Predictors of the placebo response in a nutraceutical randomized controlled trial for depression

Objective The placebo response in depression studies is the change in symptoms amongst those who receive an inactive *** well-designed randomized controlled trials(RCTs)of depression have a high proportion of placebo responders,with little understanding as to *** present study assesses characteristics associated with the placebo response in a nutraceutical trial with a large proportion of placebo *** This is a secondary analysis of a nutraceutical depression RCT which identified no overall treatment benefit relative to placebo(n=69 in placebo group).We investigated participant characteristics such as socio-demographics,clinical features,and recruitment methods,and their association with the placebo *** genetic polymorphisms were also *** response was measured based on change in Montgomery-Asberg Depression Rating Scale *** association of these hypothesis-driven variables of interest and the placebo response was examined using linear mixed effects *** Greater levels of education,particularly pursuing post-high school education,better self-reported general health,marriage/de facto,greater improvement in the first trial week,and more failed antidepressant therapies in the current depressive episode were associated with greater placebo *** increased placebo response was not found in those recruited via social media nor in those with concomitant antidepressant *** nucleotide polymorphisms from the tryptophan hydroxylase 1(TPH1)gene(A779C and A218C)were weakly associated with greater placebo response,although the evidence was attenuated after accounting for multiple *** This is,to our knowledge,the first study within nutraceutical research for depression to assess the association between participant characteristics and variation in the placebo *** variables appeared to predict the placebo *** findings may encourage future trial design

BACKGROUND There is concern regarding potential long-term cardiotoxicity with systemic distribution of metals in total joint arthroplasty(TjA)*** To determine the association of commonly used implant metals with echocardiographic measures in TjA *** The study comprised 110 TjA patients who had a recent history of high chromium,cobalt or titanium *** underwent two-dimensional,three-dimensional,Doppler and speckle-strain transthoracic echocardiography and a blood draw to measure metal *** and sex-adjusted linear and logistic regression models were used to examine the association of metal concentrations(exposure)with echocardiographic measures(outcome).RESULTS Higher cobalt concentrations were associated with increased left ventricular end-diastolic volume(estimate 5.09;95%CI:0.02-10.17)as well as left atrial and right ventricular dilation,particularly in men but no changes in cardiac *** titanium concentrations were associated with a reduction in left ventricle global longitudinal strain(estimate 0.38;95%CI:0.70 to 0.06)and cardiac index(estimate 0.08;95%CI,-0.15 to-0.01).CONCLUSION Elevated cobalt and titanium concentrations may be associated with structural and functional cardiac changes in some *** studies are warranted to better understand the systemic effects of metals in TjA patients.

Self-assembled bright luminescent hierarchical materials from a tripodal benzoate antenna and heptadentate Eu(III) and Tb(III) cyclen complexes

The europium heptadentate coordinatively unsaturated (Eu(III)) and the terbium (Tb(III)) 1,4,7,10-tetraazacyclododecane (cyclen) complexes 1 and 2 were used in conjunction with ligand 3 (1,3,5-benzene-trisethynylbenzoate) to form the supramolecular self-assembly structures 4 and 5;this being investigated in both the solid and the solution state. The resulting self-assemblies gave rise to metal centered emission (both in the solid and solution) upon excitation of 3, confirming its role as a sensitizing antenna. Drop-cased examples of ligand 3, and the solid forms of 4 and 5, formed from both organic and mixture of organic-aqueous solutions, were analyzed using Scanning Electron Microscopy, which showed significant changes in morphology;the ligand giving rise to one dimensional structures, while both 4 and 5 formed amorphous materials that were highly dense solid networks containing nanoporous features. The surface area (216 and 119 m2·g^-1 for 4 and 5 respectively) and the ability of these porous materials to capture and store gases such as N2 investigated at 77 K. The self-assembly formation was also investigated in diluted solution by monitoring the various photophysical properties of 3–5. This demonstrated that the most stable structures were that consisting of a single antennae 3 and three complexes of 1 or 2 (e.g., 4 and 5) in solution. By monitoring the excited state lifetimes of the Eu(III) and Tb(III) ions in H2O and D2O respectively, we showed that their hydration states (the q-value) changed from -2 to 0, upon formation of the assemblies, indicating that the three benzoates of 3 coordinated directly to the each of the three lanthanide centers. Finally we demonstrate that this hierarchically porous materials can be used for the sensing of organic solvents as the emission is highly depended on the solvent environment;the lanthanide emission being quenched in the presence of acetonitrile and THF, but greatly enhanced in the presence of methanol.

Current and future opportunities for liquid biopsy of circulating biomarkers to aid in early cancer detection

Early diagnosis of cancer can significantly improve treatment and survival *** and tissue biopsy are the gold standard diagnostic approaches but are costly,invasive,and often unable to detect early-stage *** past decade has marked an acceleration in the discovery and development of liquid biopsy tests for aiding in the detection of various types of tumor markers in non-tissue samples,such as *** biopsy markers include circulating tumor cells,as well as tumor cell fragments,nucleic acids,and *** biopsy may be useful in screening patients considered to be at high risk of developing cancer,for refining diagnosis when combined with other test results,and for early detection of *** in big data analytics,informatics,and artificial intelligence will make it possible to combine patient history,clinical data,and liquid biopsy marker profiles to achieve more accurate and earlier *** this review,we summarize the current use of liquid biopsy in cancer care,including the development of multi-analyte panels to improve diagnostic accuracy and detect several cancer types in a single *** highlight recent advances for potential future applications of liquid biopsy to aid in the diagnosis of early-stage lung *** also discuss the opportunities and challenges of integrating liquid biopsy into current algorithms for cancer screening and diagnosis.

Discrete element modelling of the quasi-static uniaxial compression of individual infant formula agglomerates

Infant formula is usually produced in an agglomerated powder form. These agglomerates are subjected to many transient forces following their manufacture. These can be difficult to quantify experimentally because of their small magnitudes and short durations, Numerical models have the potential to address this gap in the experimental data. The objective of the research described here was to calibrate a discrete element model for these agglomerates using experimental data obtained for quasi-static loading, and to use this model to study the mechanics of the particle response in detail. The Taguchi method was previously proposed as a viable calibration approach for discrete element models. In this work, the method was assessed for calibration of the model parameters （e.g., bond stiffnesses and strengths） considering three responses： the force at failure, strain at failure and agglomerate stiffness. The Weibull moduli for the simulation results and the experimental data were almost identical following calibration and the 37% characteristic stresses were similar. An analysis of the energy terms in the model provided useful insight into the model response. The bond energy and the normal force exerted on the platens were strongly correlated, and bond breakage events coincided with the highest energy dissipation rates.

Glade and Forest-Edge Plant Community Attributes for Three Glade Types in Arusha National Park, Tanzania

The aim of the study was to compare glade and forest-glade edge plant community attributes for man-made, upper and lower natural glades. Two plots were set up in twelve glades at 2.5, 12.5 m and three plots in thirteen glades at 2.5, 12.5 and 22.5 m depending on the size of the glade. The results showed that plants total abundance, percentage basal cover, species richness and species diversity for upper and lower natural glades differed significantly p 2 = 15.95, df = 4, p Eleusine jaegeri, Heterogon contortus, Senna mimisoides, Digitaria scalarum, Clutia abyssinica) were habitat indicators for disturbed areas, wet humid grey/black clay soils for upper natural glades and three (Solanum incanum, Senna didymobotrya, and Pennistem mezianum, were indicators of overgrazed areas and black clay soils for man-made glades.
