Exosomes:the next-generation therapeutic platform for ischemic stroke

Current therapeutic strategies for ischemic stroke fall short of the desired objective of neurological functional ***,there is an urgent need to develop new methods for the treatment of this *** are natural cell-derived vesicles that mediate signal transduction between cells under physiological and pathological *** have low immunogenicity,good stability,high delivery efficiency,and the ability to cross the blood–brain *** physiological properties of exosomes have the potential to lead to new breakthroughs in the treatment of ischemic *** rapid development of nanotechnology has advanced the application of engineered exosomes,which can effectively improve targeting ability,enhance therapeutic efficacy,and minimize the dosages *** in technology have also driven clinical translational research on *** this review,we describe the therapeutic effects of exosomes and their positive roles in current treatment strategies for ischemic stroke,including their antiinflammation,anti-apoptosis,autophagy-regulation,angiogenesis,neurogenesis,and glial scar formation reduction ***,it is worth noting that,despite their significant therapeutic potential,there remains a dearth of standardized characterization methods and efficient isolation techniques capable of producing highly purified *** optimization strategies should prioritize the exploration of suitable isolation techniques and the establishment of unified workflows to effectively harness exosomes for diagnostic or therapeutic applications in ischemic ***,our review aims to summarize our understanding of exosome-based treatment prospects in ischemic stroke and foster innovative ideas for the development of exosome-based therapies.

High-throughput and simultaneous inertial separation of tumor cells and clusters from malignant effusions using spiral-contraction-expansion channels

Tumor cell clusters are regarded as critical factors in cancer pathophysiology,and increasing evidence of their higher treatment resistance and metastasis compared to single tumor cells has been ***,existing cell separation methods that are designed for single tumor cells cannot be used to simultaneously purify tumor cell *** address this problem,we demonstrated a microfluidic approach for the high-throughput,continuous-flow ternary separation of single tumor cells,tumor cell clusters,and WBCs from clinical pleural or abdominal effusions by coupling slanted spiral channels and periodic contraction-expansion *** first systematically explored the influence of particle size and flow rate on particle *** separation performance indicated that 94.0%of WBCs were removed and more than 97%of MDA-MB-231 tumor cells were recovered at a high flow rate of 3500µL/***,more than 90%of tumor cell clusters were effectively preserved after ***,we successfully applied our device for the ternary separation of single tumor cells,tumor cell clusters,and WBCs from different malignant effusions collected from patients with metastatic ***,our spiral-contraction-expansion device has potential as a sample pretreatment tool for the cytological diagnosis of malignant effusions.

Exploring the mechanical behavior and microstructure of compacted loess subjected to dry-wet cycles and chemical contamination

Due to climatic factors and rapid urbanization,the soil in the Loess Plateau,China,experiences the coupled effects of dry-wet cycles and chemical *** the mechanical behavior and corresponding microstructural evolution of contaminated loess subjected to dry-wet cycles is essential to elucidate the soil degradation ***,direct shear and consolidation tests were performed to investigate the variations in mechanical properties of compacted loess contaminated with acetic acid,sodium hydroxide,and sodium sulfate during dry-wet *** mechanical response mechanisms were investigated using zeta potential,mineral chemical composition,and scanning electron microscopy(SEM)*** results indicate that the mechanical deterioration of sodium hydroxidecontaminated loess during dry-wet cycles decreases with increasing contaminant concentration,which is mainly attributed to the thickening of the electrical double layer(EDL)by Nat and the precipitation of calcite,as well as the formation of colloidal flocs induced by OH,thus inhibiting the development of large pores during the dry-wet *** contrast,the attenuation of mechanical properties of both acetic acid-and sodium sulfate-contaminated loess becomes more severe with increasing contaminant concentration,with the latter being more particularly *** is primarily due to the reduction of the EDL thickness and the erosion of cement in the acidic environment,which facilitates the connectivity of pores during dry-wet ***,the salt expansion generated by the drying process of saline loess further intensifies the structural ***,the mechanical performance of compacted loess is sensitive to both pollutant type and concentration,exhibiting different response patterns in the dry-wet cycling condition.

Terrestrial vegetation is a crucial component of the Earth system,and its changes not only represent one of the most distinct aspects of climate change but also exert significant feedback within the climate system by exchanging energy,moisture,and carbon *** quantitatively and mechanistically study climate-vegetation feedback,numerical vegetation models have been developed on the theory of ecophysiological constraints on plant functional *** models eventually can simulate vegetation distribution and succession across different spatial and temporal scales,and associated terrestrial carbon cycle processes by categorizing vegetation into biomes according different plant functional types and their associated environmental *** we review the developing history of vegetation models and provide recent advances and future *** 21st century,static vegetation models,as developed statistical models,can only simulate equilibrated characteristics of vegetation *** last several decades,Dynamic Global Vegetation Models(DGVMs)have been developed to simulate instantaneous responses of vegetation to climate change and associated dynamics,and can be coupled with Earth system models to investigate interactions among atmosphere,ocean,and *** are also widely applied to investigate the dynamics accounting for changes in the geographic distribution patterns of land surface vegetation at different spatial and temporal scales and to assess the impacts of terrestrial carbon and water fluxes and land use *** suggest that future vegetation modeling could integrate with machine learning,and explore vegetation transient response and feedback as well as impacts of process hierarchies and human activities on climate and ecosystem.

