Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. Reality of evidence-based practice in palliative care

There has been a paradigm shift in medicine away from tradition, anecdote and theoretical reasoning from the basic sciences towards evidence-based medicine(EBM). In palliative care however, statistically significant benefits may be marginal and may not be related to clinical meaningfulness. The typical treatment vs. placebo comparison necessitated by ‘gold standard' randomised controlled trials(RCTs) is not necessarily applicable. The complex multimorbidity of end of life care involves considerations of the patient's physical, psychological, social and spiritual needs. In addition, the field of palliative care covers a heterogeneous group of chronic and incurable diseases no longer limited to cancer. Adequate sample sizes can be difficult to achieve, reducing the power of studies and high attrition rates can result in inadequate follow up periods. This review uses examples of the management of cancer-related fatigue and death rattle(noisy breathing) to demonstrate the current state of EBM in palliative care. The future of EBM in palliative care needs to be as diverse as the patients who ultimately derive benefit. Non-RCT methodologies of equivalent quality, validity and size conducted by collaborative research networks using a ‘mixed methods approach' are likely to pose the correct clinical questions and derive evidencebased yet clinically relevant outcomes.

The article examines the role of the seas in Sumerian and Akkadian royal inscriptions and literary texts from the third millennium BC *** tracing the presence of the sea in these texts,it becomes clear that the Upper and Lower Sea—or the Mediterranean and the Persian Gulf,though the former could shift geographically—could stand as temiinal points for imperial control;as an obstacle or opponent to tackle;and moreover,as markers of distant,even constructed ***,the sea functioned as both a real and imagined border within the Mesopotamian worldview.

Interactome Analysis of Microtubule-targeting Agents Reveals Cytotoxicity Bases in Normal Cells

Cancer causes millions of deaths annually and microtubule-targeting agents （MTAs） are the most commonly-used anti-cancer drugs. However, the high toxicity of MTAs on normal cells raises great concern. Due to the non-selectivity of MTA targets, we analyzed the interaction net- work in a non-cancerous human cell. Subnetworks of fourteen MTAs were reconstructed and the merged network was compared against a randomized network to evaluate the functional rich- ness. We found that 71.4% of the MTA interactome nodes are shared, which affects cellular pro- cesses such as apoptosis, cell differentiation, cell cycle control, stress response, and regulation of energy metabolism. Additionally, possible secondary targets were identified as client proteins of interphase microtubules. MTAs affect apoptosis signaling pathways by interacting with client pro- teins of interphase microtubules, suggesting that their primary targets are non-tumor cells. The paclitaxel and doxorubicin networks share essential topological axes, suggesting synergistic effects. This may explain the exacerbated toxicity observed when paclitaxel and doxorubicin are used in combination for cancer treatment.

Effect of calcium silicate on nutrient use of lowland rice and greenhouse gas emission from a paddy soil under alternating wetting and drying

In intensively irrigated rice cultivation,plant-available silicon(Si)is a crucial nutrient for improving rice *** a source of Si,calcium silicate(CaSiO3)was amended to evaluate the effect of silicate fertilizer on rice production,nitrogen(N)use efficiency,and greenhouse gas(GHG)emission under alternating wetting and drying in a pot experiment using a tropical soil from a paddy field of the International Rice Research Institute(IRRI)in the *** levels of CaSiO3 amendment,0,112.7,224.5,and 445.8 kg ha^-1,with the recommended N rate were *** results showed that although CaSiO3amendment of 112.7 kg ha^-1resulted in higher rice straw,improved N use efficiency,and reduced N2O emission,there was no difference in grain yield among the four levels of CaSiO3 amendment owing to relatively lower harvest ***,CaSiO3 amendment showed a reverse trend between CH4 and N2O emissions as it reduced N2O emission while led to significantly increased CH4 emission and global warming ***,CaSiO3 amendment was a possible alternative to improve N use efficiency and increase rice straw biomass,but it needs to be reviewed in line with grain yield production and GHG *** is also imperative to test an optimal method of silicate fertilizer amendment in future research in order to compromise a negative impact in tropical soils.

Colorectal cancer with liver metastases:Neoadjuvant chemotherapy,surgical resection first or palliation alone?

Colorectal cancer (CRC) is one of the commonest cancers with 1.2 million new cases diagnosed each year in the world. It remains the fourth most common cause of cancer-related mortality in the world and accounts for > 600000 cancer-related deaths each year. There have been significant advances in treatment of metastatic CRC in last decade or so, due to availability of new active targeted agents and more aggressive approach towards the management of CRC, particularly with liver-only-metastases; however, these drugs work best when combined with conventional chemotherapy agents. Despite these advances, there is a lack of biomarkers to inform us about the accurate management of the patients with metastatic CRC. It is therefore imperative to carefully select the patients with comprehensive multi-disciplinary team input in order to optimise the management of these patients. In this review we will discuss various treatment options available in management of colorectal liver metastases with potential guidance on how and when to choose these options along with consideration on future directions in management of this disease.

