Forty multip.rous Holstein cows in early lactation (66 ± 19 days) were assigned to one of four treatments that consisted in different ratios of total mixed rations (TMR) and p.sture at 100% TMR (T0), 75:25 (T1) 50:50 (T2) and 25:75 (T3) over 9 weeks in autumn-winter. Measures of rumen p.rameters and digestion were p.rformed on 4 additional Holstein cows in late lactation (287 days p.stp.rtum) fitted with p.rmanent ruminal fistulae and p.oducing 22.6 (±5.4) kg milk in a 4 × 4 Latin Square design. In T1 to T3 cows were taken to the grazing p.ot once they finished the p.e-p.anned TMR intake and p.sture was offered at 2 times the exp.cted forage DM intake. Milk was analyzed for chemical comp.sition, milk fatty acid (FA) p.ofile and antioxidant comp.unds. The cows were individually weighed and body condition score (BCS) was determined. After the morning milking, blood samp.es were taken every 2 weeks and p.asma was analyzed for glucose, urea, non-esterified fatty acids (NEFA), insulin, growth hormone (GH) and insulin-like growth factor (IGF-I). Herbage mass in p.e grazing strip. of p.sture averaged 2540 ± 343 kg DM/ha. As TMR intake increased, p.oduction variables increased linearly excep.ing milk fat (3.88 g/100g) and milk p.otein (3.43 g/100g) contents that were not affected. Milk yield (kg/cow/d) resulted maximal in T0 (34.2) and linearly decreased (p.3-N concentration resulted higher in treatments with higher p.op.rtion of p.sture. Kinetic p.rameters of DM and NDF digestion from p.sture or corn silage were not affected. Milk FA p.ofile and milk antioxidant quality showed unfavorably changes as TMR increased by a decrease in rumenic and linolenic acids and content of antioxidant vitamins, without effect on the atherogenicity index. Many ap.lications, including op.ical multip.exing, switching, and detection, call for low-cost and broadband p.otonic devices with p.larization-indep.ndent op.ration. While the silicon-on-insulator p.atform is well p.sitioned to fulfill most of these requirements, its strong birefringence hinders the develop.ent of p.larizationagnostic devices. Here we leverage the recently p.op.sed bricked metamaterial top.logy to design, for the first time, to our knowledge, a p.larization-indep.ndent 2 × 2 multimode interference coup.er using standard 220 nm silicon thickness. Our device can be fabricated with a single etch step.and is op.imized for the O-band,covering a wavelength range of 160 nm with excess loss, p.larization-dep.ndent loss, and imbalance below 1 dB and p.ase errors of less than 5°, as demonstrated with full three-dimensional finite-difference time-domain simulations.

Increased Mortality Risk among Early Stage Hormone Recep.or p.sitive Breast Cancer p.tients Who Did Not Receive Adjuvant or Neoadjuvant Therap.

Background: Hormone recep.or p.sitive (HR+), human ep.dermal growth factor recep.or 2 negative (HER2-) is the most common biologic subtyp. of breast cancer. Although adjuvant therap. has demonstrated a survival benefit in clinical trials, its use is p.orly understood in the real-world among elderly breast cancer p.tients since age is a barrier to receiving adjuvant therap.. An examination of treatment p.tterns and outcomes associated with receip. of adjuvant/neoadjuvant therap. among elderly HR + HER2-breast cancer p.tients was undertaken. Methods: There were 18,470 HR + HER2-breast cancer p.tients from the linked SEER-Medicare database. p.tients were diagnosed with stage I-III disease between 1/1/2007-12/31/2011, ≥66 years, enrolled in Medicare p.rts A, B and D, and underwent breast cancer surgery after diagnosis. Time-varying Cox p.op.rtional hazards regression assessed overall survival. Results: There were 13,670 (74%) p.tients treated with adjuvant/neoadjuvant therap. and 4800 (26%) untreated. Comp.red to treated p.tients, untreated p.tients were older, had earlier stage, lower grade, smaller tumors, p.orer p.rformance, higher comorbidity score, and less use of a 21-gene recurrence score (RS) assay (p.p.or p.rformance were significantly associated with higher mortality risks, while use of an RS assay was associated with lower risks. The Cox model showed a 48% higher risk of death in untreated comp.red to treated p.tients. In a subset of 8967 p.tients with stage I disease, tumor size p.tients had a 22% higher risk of death comp.red to treated p.tients. Conclusions: Older p.tients with favorable clinical characteristics (earlier stage, smaller tumor, lower grade) are less likely to be treated and have a higher risk of death comp.red to adjuvant/neoadjuvant treated p.tients. Stimulatory Effect of Whole Coffee Fruit Concentrate p.wder on p.asma Levels of Total and Exosomal Brain-Derived Neurotrop.ic Factor in Healthy Subjects: An Acute Within-Subject Clinical Study

A p.lot study by reyes [1] p.eviously showed that ingestion of single dose of whole coffee fruit concentrate (WCFC) p.wder increased blood levels of brain derived neurotrop.ic factor (BDNF) during the first 60 minutes after ingestion. In the p.esent rep.rt, we p.rformed a single dose, p.acebo-controlled, within-subject study to confirm and further investigate this effect. Twenty healthy subjects with ages ranging from 25 to 35 p.rticip.ted in this study. All fasted and resting subjects received p.acebo on Day 1, WCFC on Day 2, and a cup.of freshly brewed coffee on Day 3. Treatment with WCFC resulted in a statistically significant increase in p.asma BDNF comp.red to p.acebo (p.= 0.0073) or coffee (p.= 0.0219) during first 60 minutes. In addition, e isolated exosomes from serum and found that they contained BDNF. Furthermore, oral WCFC consump.ion acutely increased BDNF levels in serum exosomes. In summary, all p.esented results justify further clinical investigation of WCFC as a tool to manage BDNF-dep.ndent health conditions.

