BACKGROUND The impact of perioperative blood transfusion on short-and long-term outcomes in pediatric living donor liver transplantation(PLDLT)must still be ascertained,mainly among young children.Clinical and surgical postoperative complications related to perioperative blood transfusion are well described up to three months after adult liver transplantation.AIM To determine whether transfusion is associated with early and late postoperative complications and mortality in small patients undergoing PLDLT.METHODS We evaluated the effects of perioperative transfusion on postoperative complications in recipients up to 20 kg of body weight,submitted to PLDLT.A total of 240 patients were retrospectively allocated into two groups according to postoperative complications:Minor complications(n=109)and major complications(n=131).Multiple logistic regression analysis identified the volume of perioperative packed red blood cells(RBC)transfusion as the only independent risk factor for major postoperative complications.The receiver operating characteristic curve was drawn to identify the optimal volume of the perioperative RBC transfusion related to the presence of major postoperative complications,defining a cutoff point of 27.5 mL/kg.Subsequently,patients were reallocated to a low-volume transfusion group(LTr;n=103,RBC≤27.5 mL/kg)and a high-volume transfusion group(HTr;n=137,RBC>27.5 mL/kg)so that the outcome could be analyzed.RESULTS High-volume transfusion was associated with an increased number of major complications and mortality during hospitalization up to a 10-year follow-up period.During a short-term period,the HTr showed an increase in major infectious,cardiovascular,respiratory,and bleeding complications,with a decrease in rejection complications compared to the LTr.Over a long-term period,the HTr showed an increase in major infectious,cardiovascular,respiratory,and minor neoplastic complications,with a decrease in rejection complications.Additionally,Cox hazard regression found that high-volume RBC transfusion in

Objective:To investigate the in vitro antimicrobial potential of extracts of stem,leaves,flowers,pods and seeds of Moringu okifera(M.okifera) against Vibrio spp.from hatchery water and the prawn Mmrobrachium amazonicum.Methods:The ethanol extracts of stem,leaves,pods and seeds and chloroform extract of flowers of M.oleifera were tested against Vibrio cholerue(V.cholerae) serogroups non-O1/non-O139(n=4).Vibrio vulnificus(n=l) and Vibrio mimicus(n=1).Escherichia coli(E.coli)(ATCC(?) 25922) was used as quality control.Vibrio species were obtained from Macmbrachium amazonicum prawns and from hatchery water from prawn farming.The Minimum Inhibitory Concentration(MIC) was determined by broth microdilution method.Results:The best result was obtained with the ethanol extract of pods,which inhibited three strains of the V.choleme,Vibrio vulnificus,Vibrio minimis and E.coli(MIC range 0.312-5.000 mg/mL).The chloroform extract of flowers was effective against all V.chokrae strains and E.coli(MIC range 0.625-1.250 mg/mL).However,the ethanol extracts of stem and seeds showed low effectiveness in inhibiting the bacterial growth.Conclusions:The extracts of pods,flowers and leaves of M.oleifera have potential for the control of Vibrio spp.Further studies are necessary to isolate the bioaetive compounds responsible for this antimicrobial activity.

Cervical cancer is one of the most preventable and treatable malignancies, however, it still has high incidence and mortality rates, especially in less developed countries. For Brazil, the estimate is more than 16,000 new cases and more than 6000 deaths annually. Despite this, vaccines against HPV (responsible for cervical cancer) are effective and promote a significant decrease in infections and neoplastic lesions of the cervix. As recommended by the World Health Organization, girls aged 9 to 14 should be vaccinated before becoming sexually active. Although the vaccine was developed in 2006, in Brazil immunization only started to be carried out in 2014, initially only for girls between 9 and 11 years old. The present work is an integrative review, with the objective of explaining the current situation of vaccination against HPV throughout the country. Using the PubMed, MED-LINE and Google Scholar databases, a search for scientific articles was carried out during the year 2022, using the descriptors “Vaccine”, “HPV” and “Brazil”, simultaneously selecting 17 articles. The Brazilian recommendations for vaccination against HPV have undergone several strategic changes over time, in the following points: age group and gender of the target population, major site of application of vaccines and number of doses to be applied. Currently, in Brazil, the recommendation for vaccination against HPV is neutral in gender, between 9 and 14 years old and in immunosuppressed people up to 45 years old, with a vaccination schedule of 2 doses with an interval of 6 months between them, and 3 doses for those from 15 years and immunocompromised individual (0 - 2 - 6 months). The tetravalent vaccine offered by public funding continues to be applied at health centers, despite several favorable criticisms of the return of application in schools. In scenarios of climatic change when increased global temperatures can be expected,it is essential to search for technologies that favor sapling survival and growth after planting and increase yield in the field.Kaolin-based particle films(KBPF)have been applied as barriers against the deleterious effects of high levels of solar radiation.The objective of the present study was to assess the effects of applying purified calcined kaolin-based particle film to young eucalyptus plants.Five treatments were carried out:0%(just water),3%,5%,7%,and 10%calcined kaolin applied to the adaxial part of the leaves.A complete randomized block design was used with five treatments and ten replicates.The plants were assessed for height,diameter at ground level,gas exchanges(net photosynthetic CO_(2) assimilation,stomatal conductance and transpiration),instantaneous(WUE)and intrinsic water use efficiency(IWUE),chlorophyll a fluorescence(maximum quantum yield of photosystem II(PSII)(Fv/Fm),concentration of active reaction centers in relation to the quantity of photons absorbed(RC/ABS)and the maximum ratio of quantum yields of photochemical and concurrent non-photochemical processes in PSII(Fv/F0),SPAD reading and leaf ontogeny.The 3%KBPF concentration showed the best responses in biometric assessments 80 days after planting(DAP)and were corroborated by the responses of the leaf,stem and shoot dry matter production as a whole.The leaf ontogeny assessments showed positive responses following KBPF application when considering leaf development,with 7%KBPF concentration resulting in the highest mean values.The mean specific leaf mass had negative response to high KBPF concentrations.At 60 DAP,the gas exchange variables during both assessment periods declined with an increase in KBPF concentration.Significant differences as a result of KBPF applications were found only at the start of the assessments(34 DAP)for both WUE and for IWUE.Leaf‘greenness’(SPAD reading)at 47 days showed a quadratic relationship in both periods.The variables of ch