Plant height has been a major target for selection of high-yielding varieties in *** height-reducing loci(Rht-B1 and Rht-D1)have been widely used since the Green ***,these genes also negatively affect other agronomic traits such as kernel *** alternative height-reducing loci could benefit wheat *** study focused on the genetics of plant height in 260 historical and contemporary winter wheat accessions via genome-wide association studies using 38,693 single nucleotide polymorphism(SNP)markers generated through genotyping by sequencing,two Kompetitive Allele Specific Polymorphismmarkers,and phenotypic data recorded in two seasons(2016 and 2018).The 260 accessions showed wide variation in plant *** accessions developed after 1960 were shorter than earlier *** broad-sense heritability for plant height was high(H2=0.82),whichwas also supported by a high correlation(r=0.82,P<0.0001)between heights from the two *** detected a total of 16 marker–trait associations(MTAs)for plant height at–lg(P)≥4.0 on chromosomes 1A,2B,2D,3B,4D,5A,5D,6A,6B,7A,and *** detected three of the MTAs(QPLH-2D,QPLH-4B.2,and QPLH-4D)consistently in individual-year and combined-year *** MTAs individually explained 10%–16%of phenotypic *** heightreducing alleles at these threeMTAs appeared after 1960 and increased in frequency *** the genes near these loci were gibberellic acid insensitive(GAI)and GRAS transcription factor(GIBBERELLIC-ACID INSENSITIVE(GAI),REPRESSOR of GAI(RGA),and SCARECROW(SCR)).The evidence from this study and previous reports suggests that QPLH-2D is Rht8.A gene encoding a GRAS transcription factor is located near *** of the QPLH-2D locus and associated candidate genes awaits further study.

Founder effects and the evolution of asymmetrical sexual isolation in a rapidly-speciating clade

Sexual isolation resulting from differences in mate choice behaviors is a hallmark of rapidly-speciating lineages. When present, asymmetrical sexual isolation may provide insights into the mechanisms responsible for the evolutionary change in mate signaling traits. In particular, Kaneshiro＇s hypothesis suggests that divergence in sexual characters between populations may arise in allopatry when ＇derived＇ founding populations are subject to severe population bottlenecks, accompanied by a relaxation of sexual selection relative to ＇ancestral＇ source populations. In the present study, we tested predictions of asymmetrical sexual isolation between two allopatric species of Hawaiian Laupala crickets, representing ＇ancestral＇（L. makaio） and ＇derived＇（L. nigra） taxa. While crickets in this genus are notable for rapid divergence of male courtship songs, these species share similar song types, thus suggesting that patterns of sexual isolation are likely due to other mating cues. Analysis of behavioral responses in conspeci- fic and heterospecific ＇no-choice＇ mating trials revealed pronounced asymmetrical isolation in the direction predicted by Kane- shiro＇s hypothesis, wherein we observed a significant reduction in mating success for crosses involving ＇derived＇ males paired with ＇ancestral＇ females, compared to the reciprocal heterospecific and both conspecific pairings. Further dissection of courtship behaviors suggested this difference did not reflect male mate choice, but rather, marked reduced spermatophore acceptance rates by ＇ancestral＇ females paired with ＇derived＇ males. The results are discussed with respect to founder effect models of speciatiou and the potential role of chemosensory signals in mate choice in these species [Current Zoology 59 （2）： 230-238, 2013].

Systematic review of prognostic importance of extramuralvenous invasion in rectal cancer

AIM: To systematically review the survival outcomes relating to extramural venous invasion in rectal ***: A systematic review was conducted using PRISMA guidelines. An electronic search was carried out using MEDLINE, EMBASE, CINAHL, Cochrane library databases, Google scholar and Pub Med until October 2014. Search terms were used in combination to yield articles on extramural venous invasion in rectal cancer. Outcome measures included prevalence and 5-year survival rates. These were graphically displayed using Forest plots. Statistical analysis of the data was carried ***: Fourteen studies reported the prevalence of extramural venous invasion(EMVI) positive patients. Prevalence ranged from 9%-61%. The pooled prevalence of EMVI positivity was 26% [Random effects: Event rate 0.26(0.18, 0.36)]. Most studies showed that EMVI related to worse oncological outcomes. The pooled overall survival was 39.5% [Random effects: Event rate 0.395(0.29, 0.51)].CONCLUSION: Historically, there has been huge variation in the prevalence of EMVI through inconsistent reporting. However the presence of EMVI clearly leads to worse survival outcomes. As detection rates become more consistent, EMVI may be considered as part of risk-stratification in rectal cancer. Standardised histopathological definitions and the use of magnetic resonance imaging to identify EMVI will improve detection rates in the future.