improving iEC thermal model for oil natural air natural transformers using optimised parameters based on dynamic simulation

Accurate assessment of hot-spot temperature is essential for the safe operation of power *** dynamic thermal models cannot estimate hot-spot temperature accurately since some input parameters are roughly determined by transformer capacity and cooling mode while ignoring the effect of winding structure,tank dimensions,and material physical *** improve the accuracy of temperature assessment,empirical parameters of the iEC thermal model including thermal constants,winding and oil exponents are optimised with the help of numerical simulation in this *** on energy conservation and heat transfer theory,a computational fluid dynamic(CFD)model of a transformer in oil natural air natural(ONAN)cooling mode is *** CFD model simulates the entity's structure,sizes,and multi-stage heat dissipation processes realistically,so it can more precisely calculate the dynamic hot-spot *** to the simulated temperature curves at different operating conditions,the thermal constants and oil exponent are estimated using non-linear regression,and the winding exponent is optimised using linear regression.A case study is conducted on an ONAN *** shows the improved iEC model with optimised parameters can more accurately evaluate hot-spot temperature,and the absolute error is decreased by 2.4 K(38.7%)compared with traditional thermal models.

A Knowledge-enhanced Two-stage Generative Framework for Medical Dialogue information Extraction

This paper focuses on term-status pair extraction from medical dialogues(MD-TSPE),which is essential in diagnosis dia-logue systems and the automatic scribe of electronic medical records(EMRs).in the past few years,works on MD-TSPE have attracted increasing research attention,especially after the remarkable progress made by generative ***,these generative methods output a whole sequence consisting of term-status pairs in one stage and ignore integrating prior knowledge,which demands a deeper un-derstanding to model the relationship between terms and infer the status of each *** paper presents a knowledge-enhanced two-stage generative framework(KTGF)to address the above *** task-specific prompts,we employ a single model to com-plete the MD-TSPE through two phases in a unified generative form:We generate all terms the first and then generate the status of each generated *** this way,the relationship between terms can be learned more effectively from the sequence containing only terms in the first phase,and our designed knowledge-enhanced prompt in the second phase can leverage the category and status candidates of the generated term for status ***,our proposed special status"not mentioned"makes more terms available and en-riches the training data in the second phase,which is critical in the low-resource *** experiments on the Chunyu and CMDD datasets show that the proposed method achieves superior results compared to the state-of-the-art models in the full training and low-re-sourcesettings.

impact of wetting-drying cycles and acidic conditions on the soil aggregate stability of yellow‒brown soil

Soil aggregate is the basic structural unit of soil,which is the foundation for supporting ecosystem functions,while its composition and stability is significantly affected by the external *** study was conducted to explore the effect of external environment(wetting-drying cycles and acidic conditions)on the soil aggregate distribution and stability and identify the key soil physicochemical factors that affect the soil aggregate *** yellow‒brown soil from the Three Gorges Reservoir area(TGRA)was used,and 8 wetting-drying conditions(0,1,2,3,4,5,10 and 15 cycles)were simulated under 4 acidic conditions(pH=3,4,5 and 7).The particle size distribution and soil aggregate stability were determined by wet sieving method,the contribution of environmental factors(acid condition,wetting-drying cycle and their combined action)to the soil aggregate stability was clarified and the key soil physicochemical factors that affect the soil aggregate stability under wetting-drying cycles and acidic conditions were determined by using the Pearson’s correlation analysis,Partial least squares path modeling(PLS‒PM)and multiple linear regression *** results indicate that wetting-drying cycles and acidic conditions have significant effects on the stability of soil aggregates,the soil aggregate stability gradually decreases with increasing number of wetting-drying cycles and it obviously decreases with the increase of ***,the combination of wetting-drying cycles and acidic conditions aggravate the reduction in the soil aggregate *** wetting-drying cycles,acidic conditions and their combined effect imposes significant impact on the soil aggregate stability,and the wetting-drying cycles exert the greatest *** soil aggregate stability is significantly correlated with the pH,Ca^(2+),Mg^(2+),maximum disintegration index(MDi)and soil bulk density(SBD).The PLS‒PM and multiple linear regression analysis further reveal that the soil aggre

The electrophilic addition to an alkene with an electrophile has been widely studied and applied in organic *** organic chemistry textbook describes the classical reaction between an alkene and an iodine electrophile(such as elemental iodine and N-iodosuccinimide(niS))as a typical ionic reaction,in which an iodonium ion is formed and then attacked by a ***,in this article,we report a new and unusual reaction mode between an alkene and niS;that is,a single electron transfer(SET)process occurs between these two reactants by forming an electron-donor acceptor *** only does this unusual single electron transfer reaction between an alkene and niS add fundamentally important knowledge to organic chemistry,it also provides a valuable synthetic method as the new SET reaction mode with opposite regioselectivity as compared with the traditional ionic mode.
