Senescence in adipose-derived stem cells and its implications in nerve regeneration

Adult mesenchymal stem cells, specifically adipose-derived stem cells have self-renewal and multiple differentiation potentials and have shown to be the ideal candidate for therapeutic applications in regenerative medicine, particularly in peripheral nerve regeneration. Adipose-de- rived stem cells are easily harvested, although they may show the effects of aging, hence their potential in nerve repair may be limited by cellular senescence or donor age. Cellular senescence is a complex process whereby stem cells grow old as consequence of intrinsic events （e.g., DNA damage） or environmental cues （e.g., stressful stimuli or diseases）, which determine a permanent growth arrest. Several mechanisms are implicated in stem cell senescence, although no one is exclusive of the others. In this review we report some of the most important factors modulating the senescence process, which can influence adipose-derived stem cell morphology and function, and compromise their clinical application for peripheral nerve regenerative cell therapy.

Multivariate analysis to predict letrozole efficacy in improving sperm count of non-obstructive azoospermic and cryptozoospermic patients： a pilot study

We tested the hypothesis that letrozole increases sperm count in non-obstructive azoospermic or cryptozoospermic patients with a testosterone （T）/17-beta-2-oestradiol （E2） ratio 〈 10. Forty-six patients with no chromosomal aberrations were randomized into two groups： 22 received letrozole 2.5 mg per day for 6 months （Group 1：6 azoospermic＋ 16 cryptozoospermic patients）, while 24 received a placebo （Group 2：5 azoospermic＋19 cryptozoospermic patients）. The following data were collected： two semen analyses, clinical history, scrotal Duplex scans, body mass index （BMI）, Y microdeletion, karyotype and cystic fibrosis screens and follicle-stimulating hormone （FSH）, luteinizing hormone （LH）, E2, T and prolactin levels. Both before and after letrozole or placebo administration, the patients underwent two semen analyses and hormonal assessments. The differences were evaluated using the Mann-Whitney Utest. The relationships between sperm concentration after letrozole administration with respect to FSH, TIE2 ratio, bilateral testicle volume and BMI before letrozole administration were assessed using multivariate analysis. The side effects were assessed using the chi-square test. Group 1 had sperm concentration （medians： 400-1.290× 10^6 ml^-1; P〈0.01） and motility （medians： class A from 2% to 15%; P〈0.01）, FSH, LH and T significantly increased, while Group 2 did not. E2 levels diminished significantly in Group 1, but not in Group 2. Eight patients in Group 1 demonstrated side effects, whereas no patient side effects were observed in Group 2. The sperm concentration after letrozole administration is inversely related to TIE2, FSH and BMI; a direct relationship emerged between sperm concentration and testicular volume.

The present study aimed to determine whether natural dehydrated feed (DNF) can replace dry extruded feed while preserving the required daily intake, nutrient digestibility and benefits for immunity and antioxidant response in cats. The animals were divided into two groups of seven animals each. One group received DNF, and the other consumed commercial extruded dry feed (DCF). The experiment lasted 45 days and we measured daily consumption and collected blood and feces samples. The animals in the DNF group showed greater digestibility of fiber and mineral matter than DCF. Cats in the DNF group had a higher volume of feces. DNF cats had lower leukocyte and granulocyte counts and cholesterol levels. Serum enzymatic antioxidant responses and non-enzymatic were higher in DNF cats. Cats in the DNF group showed lower serum acute phase proteins in addition to a tendency toward a higher concentration of transferrin than in the DCF group. Fecal Escherichia coli and total coliform counts of cats fed DNF were higher than DCF. We conclude that the daily consumption of cats is adequate. This feed had greater digestibility of fiber and mineral matter, in addition to an improvement in antioxidant and immunological responses.

A novel process is reported which produces an asymmetric supercapacitor through the complete recycling of end-of-life lithium ion *** electrodic powder recovered by industrial scale mechanical treatment of spent batteries was leached and the dissolved metals were precipitated as mixed metals *** battery-type positive electrodes were produced by electrodeposition into nanoporous alumina templates from the electrolytic baths prepared by dissolution of the precipitated *** impact of the different metals contained in the electrodic powder was evaluated by benchmarking the electrochemical performances of the recovered nanowires-based electrodes against electrodes produced by using high-purity *** of inactive Cu in the nanowires lowered the final capacitance of the electrodes while Ni showed a synergistic effect with cobalt providing a higher capacitance with respect to synthetic Co *** carbonaceous solid recovered after leaching was indepth characterized and tested as negative *** the chemical and electrochemical characterization indicate that the recovered graphite is characterized by the presence of oxygen functionalities introduced by the leaching *** has led to the obtainment of a recovered graphite characterized by an XPS C/O ratio,Raman spectrum and morphology close to literature reports for reduced graphene *** asymmetric supercapacitor assembled using the recovered nanowires-based positive electrodes and graphite as negative electrodes has shown a specific capacitance of 42 F g^(-1), computed including the whole weight of the positive electrode and recovered graphite,providing a maximum energy density of ~9 Wh kg^(-1) and a power density of 416 W kg^(-1) at 2.5 mA cm^(-2).