AIM To investigate the modulatory effect of B-1 cells on murine peritoneal macrophages infected with Leishmania major(L. major) in vitro.METHODS Peritoneal macrophages obtained from BALB/c andBALB/c XID mice were infected with L. major and cultured in the presence or absence of B-1 cells obtained from wild-type BALB/c mice. Intracellular amastigotes were counted, and interleukin-10(IL-10) production was quantified in the cellular supernatants using an enzymelinked immunosorbent assay. The levels of the lipid mediator prostaglandin E2(PGE2) were determined using a PGE2 enzyme immunoassay kit(Cayman Chemical, Ann Arbor, MI), and the number of lipid bodies was quantified in the cytoplasm of infected macrophages in the presence and absence of B-1 cells. Culturing the cells with selective PGE2-neutralizing drugs inhibited PGE2 production and confirmed the role of this lipid mediator in IL-10 production. In contrast, we demonstrated that B-1 cells derived from IL-10 KO mice did not favor the intracellular growth of L. major.RESULTS We report that B-1 cells promote the growth of L. major amastigotes inside peritoneal murine macrophages. We demonstrated that the modulatory effect was independent of physical contact between the cells, suggesting that soluble factor(s) were released into the cultures. We demonstrated in our co-culture system that B-1 cells trigger IL-10 production by L. major-infected macrophages. Furthermore, the increased secretion of IL-10 was attributed to the presence of the lipid mediator PGE2 in supernatants of L. major-infected macrophages. The presence of B-1 cells also favors the production of lipid bodies by infected macrophages. In contrast, we failed to obtain the same effect on parasite replication inside L. major-infected macrophages when the B-1 cells were isolated from IL-10 knockout mice. CONCLUSION Our results show that elevated levels of PGE2 and IL-10 produced by B-1 cells increase L. major growth, as indicated by the number of parasites in cell cultures.

AIM: To evaluated the therapeutic and prophylactic effect of thalidomide on 2, 4, 6-trinitrobenzene sulfonic acid (TNBS)-induced colitis. Thalidomide has been reported to downregulate the expression of tumor necrosis factor a (TNF-α), IL-12, and vascular endothelial growth factor (VEGF), hallmarks of intestinal inflammation in Crohn's disease (CD). METHODS: Male Wistar rats were divided in five groups of ten animals each. Four groups received a rectal infusion of TNBS in ethanol. The first group was sacrificed 7 d after colitis induction. The second and third groups received either thalidomide or placebo by gavage and were sacrificed at 14 d. The fourth group received thalidomide 6 h before TNBS administration, and was sacrificed 7 d after induction. The fifth group acted as the control group and colitis was not induced. Histological inflammatory scores of the colon were performed and lamina propria CD4+ T cells, macrophages, and VEGF+ cells were detected by immunohistochemistry. TNF-a and IL-12 were quantified in the supernatant of organ cultures by ELISA. RESULTS: Significant reduction in the inflammatory score and in the percentage of VEGF+ cells was observed in the group treated with thalidomide compared with animals not treated with thalidomide. Both TNF-αand IL-12 levels were significantly reduced among TNBS induced colitis animals treated with thalidomide compared with animals that did not receive thalidomide. TNF-αlevels were also significantly reduced among the animals receiving thalidomide prophylaxis compared with untreated animals with TNBS-induced colitis. Intestinal levels of TNF-αand IL-12 were significantly correlated with the inflammatory score and the number of VEGF+ cells. CONCLUSION: Thalidomide significantly attenuates TNBS-induced colitis by inhibiting the intestinal production of TNF-α, IL-12, and VEGF. This effect may support the use of thalidomide as an alternate approach in selected patients with CD.

The present study evaluates the performance of three numerical weather forecasting models: Global Forecast System (GFS), Brazilian Regional Atmospheric Modelling System (BRAMS) and ETA Regional Model (ETA), by means of the Mean Error (ME) and the Root Mean Square Error (RMSE), during the most rainy four months period (January to April 2012) on Eastern Amazonia. The models displayed errors of superestimation and underestimation with respect to the observed precipitation, mainly over center-north of Pará and all of Amapá, where the precipitation is higher. Among the analyzed models, GFS shows the best performance, except during January and March, when the model to underestimated precipitation, possibly due to the anomalously high values recorded.

Indiscriminate disposal of residues of domestic and/or industrial origin in aquatic ecosystems resulting from urban activities causes concern about their effects on the fauna. Substances absorbed by animals, such as those cited above, are metabolized by the liver, as it is the main target organ of several pollutants and also exercises basic vital functions in the organism. For this reason the liver is used as the main and most reliable biomarker in environmental studies. Thus the objective of the present study was to identify hepatic alterations resulting from exposure to water contaminated by a few commercial brands of biodegradable detergents and their effects when present together with several other contaminants found in nature. For such, a solution of 1 ppm of biodegradable detergents and water from an urban lake were tested on two widely distributed native Brazilian fish (Astyanax altiparanae and Prochilodus lineatus). Several alterations were found on the liver, including cytoplasm vacuolization, nuclear displacement, macrophage recruitment and reduced liver glycogen, showing a degenerative process in the liver of the fish, due to the action of contaminants.
