Chondroitinase ABC combined with Schwann cell transplantation enhances restoration of neural connection and functional recovery following acute and chronic spinal cord injury

Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal *** we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord *** report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal *** Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder ***,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord *** findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.

By introducing a discrete memristor and periodic sinusoidal functions,a two-dimensional map with coexisting chaos and hyperchaos is *** coexisting chaotic and hyperchaotic attractors under different Lyapunov exponents are firstly found in this discrete map,along with which other regimes of coexistence such as coexisting chaos,quasiperiodic oscillation,and discrete periodic points are also *** hyperchaotic attractors can be flexibly controlled to be unipolar or bipolar by newly embedded constants meanwhile the amplitude can also be controlled in combination with those coexisting *** on the nonlinear auto-regressive model with exogenous inputs(NARX)for neural network.the dynamics of the memristive map is well predicted,which provides a potential passage in artificial intelligencebased applications.

GOAL: the comprehensive gene ontology analysis layer

Homogeneity or heterogeneity of cells is the most fundamental and important features of analyzing biological associations of genes and gene products. Recent bioinformatics technology requires an automated high-throughput analysis application that can handle massively produced data from next generation sequences and dramatically increased size of public proteomic/genomic databases. Although Gene ontology(GO) database has been newly spotlighted on its wide coverage of machine-readable terminologies, its complex DB schema and vast amount of applications utilizing GO without deep considerations of GO term relations dilute the actual power of GO-based analysis and resulted in misleading/under estimated outcomes. Meanwhile, our recent studies showed that BSM score, a new way of measuring functional similarity, clearly outperformed existing conventional methods. However, implementing BSM score that requires integrating multiple databases and calculating scoring matrix is not trivial and even difficult for bioinformatics experts; therefore, a web-based graphical user interface(GUI) tool, Gene Ontology Analysis Layer(GOAL: http://***/chenlab/goal) is introduced to provide user-friendly GO application powered by state of art functional similarity metric,BSM score.

Surface flashover is a devastating electronic avalanche along the gas–solid interface when a high electric field is applied,which is a potential issue that threatens the safe operations of advanced power electronic,electrical,and spacecraft ***,the underlying physical mechanisms for surface flashover development are still under investigation owing to the complex charge transport processes through the gas phase,solid phase,and gas–solid *** this study,the history of surface flashover theory in the last 50 years is introduced,and several k.y questions are reviewed from the perspective of the competing mechanisms of charge transport:the role of each phase in a surface flashover,the origin of surface charging,and effects of traps in solid on surface ***,some suggestions involve charge transport processes in each phase,and their correlations are put forward,and a predictable‘charge transport competitive flashover model’is proposed by clarifying the competing mechanisms of charge transport processes through multiple *** study summarises the history and hot topics of physical mechanisms of surface flashover proposed based on classic and recent progress and offers promising routes for developing a more precise surface flashover theory and improving surface flashover performances.

Paleozoic and Mesozoic Basement Magmatisms of Eastern Qaidam Basin,Northern Qinghai-Tibet Plateau:LA-ICP-MS Zircon U-Pb Geochronology and its Geological Significance

The eastern margin of the Qaidam Basin lies in the k.y tectonic location connecting the Qinling, Qilian and East k.nlun orogens. The paper presents an investigation and analysis of the geologic structures of the area and LA-ICP MS zircon U-Pb dating of Paleozoic and Mesozoic magmatisms of granitoids in the basement of the eastern Qaidam Basin on the basis of 16 granitoid samples collected from the South Qilian Mountains, the Qaidam Basin basement and the East k.nlun Mountains. According to the results in this paper, the basement of the basin, from the northern margin of the Qaidam Basin to the East k.nlun Mountains, has experienced at least three periods of intrusive activities of granitoids since the Early Paleozoic, i.e. the magmatisms occurring in the Late Cambrian （493.1±4.9 Ma）, the Silurian （422.9±8.0 Ma-420.4±4.6 Ma） and the Late Permian-Middle Triassic （257.8±4.0 Ma-228.8＋1.5 Ma）, respectively. Among them, the Late Permian - Middle Triassic granitoids form the main components of the basement of the basin. The statistics of dated zircons in this paper shows the intrusive magmatic activities in the basement of the basin have three peak.ages of 244 Ma （main）, 418 Ma, and 493 Ma respectively. The dating results reveal that the Early Paleozoic magmatism of granitoids mainly occurred on the northern margin of the Qaidam Basin and the southern margin of the Qilian Mountains, with only weak.indications in the East k.nlun Mountains. However, the distribution of Permo-Triassic （P-T） granitoids occupied across the whole basement of the eastern Qaidam Basin from the southern margin of the Qilian Mountains to the East k.nlun Mountains. An integrated analysis of the age distribution of P-T granitoids in the Qaidam Basin and its surrounding mountains shows that the earliest P-T magmatism （293.6-270 Ma） occurred in the northwestern part of the basin and expanded eastwards and southwards, resulting in the P-T intrusive magmatism that ran through the whole basin basement. Long-term tunnel behaviour and ground movements after tunnelling in clayey soils

Long term ground movements above a tunnel may continue to increase with time after tunnelling in clayey soils as the tunnellinginduced excess pore water pressures dissipate,whilst the changing earth pressure acting on the tunnel leads to further tunnel deformation during *** the tunnel itself introduces new drainage conditions;that is,depending on the drainage condition of the tunnel lining,the effective stresses around the tunnel change with time,inducing further soil consolidation.A seepage rate from low permeability clayey soil is often very small and the groundwater seeping into the tunnel can evaporate *** a tunnel may look.impermeable because the surface look. dry,it is possible that the tunnel drainage conditions are actually *** paper summarises the investigation of soil-tunnel consolidation interaction,particularly focusing on ground surface movements and tunnel lining deformation in the interest of engineering *** results show that tunnel lining permittivity relative to the permeability of the surrounding ground plays an important role on both long-term ground movements as well as tunnel lining *** findings published in literature are reviewed step by step starting from a single tunnel,twin tunnels to complex cross passage *** mechanisms of tunnelling-induced soil consolidation for these structures are identified and,where applicable,possible engineering methodologies to assess the magnitude of long-term ground surface settlements and tunnel lining loads are proposed.

Learning to synthesize:robust phase retrieval at low photon counts

The quality of inverse problem solutions obtained through deep learning is limited by the nature of the priors learned from examples presented during the training *** in the case of quantitative phase retrieval,spatial frequencies that are underrepresented in the training database,most often at the high band,tend to be suppressed in the *** hoc solutions have been proposed,such as pre-amplifying the high spatial frequencies in the examples;however,while that strategy improves the resolution,it also leads to high-frequency artefacts,as well as low-frequency distortions in the ***,we present a new approach that learns separately how to handle the two frequency bands,low and high,and learns how to synthesize these two bands into full-band *** show that this“learning to synthesize”(LS)method yields phase reconstructions of high spatial resolution and without artefacts and that it is resilient to high-noise conditions,e.g.,in the case of very low photon *** addition to the problem of quantitative phase retrieval,the LS method is applicable,in principle,to any inverse problem where the forward operator treats different frequency bands unevenly,i.e.,is ill-posed.
