Recent updates of precision therapy for gastric cancer: Towards optimal tailored management

Signaling pathways of gastric carcinogenesis and gastric cancer progression are being avidly studied to seek optimal treatment of gastric cancer. Among them, hepatocyte growth factor (HGF)/c-MET, phosphoinositide 3-kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR) and janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) pathways have been widely investigated. Their aberrant expression or mutation has been significantly associated with advanced stage or poor prognosis of gastric cancer. Recently, aberrations of immune checkpoints including programmed cell death-1/programmed cell death ligand-1 (PD-1/PD-L1) have been suggested as an important step in the formation of a microenvironment favorable for gastric cancer. Accomplishments in basic research have led to the development of novel agents targeting these signaling pathways. However, phase III studies of selective anti-HGF/c-MET antibodies and mTOR inhibitor failed to show significant benefits in terms of overall survival and progression-free survival. Few agents directly targeting STAT3 have been developed. However, this target is still critical issue in terms of chemoresistance, and SH2-containing protein tyrosine phosphatase 1 might be a significant link to effectively inhibit STAT3 activity. Inhibition of PD-1/PD-L1 showed durable efficacy in phase I studies, and phase III evaluation is warranted. Therapeutic strategy to concurrently inhibit multiple tyrosine kinases is a reasonable option, however, lapatinib needs to be further evaluated to identify good responders. Regorafenib has shown promising effectiveness in prolonging progression-free survival in a phase II study. In this topic highlight, we review the biologic roles and outcomes of clinical studies targeting these signaling pathways.

Homeobox genes, including HOX and non-HOX genes, have been identified to be expressed aberrantly in solid tumors. In gastrointestinal(GI) cancers, most studies have focused on the function of non-HOX genes including caudal-related homeobox transcription factor 1(CDX1) and CDX2. CDX2 is a crucial factor in the development of pre-cancerous lesions such as Barrett's esophagus or intestinal metaplasia in the stomach, and its tumor suppressive role has been investigated in colorectal cancers. Recently, several HOX genes were reported to have specific roles in GI cancers; for example, HOXA13 in esophageal squamous cell cancer and HOXB7 in stomach and colorectal cancers. HOXD10 is upregulated in colorectal cancer while it is silenced epigenetically in gastric cancer. Thus, it is essential to examine the differential expression pattern of various homeobox genes in specific tumor types or cell lineages, and understand their underlying mechanisms. In this review, we summarize the available research on homeobox genes and present their potential value for the prediction of prognosis in GI cancers.

Composites were prepared,through hot pressing,using carbon materials with different pore size distributions as additives for commercial Bi_(0.5)Sb_(1.5)Te_(3) thermoelectric material(BST,p-type).Thermoelectric properties of the composites were measured in a temperature range of 298-473 *** conductivity of the composites,especially lattice thermal conductivity,was effectively decreased due to the mesoporous properties of the incorporated carbon *** electrical conductivity of the composites slightly decreased due to the electron scattering at the interface between the carbon material and the commercial BST *** composite with 0.2 vol.%mesoporous carbon powder(36%mesoporosity)exhibited a figure of merit value approximately 10.7%higher than that of commercial BST without *** behavior resulted in 116%improved output power in the composite block-based single element compared with a bare BST thermoelectric *** enhanced figure of merit was attributed to the effective reduction of lattice thermal conductivity by acoustic phonons scattering at the interface between the BST matrix and the mesoporous carbon as well as at the pore surfaces within the mesoporous *** utilizing mesoporous carbon materials used in this study,the shortcomings and economic difficulties of the composite process with low dimensional carbon additives(carbon nanotubes,graphene,and nanodiamond)can be overcome for extensive practical *** carbon powder with a tailored porosity distribution revealed the validity of bulk-type carbon additives to enhance the figure of merit of commercial thermoelectric materials.

Tip vortex cavitation noise of marine propeller became primary concems to reduce hazardous environmental impacts from commercial ship or to keep the underwater surveillance of naval ships. The investigations of the tip vortex and its induced noise are normally conducted through the model test in a water cavitation tunnel. However the Reynolds number of model-test is much smaller than that of the full-scale, which subsequently results in the difference of tip vortex cavitation inception. Hence, the scaling law between model- and full-scales needs to be identified prior to the prediction and assessment of propeller noise in full scale. From previous researches, it is generally known that the incipient caivtation number of tip vortex can be represented as a power of the Reynolds number. However, the power exponent for scaling, which is the main focus of this research, has not been clearly studied yet. This paper deals with the estimation of scaling exponent based on tip vortex cavitation inception test in both full- and model-scale ships. Acoustical measurements as well as several kind of signal processing technique for an inception criterion suggest the scaling exponent as 0.30. The scaling value proposed in this study shows slight difference to the one of most recent research. Besides, extrapolation of model-ship noise measurement using the proposed one predicts the full-scale noise measurement with an acceptable discrepancy.

