Saccharolobus islandicus REy15A represents one of the very few archaeal models with versatile genetic tools,which include efficient genome editing,gene silencing,and robust protein expression ***,plasmid vectors constructed for this crenarchaeon thus far are based solely on the pRN2 cryptic *** this plasmid coexists with pRN1 in its original host,early attempts to test pRN1-based vectors consistently failed to yield any stable host-vector system for *** hypothesized that this failure could be due to the occurrence of CRISPR immunity against pRN1 in this *** identified a putative target sequence in orf904 encoding a putative replicase on pRN1(target N1).Mutated targets(N1a,N1b,and N1c)were then designed and tested for their capability to escape the host CRISPR immunity by using a plasmid inter-ference *** results revealed that the original target triggered CRISPR immunity in this archaeon,whereas all three mutated targets did not,indicating that all the designed target mutations evaded host *** mutated targets were then incorporated into orf904 individually,yielding corresponding mutated pRN1 backbones with which shuttle plasmids were constructed(pN1aSD,pN1bSD,and pN1cSD).*** transformation revealed that pN1aSD and pN1bSD were functional shuttle vectors,but pN1cSD lost the capability for *** results indicate that the missense mutations in the conserved helicase domain in pN1c inactivated the *** further showed that pRN1-based and pRN2-based vectors were stably maintained in the archaeal cells either alone or in combination,and this yielded a dual plasmid system for genetic study with this important archaeal model.

In recent decades,the spatio-temporal patterns of China’s croplands have been reshaped by disturbances from anthropogenic activities,with complex changes in the topographic characteristics of ***-climbing of croplands(SCCL)is an important issue that threatens sustainable agricultural *** providing land with prominent location advantages,SCCL weakens the water and fertilizer retention capacity for cropland,intensifies various geological disasters,and adversely affects the ecological environment and food yield of these *** is crucial to determine the spatio-temporal variation features and effects of SCCL in China to formulate more accurate cropland protection policies and to maintain food security;however,the current lack of relevant studies is detrimental for capturing trends in cropland resources and sustainable cropland *** this study,we constructed a multi-scale slope spectrum for cropland and total terrain to explore the spatial differences and trends of SCCL from a three-dimensional *** evaluated the natural and socioeconomic effects of SCCL in China from multiple *** indicate that the proportion of cropland with slopes below 2°,5°,and 6°in China decreased by 0.43%,0.47%,and 0.50%from 1980 to 2020,*** became apparent during 1980-1990 and 2010-2020,especially over the recent *** cropland climbing index(CCI)and upper limited slope change(ULSC)to measure the spatio-temporal pattern of SCCL were 0.99%and 1.17°,respectively,during *** the agricultural regional scale,the SCCL was also concentrated in 1980-1990 and 2010-2020,and it is more pronounced in the southern *** proportion of provinces and prefecture-level cities with high-intensity SCCL during 1980-2020 were 87.10%and 49.73%,*** was comparatively more pronounced and broader from 2010 to *** this period,17.84%of prefecture-level cities had no SCCL,and the average CCI for all prefecture-level citi

Four new sesquiterpene lactones(SLs)(1–4),along with a biosynthetically related SL(5),have been isolated from the leaves of Magnolia *** A(1)is notable as the first C18 homogemarane type SL,featuring a unique 1,7-dioxaspiro[4.4]nonan-6-one *** 2 and 3,representing the first instances of chlorine-substituted gemarane-type SL analogs in natural products,were also *** structures of these isolates were elucidated through a combination of spectroscopic data analysis,electronic circular dichroism calculations,and X-ray single-crystal diffraction *** isolates demonstrated anti-inflammatory activity in lipopolysaccharide-stimulated RAW264.7 ***,3–5 showed a significant inhibitory effect on nitric oxide production,with IC50 values ranging from 0.79 to 4.73μmol·L^(−1).Additionally,4 and 5 exhibited moderate cytotoxic activities against three cancer cell lines,with IC50 values between 3.09 and 11.23μmol·L^(−1).