The widespread diffusion of renewable energy sources calls for the development of high-capacity energy storage systems as the A-CAES(Adiabatic Compressed Air Energy Storage)*** this framework,low temperature(100℃–200℃)A-CAES(LT-ACAES)systems can assume a key role,avoiding some critical issues connected to the operation of high temperature *** this paper,two different LT-ACAES configurations are *** two configurations are characterized by the same turbomachines and compressed air storage section,while differ in the TES section and its integration with the *** particular,the first configuration includes two separated cycles:the working fluid(air)cycle and the heat transfer fluid(HTF)*** heat exchangers connect the two cycles allowing to recover thermal energy from the compressors and to heat the compressed air at the turbine *** different HTFs were considered:air(case A)and thermal oil(case B).The second configuration is composed of only one cycle,where the operating fluid and the HTF are the same(air)and the TES section is composed of three different packed-bed thermal storage tanks(case C).The tanks directly recover the heat from the compressors and heat the air at each turbine inlet,avoiding the use of heat *** LT-ACAES systems were modelled and simulated using the ASPEN-Plus and the MATLAB-Simulink *** main aim of this study was the detailed analysis of the reciprocal influence between the turbomachinery and the TES system;furthermore,the performance evaluation of each plant was carried out assuming both on-design and off-design operating ***,the different configurations were compared through the main performance parameters,such as the round-trip efficiency.A total power output of around 10 MW was set,leading to a TES tank volume ranging between 500 and 700 m^(3).The second configuration with three TES systems appears to be the most promising in terms of round-trip efficiency

Injection drug users(IDUs)are at risk of hepatitis C virus(HCV)infection,due to needle and syringe *** HCV infection is a major cause of liver-related morbidity and mortality but can be cured with antiviral treatment leading to sustained viral response(SVR).It is well demonstrated that,when close cooperation between specialists in drug addiction and psychiatrists is assured,patients on maintenance treatment with methadone/buprenorphine can be treated for HCV with response rate,tolerability and side effects similar to those reported in *** guidelines recommend that active injection drug use should not exclude patients from HCV treatment,but many services remain reluctant to treat *** significant pharmacodynamic interactions were reported between approved direct anti-viral agents(DAAs)and buprenorphine or *** adjustments are not recommended;therefore DAAs appear to be the"perfect"therapy for patients taking opiate substitutive *** suggestions have been recently recognized by the European Association for the Study of the Liver(EASL)and included in EASL Recommendations on Treatment of Hepatitis C *** confirm that HCV treatment for IDUs should be considered on an individualized basis and delivered within a multidisciplinary team setting;a history of intravenous drug use and recent drug use at treatment initiation are not associated with reduced SVR and decisions to treat must be made on a case-by-case basis.

We investigated whether letrozole （2.5 mg day-1） improves sperm count in non-obstructive azoospermia （NOA） patients. Four men were included in this study, and they had folliculo-stimulating hormone and other hormone levels within the normal range and no varicoceles or chromosomal aberrations. These four patients were administered letrozole for 3 months. Sperm count, testicular volume, gonadotropin, testosterone （T） and estradiol （E2） blood levels were assessed before, during and 1 week after the suspension of treatment. All patients showed spermatozoa in their ejaculate, increased gonadotropin and T levels and lower E2 levels （P〈0.05 in all cases）, when letrozole was administered. This suggests that letrozole treatment might improve sperm count in an NOA sub-population; however, more studies, including the proper controls, are needed to confirm its efficacy.

Driven by the rise of Web 2.0 and the non-stop spread of mobile device sensors,the concept of Public Participation GIS(PPGIS)is knowing a revolutionary era which perfectly fits the new vision of Digital Earth(DE).This research investigates the opportunity to build up a PPGIS prototype with completely Free and Open Source Software(FOSS)architecture,in which data directly come from field surveys carried out by users.A sample application related to urban monitoring is addressed,which concentrates on user field-collected data concerning reports of road pavement *** Data Kit suite allows users to collect geotagged multimedia information using mobile devices with onboard location sensors(e.g.a Global Positioning System receiver).On field-captured data are sent to a server and stored into a PostgreSQL database with PostGIS spatial *** then disseminates data on the *** the client-side,different solutions allow data visualization on both desktop computers and mobile *** designed architecture provided support for FOSS usage in the process of gathering,uploading,and Web publishing information collected in *** implementation of the developed PPGIS could thus substantially increase public active participation in data creation and sharing,besides enlarging DE knowledge up to the local level.