Owing to the tremendous demands for high-resolution pixel-scale thin lenses in displays,we developed a graphenebased ultrathin square subpixel lens(USSL)capable of electrically tuneable focusing(ETF)with a performance competitive with that of a typical mechanical refractive *** fringe field due to a voltage bias in the graphene proves that our ETF-USSL can focus light onto a single point regardless of the wavelength of the visible light—by controlling the carriers at the Dirac point using radially patterned graphene layers,the focal length of the planar structure can be adjusted without changing the curvature or position of the lens.A high focusing efficiency of over 60% at a visible wavelength of 405 nm was achieved with a lens thickness of <13 nm,and a change of 19.42% in the focal length with a 9% increase in transmission was exhibited under a driving *** design is first presented as an ETF-USSL that can be controlled in pixel units of flat panel displays for visible *** can be easily applied as an add-on to high resolution,slim displays and provides a new direction for the application of multifunctional autostereoscopic displays.

Quaternary intraplate magmatism formed several volcanic islands and seamounts,including Dokdo(DD),Ulleungdo(UD),Simheungtack(ST),Anyongbok,and Isabu in the southwest of the East Sea back-arc *** this study,we present whole-rock geochemical,zircon U–Pb age,and in situ O–Hf isotope data for the submerged volcanic rocks from DD,UD,and ST to provide new insights into the eruption timing of these volcanoes and constrain the magma evolution *** samples used in this study were trachytes and exhibited ferroan,alkalic,and metaluminous to weakly peraluminous *** showed light rare earth element(REE)-enriched patterns with(La/Yb)N ratios of 25.3–31.7 and mostly negative Eu anomalies in a chondrite-normalized REE *** addition,they were enriched in large-ion lithophile elements and high field strength elements;they exhibited positive Pb anomalies and strongly negative Ba,Sr,P,and Ti *** zircons yielded a weighted-mean 206Pb/238U age of 2.61,0.348–0.704,and 2.76–2.94 Ma for the DD,UD,and ST trachytes,*** zircons exhibited lowerδ^(18)O values than normal depleted mantle values,regardless of the crystallization age and spatial distribution of ***δ^(18)O values showed no correlation with U contents or Th/U ratios,indicating that the lowδ^(18)O signatures were of primary magmatic *** Hf isotopic compositions of the zircons were relatively heterogeneous but predominately characterized by positive eHf *** O–Hf mixing modeling revealed that low-δ^(18)O rocks with positive eHf values from the UD and ST volcanoes were derived from a hybrid source of recycled juvenile crustal materials with low-δ^(18)O and positive eHf signatures and an enriched mantle source with normalδ^(18)O and negative eHf *** juvenile oceanic crust in the source was likely metasomatized by seawater at high temperatures prior to *** contrast,the felsic magma that formed the DD volcanoes may have assimilated with regional

随着电力行业碳减排任务的推进,电-碳耦合愈加紧密,为综合能源系统优化运行带来了新机遇、新挑战。根据新型电力系统多能互补、清洁低碳、安全可控、灵活高效、开放互动的基本特征,提出需求侧动态电-碳双响应的日前多级市场交易方法。首先,基于碳排放流理论,将源侧碳排放信息传递至各园区,实现差异运行方式下电力系统源-网-荷的精细化“电-碳”联动。然后,构建需求侧电-碳-绿证多元交易市场,鼓励更多主体参与市场,充分挖掘需求侧在经济、环境等方面的价值。最后,建立主网与各园区间的双层综合能源系统优化调度模型。上层主网发布动态电价与动态碳排放因子,下层需求侧的各园区生成有效的减排方案,使得源荷双侧形成有效互动,综合能源系统整体实现低碳经济运行。仿真结果表明,所提策略能够在较为精确表征碳排放的基础上,通过完善需求侧多元交易市场,深度挖掘园区的碳减排潜力,提高新能源的消纳率,有效平衡系统的经济性、安全性和低碳性。

Improving ionic conductivity of von-Alpen-type NASICON ceramic electrolytes via magnesium doping

NASICON (sodium (Na) superionic conductor) compounds have attracted considerable attention as promising solid electrolyte materials for advanced Na-based batteries. In this study, we investigated the improvement in ionic conductivities of von-Alpen-type NASICON (vA-NASICON) ceramic electrolytes by introducing a magnesium ion (Mg^(2+)) as a heterogeneous element. The optimal Mg-doped vA-NASICON exhibited a high ionic conductivity of 3.64×10^(−3) S·cm^(−1), which was almost 80% higher than that of un-doped vA-NASICON. The changes in physicochemical properties of the vA-NASICONs through the Mg introduction were systematically analyzed, and their effects on the ionic conductivities of the vA-NASICON were studied in detail. When the optimal ratio of Mg^(2+) was used in a synthetic process, the relative density (96.6%) and grain boundary ionic conductivity (σgb) were maximized, which improved the total ionic conductivity (σt) of the vA-NASICON. However, when Mg^(2+) was introduced in excess, the ionic conductivity decreased because of the formation of an undesired sodium magnesium phosphate (NaxMgyPO_(4)) secondary phase. The results of this study are expected to be effectively applied in the development of advanced sodium-based solid electrolytes with high ionic conductivities.