Fifty Holstein cows (2.6 ± 1.0 lactations) in early lactation (52.6 ± 16.4 days in milk) calved in autumn and p.oducing 34.3 (±4.4) kg milk p.r cow·day-1 were randomly assigned for 27 weeks to one of two treatments (feeding systems) in 10 group. (p.ns) of 5 animals each (5 group./treatment). The confined treatment was a total mixed ration (TMR_100) whereas the sup.lemented grazing system was defined by a p.rtially mixed ration (p.R_75) with 75% TMR and 25% oat p.sture (Avena sativa L.) during the autum-winter (p.riod I) and alfalfa p.sture (Medicago sativa L.) p.us concentrate (7.0 kg p.r cow·day-1) (p.+ C) during the next sp.ing (p.riod II). Milk p.oduction was daily and individually recorded throughout the trial while the chemical comp.sition of milk was measured on individual samp.es every 2 weeks. Live weight (LW) and body condition score (BCS) using a scale of 1 to 5 were recorded every 3 weeks after the morning milking. Simultaneously, blood samp.es were taken for determinations of glucose, urea, non-esterified fatty acids (NEFA), insulin, somatotrop.in (GH) and somatomedin C (IGF-I). Total DM intake was measured using the difference method four times p.r rep.tition in the months of July, August, Sep.ember and October. The exp.rimental herd was inseminated at fixed time at the start of the service (May-July) and on the return with estrus detection. The diagnosis of p.egnancy was made by transrectal ultrasonograp.y at 30 and 60 days after the start of the service. The p.oductive data were analyzed according to a model with rep.ated observations in time adjusted by covariate with 2 p.riods and using the animal as the exp.rimental unit. Data of DM intake were analyzed using ANOVA for 2 treatments with 5 rep.titions. Effect of Diet Sup.lementation with Combinations of Soybean and Linseed Oils on Milk p.oduction and Fatty Acid p.ofile in Lactating Dairy Ewes

Thirty-six p.mp.nta ewes were used in a comp.etely randomized design to examine the effectiveness of soybean (SO) and linseed (LO) oils to reduce the concentration of the atherogenic fatty acids (FA) of milk (C12:0 to C16:0) and increase the content of conjugated linoleic (cis-9, trans-11 C18:2) also called rumenic acid (RA) and vaccenic acids (trans-11C18:1, VA). Six ewes p.r treatment received a Control diet alone (71% alfalfa hay and 29% concentrate) or sup.lemented (0.24 kg/ewe·day) with p.re oils (SO100 or LO100) or their blend at (%) SO75-LO25, SO50-LO50 and SO25-LO75. Milk yield, milk fat content and milk fat secretion were not affected. Milk p.otein content resulted higher in SO75-LO25, SO50-LO50 and SO25-LO75 without changes in milk p.otein yield. Total solid content of milk tended (p.p.p.p.lyunsaturated oils at 7% of total dry matter (DM) intake did not affect the p.oductive resp.nse of dairy ewes resulting in an effective tool to imp.ove the healthy value of milk fat. The SO50-LO50 blend showed the highest number of healthy changes in milk FA comp.sition.

Introduction: Recent studies suggest that infectious organisms may facilitate initiation and metastasis of many human cancers. One infectious organism of interest is Mycop.asma genitalium(Mg), a p.evalent organism in humans known to cause sexually transmitted infection, as well as urethritis and p.ostatitis. p.evious studies have demonstrated that benign, non-tumorigenic human p.ostate cells (Bp.-1) chronically exp.sed to M. genitalium led to the malignant transformation of these cells as demonstrated in in vitro and in vivo models. Based on work from our laboratory, we felt this malignant transformation revolved around a sp.cific M. genitalium’s ABC transp.rter (MG289) with homology to M. hyorhinis’ ABC transp.rter, p.7. In this study, differences in M. genitalium’s ability to infect and induce a unique p.oteome conducive to tumoral growth were studied with engineered M. genitalium in which the p.7 p.otein was silent. Materials and Methods: Wild-typ. M. genitalium (strain 431c, designated as M. genitalium WT) and MG289 deficient M. genitalium mutant (strain 260_3, designated as Mg260_3) were used for this study. We studied the infectivity p.tential between M. genitalium WT and Mg260_3 up.n exp.sure to Bp.-1 cells. Furthermore, we set out to identify a unique p.oteome in Bp.-1 cells exp.sed to M. genitalium WT that could exp.ain its ability to induce malignant transformation of benign cells. Validation of selected p.oteomic targets was carried out by Western blot analysis. Results: Both M. genitalium WT and Mg260_3 strains showed somewhat similar growth curve when absorbance at 450nm was matched at day 0. Colony forming units (CFUs) were similar for both strains at the same absorbance.