Ischemia-reperfusion(I/R)injury paradoxically occurs during reperfusion following ischemia,exacerbating the initial tissue *** limited understanding of the intricate mechanisms underlying I/R injury hinders the development of effective therapeutic *** Wnt signaling pathway exhibits extensive crosstalk with various other pathways,forming a network system of signaling pathways involved in I/R *** review article elucidates the underlying mechanisms involved in Wnt signaling,as well as the complex interplay between Wnt and other pathways,including Notch,phosphatidylinositol 3-kinase/protein kinase B,transforming growth factor-β,nuclear factor kappa,bone morphogenetic protein,N-methyl-D-aspartic acid receptor-Ca2+-Activin A,Hippo-yes-associated protein,toll-like receptor 4/toll-interleukine-1 receptor domain-containing adapter-inducing interferon-β,and hepatocyte growth factor/mesenchymal-epithelial transition *** particular,we delve into their respective contributions to key pathological processes,including apoptosis,the inflammatory response,oxidative stress,extracellular matrix remodeling,angiogenesis,cell hypertrophy,fibrosis,ferroptosis,neurogenesis,and blood-brain barrier damage during I/R *** comprehensive analysis of the mechanisms involved in Wnt signaling during I/R reveals that activation of the canonical Wnt pathway promotes organ recovery,while activation of the non-canonical Wnt pathways exacerbates ***,we explore novel therapeutic approaches based on these mechanistic findings,incorporating evidence from animal experiments,current standards,and clinical *** objective of this review is to provide deeper insights into the roles of Wnt and its crosstalk signaling pathways in I/R-mediated processes and organ dysfunction,to facilitate the development of innovative therapeutic agents for I/R injury.

To investigate the role of co-stimulatory and co-inhibitory molecules on immune tolerance in immune thrombocytopenia(ITP),this study mapped the immune cell heterogeneity in the bone marrow of ITP at the single-cell level using Cytometry by Time of Flight(CyTOF).Thirtysix patients with ITP and nine healthy volunteers were enrolled in the *** soluble immunomodulatory molecules,more sCD25 and sGalectin-9 were detected in ITP *** the cell surface,co-stimulatory molecules like ICOS and HVEM were observed to be upregulated in mainly central memory and effector T *** contrast,co-inhibitory molecules such as CTLA-4 were significantly reduced in Th1 and Th17 cell *** a platelet count of 30×10^(9)L^(−1)as the cutoff value,ITP patients with high and low platelet counts showed different T cell immune ***-presenting cells such as monocytes and B cells may regulate the activation of T cells through CTLA-4/CD86 and HVEM/BTLA interactions,respectively,and participate in the pathogenesis of *** conclusion,the proteomic and soluble molecular profiles brought insight into the interaction and modulation of immune cells in the bone marrow of *** may offer novel targets to develop personalized immunotherapies.

Neutron-rich P,Cl,and K isotopes,particularly those with neutron numbers of approximately N=28,have attracted extensive experimental and theoretical *** utilize the ab initio valence-space in-medium similarity renormalization group approach,based on chiral nucleon-nucleon and three-nucleon forces,to investigate the exotic properties of these *** calculations of the low-lying spectra are performed.A key finding is the level inversion between 3/2^(+)_(1)and 1/2^(+)_(1)states in odd-A isotopes,attributed to the inversion ofπ0xod3/2 andπls1/2 single-particle *** ab initio calculations,which incorporate the three-nucleon forces,correlate closely with existing experimental *** calculations of effective proton single-particle energies provide deeper insights into the shell evolution for Z=14 and 16 *** results indicate that the three-body force plays important roles in the shell evolution for Z=14 and 16 sub-shells with neutron numbers ranging from 20 to ***,systematic ab initio calculations are conducted for the low-lying spectra of odd-odd *** results correspond with experimental data and provide new insights for future research into these isotopes up to and beyond the drip line.

A novel approach based on thermal diffusion was used to achieve controllable Mg content in A2B7-type La-Mg-Ni-based *** formation mechanism of the A2B7-type phase as a result of the thermal diffusion process and the effect of Mg content on hydrogen storage performance were investigated.X-ray diffraction（XRD） patterns and Rietveld refinement results showed that increased Mg transformed the LaNi5phase in the La0.74Sm0.03y0.23Ni4.32Al0.04precursor alloy into a superlattice *** electron microscopy（SEM） images showed that Mg was evenly distributed in the alloy *** in the superlattice significantly inhibited the phase decomposition of the superlattice structure during the hydrogen absorption/desorption *** A2B7-type La0.57Sm0.02y0.18Mg0.23Ni3.38Al0.03alloy composed of Gd2Co7and Ce2Ni7phases was successfully *** pressure-composition isotherm profiles showed that the alloy had a hydrogen storage capacity as high as 1.73 wt%,with good cycling *** 50 cycles of hydrogen absorption/desorption,the alloy retained a hydrogen storage capacity of 1.45 wt%,with a capacity retention rate of up to 84.28%.The Mg thermal diffusion process thus provides a new approach for the controlled preparation of La-Mg-Ni-based alloys.
