Hypertension （HTN） is a risk factor for erectile dysfunction, but its effect on vas deferens （VD） contractility and the ejaculatory response has not been delineated. NG-nitro-L-arginine methyl ester （L-NAME）, a nitric oxide synthase inhibitor, was used for induction of nitric oxide （NO）-deficient HTN. Our aim was to evaluate the effects of L-NAME-induced HTN on rat VD contractility and to determine whether sildenafil affects VD contractility. A total of 36 male rats were divided into （1） control, （2）L-NAME-HTN, （3） sildenafil treated L-NAME-HTN groups. Group 2 was treated with L-NAME （40 mg kgI per day） in drinking water for 4 weeks. Group 3 received sildenafil （1.5 mg kg^-1 per day, by oral gavage） concomitantly with L-NAME. The prostatic portion of the VD was subjected to electrical field stimulation （EFS, 1-20 Hz）, and the P2X1 agonist α,β-methylene ATP （α,β meATP, 100 μmol L^-1-1 μmol L-1） and the al-adrenoceptor agonist phenylephrine （Phe, 100 μmol L^-1-1 mmol L^-1） were used to construct concentration-response curves. These experiments were repeated in the presence of P2X receptor antagonist, pyridoxalphosphate-6-azophenyl-2＇,4＇-disulfonic acid （PPADS, 30 μmol L-1）. VD contractions in response to EFS, a,β-meATP and Phe were significantly enhanced by L-NAME. Sildenafil treatment in the L-NAME group improved the contractile response of VD to EFS （20 Hz）. In the presence of PPADS, the enhanced contractile response of VD to EFS and a,β-meATP in hypertensive rats was reversed. In the rat model of chronic NO depletion, the purinergic and adrenergic components and EFS affect VD contractility. The VD contractile response may be mediated more by the purinergic system than the adrenergic system, and sildenafil may alter the ejaculatory response in men with PE.

Optogenetics is an emerging field that combines optical and genetic approaches to non-invasively interfere with cellular events with exquisite spatiotemporal control. Although it arose originally from neuroscience, optogenetics is widely applicable to the study of many different biological systems and the range of applications arising from this tech- nology continues to increase. Moreover, the repertoire of light-sensitive proteins used for devising new optogenetic tools is rapidly expanding. Light, Oxygen, or Voltage sensing （LOV） and Blue-Light-Utilizing flavin adenine dinucleotide （FAD）（BLUF） domains represent new contributors to the optogenetic toolkit. These small （100-140-amino acids） flavoprotein modules are derived from plant and bacterial photoreceptors that respond to UV-A/blue light. In recent years, considerable progress has been made in uncovering the photoactivation mechanisms of both LOV and BLUF domains. This knowledge has been applied in the design of synthetic photoswitches and fluorescent reporters with applications in cell biology and biotechnology. In this review, we summarize the photochemical properties of LOV and BLUF photosensors and highlight some of the recent advances in how these flavoproteins are being employed to artificially regulate and image a variety of biological processes.

AIM： In the present study, antibody and peripheral blood mononuclear cells （PBMc） proliferative responses against hepatitis c virus （HcV） antigens were evaluated in HcV chronically infected patients. METHODS： Paired serum and PBMc samples were taken six months apart from 34 individuals, either treated or not, and tested by enzyme-linked immunosorbent assay （ELISA） and carboxyfluorescein succinimidyl ester staining. RSULTS： Over 70% of the patients showed specific IgG and IgM against capsid, E1 and NS3, while HVR-1 was recognized by half of the patients. An increase in the levels of the anti-capsid IgM （P = 0.027） and IgG （P = 0.0006） was observed in six-month samples, compared to baseline. Similarly, a significantly higher percent of patients had detectable IgA reactivity to capsid （P = 0.017） and NS3 （P = 0.005） after six months, compared to baseline. Particularly, IgA against structural antigens positively correlated with hepatic damage （P = 0.036）. IgG subclasses evaluation against capsid and NS3 revealed a positive recognition mediated by IgG1 in more than 80% of the individuals. On the contrary, less than 30% of the patients showed a positive proliferative response either of cD4＋ or cD8＋ T cells, being the capsid poorly recognized. cONcLUSION： These results confirm that while the cellular immune response is narrow and weak, a broad and vigorous humoral response occurs in HcV chronic infection. The observed correlation between IgA and hepatic damage may have diagnostic significance, although it warrants further confirmation.

The cyanide content of the parenchyma (pulp) of three sweet cassava cultivars: Alhaji (Tropical Manihot Selection;TMS 98/0581), Ochengbila (Tropical Manihot Selection;TMS 98/0505), and Onono (Tropical Manihot Selection;TMS 98/0524) from three local government areas of Benue State were studied. Their contents were: TMS 98/0581 (41.46 - 53.40) mg/kg, TMS 98/0505 (45.12 - 58.93) mg/kg and TMS 98/0524 (46.38 - 58.08) mg/kg for all the local government areas. For the peels, it ranged from (63.41 - 108.96) mg/kg for all cultivars. The cyanide content of the cultivars varies from one local government to another and also from one farm to another. No appreciable loss of cyanide was observed after four days of storage. The cyanide content of boiled and roasted cassava parenchyma was (5.26 - 7.85) mg/kg and (9.73 - 10.55) mg/kg respectively for all the cultivars. Air dried parenchyma at (26°c - 32°c for one week and oven dried parenchyma at 60°c for four hours ranged from (16.05 - 18.83) mg/kg and (21.82 - 24.04) mg/kg respectively for all cultivars.
