The Gsα/cAMP signaling pathway mediates the effect of a variety of hormones and factors that regulate the homeostasis of the post-natal ***,the dysregulated activity of Gsαdue to gain-of-function mutations (R201C/R201H)results in severe architectural and functional derangements of the entire bone/bone marrow *** the consequences of gain-of-function mutations of Gsαhave been extensively investigated in osteoblasts and in bone marrow osteoprogenitor cells at various differentiation stages,their effect in adipogenically-committed bone marrow stromal cells has remained *** generated a mouse model with expression of Gsα^(R201C) driven by the Adiponectin(Adq)***-Gsα^(R201C) mice developed a complex combination of metaphyseal,diaphyseal and cortical bone *** the metaphysis,Gsα^(R201C) caused an early phase of bone resorption followed by bone *** bone formation was sustained by cells that were traced by Adq-Cre and eventually resulted in a high trabecular bone mass *** the diaphysis,Gsα^(R201C),in combination with estrogen,triggered the osteogenic activity of Adq-Cre-targeted perivascular bone marrow stromal cells leading to intramedullary bone ***,consistent with the previously unnoticed presence of Adq-Cre-marked pericytes in intraosseous blood vessels,Gsα^(R201C) caused the development of a lytic phenotype that affected both cortical(increased porosity)and trabecular(tunneling resorption)*** results provide the first evidence that the Adq-cell network in the skeleton not only regulates bone resorption but also contributes to bone formation,and that the Gsα/cAMP pathway is a major modulator of both functions.

Impact of hepatitis B immunoglobulin mode of administration on treatment experiences of patients after liver transplantation: Results from an online survey

BACKGROUND Hepatitis B immunoglobulin(HBIG)in combination with a potent nucleos(t)ide analog is considered the standard of care for prophylaxis against hepatitis B virus(HBV)reinfection after liver transplantation for HBV-associated *** To evaluate patients’satisfaction,preferences,and requirements for subcutaneous(SC),intramuscular(IM),and intravenous(IV)HBIG *** A self-completion,cross-sectional,online,22-question survey was conducted to examine perceptions and satisfaction with current HBIG treatment in adults receiving HBIG treatment following liver transplantation for HBV-associated disease in France,Italy,and *** HBIG products with different administration modes were evaluated using target product profile assessment and a conjoint(trade-off)*** Ninety patients were enrolled;32%,17%,and 51%were SC,IM,and IV HBIG users,*** duration of treatment was 36.2 *** HBIG had the least negative impact on emotional well-being and social life and was perceived as the most convenient,easiest to administer,least painful,and had the highest self-rating of treatment *** IM HBIG users than SC or IV HBIG users reported that administration frequency was excessive(67%,28%,and 28%,respectively).In the target product profile assessment,76%of patients were likely to use hypothetical SC *** the conjoint exercise,administration route,frequency,and duration were key drivers of treatment *** Ease,frequency,duration,and side effects of HBIG treatment administration were key drivers of treatment preferences,and SC HBIG appeared advantageous over IM and IV HBIG for administration ease,convenience,and pain.A hypothetical SC HBIG product elicited a favorable *** demographics,personal preferences,and satisfaction with HBIG treatment modalities may influence long-term treatment compliance.

Therapeutic advances in neural regeneration for Huntington’s disease

Huntington’s disease is a neurodegenerative disease caused by the expansion mutation of a cytosine-adenine-guanine triplet in the exon 1 of the HTT gene which is responsible for the production of the huntingtin (Htt) protein. In physiological conditions, Htt is involved in many cellular processes such as cell signaling, transcriptional regulation, energy metabolism regulation, DNA maintenance, axonal trafficking, and antiapoptotic activity. When the genetic alteration is present, the production of a mutant version of Htt (mHtt) occurs, which is characterized by a plethora of pathogenic activities that, finally, lead to cell death. Among all the cells in which mHtt exerts its dangerous activity, the GABAergic Medium Spiny Neurons seem to be the most affected by the mHtt-induced excitotoxicity both in the cortex and in the striatum. However, as the neurodegeneration proceeds ahead the neuronal loss grows also in other brain areas such as the cerebellum, hypothalamus, thalamus, subthalamic nucleus, globus pallidus, and substantia nigra, determining the variety of symptoms that characterize Huntington’s disease. From a clinical point of view, Huntington’s disease is characterized by a wide spectrum of symptoms spanning from motor impairment to cognitive disorders and dementia. Huntington’s disease shows a prevalence of around 3.92 cases every 100,000 worldwide and an incidence of 0.48 new cases every 100,000/year. To date, there is no available cure for Huntington’s disease. Several treatments have been developed so far, aiming to reduce the severity of one or more symptoms to slow down the inexorable decline caused by the disease. In this context, the search for reliable strategies to target the different aspects of Huntington’s disease become of the utmost interest. 1. Introduction The tegmental wall of the tympanic cavity is a thin plate of the temporal bone that separates the middle cranial fossa(MCF) from the ear. This anatomical region consists of two areas: an anterior one, comprised of the tegmen tympani(To′th et al., 2007), and a posterior one, formed by the tegmen antri and the tegmen mastoideum(Makki et al., 2011). In some patients, the tegmental region of the temporal bone can be interrupted, causing a tegmen defect(TD). A TD is sometimes associated with a meningoencephalic herniation(MEH), in which brain tissue herniates through a TD.

