active and slow organic matter pools. The amount of sorbed glucose was calculated as the difference between the total and remaining amounts of glucose added in a soil extract. Two different models, the Freundlich and Langmuir models, were selected to assess the equilibrium isotherms of glucose sorption. The cO2-c release strongly depended on the presence of nutrients only when no biochar was added to the soil. The mineralization of organic matter in the soil amended with both biochar and glucose was equal to the sum of the mineralization of the two c sources separately. Furthermore, a significant amount of glucose can be sorbed on the biochar-amended soil, suggesting the involvement of physico-chemical mechanisms in labile organic matter protection.

Light Electrospun Polyvinylpyrrolidone Blanket for Low Frequencies Sound Absorption

Light polymeric soundproofing materials （density = 63 kg/m3） of interest for the transportation industry were fabricated through electrospinning. Blankets of electrospun polyvinylpyrrolidone （average fiber diameter = （1.6 ± 0.5） or （2.8 ± 0.5） μm） were obtained by stacking disks of electrospun mats. The sound absorption coefficients were measured using the impedance tube instrument based on ASTM E1050 and ISO 10534-2. For a given set of disks （from a minimum of 6） the sound absorption coefficient changed with the frequency （in the range 200-1600 Hz） following a bell shape curve with a maximum （where the coefficient is greater than 0.9） that shifts to lower frequencies at higher piled disks number and greater fiber diameter. This work showed that electrospinning produced sound absorbers with reduced thickness （2-3 cm） and excellent sound-absorption properties in the low and medium frequency range.

Global epidemiology of hepatitis c virus infection: an up-date of the distribution and circulation of hepatitis c virus genotypes

AIM To review Hepatitis c virus(HcV) prevalence and genotypes distribution *** We conducted a systematic study which represents one of the most comprehensive effort to quantify global HcV epidemiology,using the best available published data between 2000 and 2015 from 138 countries(about 90% of the global population),grouped in 20 geographical areas(with the exclusion of Oceania),as defined by the Global Burden of Diseases project(GBD). countries for which we were unable to obtain HcV genotype prevalence data were excluded from calculations of regional proportions,although their populations were included in the total population size of each region when generating regional genotype prevalence *** Total global HcV prevalence is estimated at 2.5%(177.5 million of HcV infected adults),ranging from 2.9% in Africa and 1.3% in Americas,with a global viraemic rate of 67%(118.9 million of HcV RNA positive cases),varying from 64.4% in Asia to 74.8% in Australasia. HcV genotype 1 is the most prevalent worldwide(49.1%),followed by genotype 3(17.9%),4(16.8%) and 2(11.0%). Genotypes 5 and 6 are responsible for the remaining common worldwide,the largest proportion of genotypes 4 and 5 is in lower-income countries. Although HcV genotypes 1 and 3 infections are the most prevalent globally(67.0% if considered together),other genotypes are found more commonly in lowerincome countries where still account for a significant proportion of HcV *** A more precise knowledge of HcV genotype distribution will be helpful to best inform national healthcare models to improve access to new treatments.