Determination of particulate polycyclic aromatic hydrocarbons in ambient air by gas chromatography-mass spectrometry after molecularly imprinted polymer extraction

A solid phase extraction procedure(SPE)is described for the quantitative analysis of polycyclic aromatic hydrocarbons(PAHs)in atmospheric particulate matter(PM),as ubiquitous environmental pollutants routinely measured in air qualitymonitoring.A SPE cartridge was used based on a molecular imprinted polymer(MIP-SPE)properly tailored for selective retention of PAHs with 4 and more benzene fused *** performance of the clean-up procedure was evaluated with the specific concern of selective purification towards saturated hydrocarbons,which are the PM components mostly interfering GC analysis of target *** optimized operative conditions,the MIP-SPE provided analyte recovery close to 95%for heavier PAHs,from benzo(α)pyrene to benzo(ghi)perylene,and close to 90%for four benzene rings PAHs,with good reproducibility(RSDs:2.5%-5.9%).Otherwise,C_(17)-C_(32) n-alkanes were nearly completely *** proposed method was critically compared with Solid Phase Micro Extraction(SPME)using a polyacrylate *** methods were successfully applied to the analysis of ambient PM2.5 samples collected at an urban polluted *** the two procedures,the MIP-SPE provided the highest recovery(R%≥93%)for PAHs with 5 and more benzene rings,but lower for lighter *** contrast,SPME showed a mean acceptable R%value(∼80%)for all the investigated PAHs,except for the heaviest PAHs in the most polluted samples(R%:110%-138%),suggesting an incomplete purification from the interfering n-hydrocarbons.

Taste sensitivity and divergence in host plant acceptance between adult females and larvae of Papilio hospiton

On the island of Sardinia the lepidopteran Papilio hospiton uses Ferula com- munis as exclusive host plant. However, on the small island of Tavolara, adult females lay eggs on Seseli tortuosum, a plant confined to the island. When raised in captivity on Seseli only few larvae grew beyond the first-second instar. Host specificity of lepidopterans is determined by female oviposition preferences, but also by larval food acceptance, and adult and larval taste sensitivity may be related to host selection in both cases. Aim of this work was：（i） to study the taste sensitivity of larvae and ovipositing females to saps of Ferula and Seseli; （ii） to cross-compare the spike activity of gustatory receptor neurons （GRNs） to both taste stimuli; （iii） to evaluate the discriminating capability between the two saps and determine which neural code/s is/are used. The results show that：（i） the spike responses of the tarsal GRNs of adult females to both plant saps are not different and therefore they cannot discriminate the two plants; （ii） larval L-lat GRN shows a higher activity in response to Seseli than Ferula, while the opposite occurs for the phagostimulant neurons, and larvae may discriminate between the two saps by means of multiple neural codes; （iii） the number of eggs laid on the two plants is the same, but the larval growth performance is better on Ferula than Seseli. Taste sensitivity differences may explain the absence of a positive relationship between oviposition preferences by adult females and plant acceptance and growth performance by larvae.

Salix L.(willow)is the largest genus of the family Salicaceae and plays an important role in riparian habitats,wetlands and in shrub *** to the diff erent implications for the species belonging to this family,it is fundamental to identify molecular tools characterizing relevant clones.A set of six multilocus and multiallelic simple sequence repeat(SSRs)markers are presented,leading to 390 polymorphic fragments considered as single dominant markers and able to discriminate successfully 92 *** *** 24 Salix *** polymorphic fragments have been used to perform genetic diversity studies,and to investigate population structures and cluster analysis in a germplasm *** results highlight the capability of the six SSRs to be powerful genetic resources in applied forestry research,both to distinguish *** clones from Salix *** to perform genetic population studies for breeding programs.

Testicular ultrasound inhomogeneity is an informative parameter for fertility evaluation

Testicular volume(TV)is proposed to be a positive predictor of male fertility status,because of the relation known between the TV and the seminiferous tubule *** of the measurement methodology,the role of TV and testicular ultrasound(US)assessments is still debated in andrological clinical *** this retrospective cohort study,we evaluated TV and testis US role in the diagnostic workup of andrological *** consecutive outpatients undergoing single-operator testis US(Modena,Italy)from March 2012 to March 2018 were enrolled,matching sonographic,hormonal,and seminal data.A total of 302 men were referred and evaluated for gynecomastia,suspected hypogonadism,couple infertility(Cl),or sexual *** the hypogonadal group,TV was lower compared to that in other groups(P<0.001),and a significant,direct correlation between TV and testosterone level was observed in nonandrogen-treated patients(R=0.911,P<0.001),suggesting that testicular size could be related to the testosterone-secreting *** the Cl group,normozoospermic patients showed higher TV compared to men with impaired semen quality(P=0.003)and azoospermia(P=0.003).However,TV was not able to discriminate between patients presenting normal and altered semen *** the contrary,testis US inhomogeneity was more frequent in patients with impaired sperm quality(55.0%;P=0.007)and azoospermia(40.0%;P=0.012),compared to patients with normozoospermia(5%),identifying thereby the sonographic pattern as an informative parameter of the fertility ***,in the Cl workup,US evaluation seems to be more informative than the TV assessment alone.
