国际上被誉为"海洋药物宝库"的芋螺毒素,具有特异结合动物体内各种离子通道和受体的特殊功能。其中的α-芋螺毒素能特异性地作用于乙酰胆碱受体,对其亚型具有极高的选择性阻断活性。因而,α-芋螺毒素不但具有重要的科学价值,而且具有巨...

国际上被誉为"海洋药物宝库"的芋螺毒素,具有特异结合动物体内各种离子通道和受体的特殊功能。其中的α-芋螺毒素能特异性地作用于乙酰胆碱受体,对其亚型具有极高的选择性阻断活性。因而,α-芋螺毒素不但具有重要的科学价值,而且具有巨大的药物开发潜力,在镇痛、戒烟戒毒、乳腺癌、肺癌、以及脑神经疾病等多种疑难杂症的研究、诊断及其治疗领域具有极好的应用前景。近年来,作者经过大量筛选研究,鉴定出5个海南产新颖α-芋螺毒素,对它们的人工合成、三维结构、作用靶点、与受体相互作用机制、药理药效等进行了深入的研究,其中3个在动物模型上,镇痛活性比吗啡强300-1200倍;另2个是戒烟戒毒的候选新药。特别是:(1)发现的新型芋螺毒素TxIB,是国际上迄今发现的唯一能将α6/α3β2β3乙酰胆碱受体和其他所有亚型的乙酰胆碱受体区分开的工具药,这在神经科学以及神经疾病的研究、诊断和治疗领域具有非常重要的意义;(2)发现的新芋螺毒素TxID,是迄今发现的活性最强的α3β4乙酰胆碱受体亚型的特异阻断剂;(3)发现的新芋螺毒素LvIA,是迄今发现的对极难区分的α3β2和α6β2两种亚型区分度最高的配体;(4)发现的新超家族芋螺毒素GeXIVA是目前活性最强的α9α10乙酰胆碱受体特异阻断剂。α6/α3β2β3与α3β4乙酰胆碱受体是治疗成瘾的药物作用靶点;α9α10乙酰胆碱受体是治疗神经痛的靶点。这些新芋螺毒素已申请国际PCT专利,其中4个新芋螺毒素已在J Biol Chem、FAsEB J、J Med Chem等上发表,它们在神经精神疾病机理的研究,以及镇痛、戒烟戒毒、帕金森症、乳腺癌、肺癌等新药研发领域具有非常巨大的应用价值,是很难得的海洋原创候选新药或先导药物。

Background and Purpose-Phosphodiesterase 4D (PDE4D) was identified recently as the first novel stroke gene to predispose to ischemic stroke independently of conventional risk factors. An association was only found with large vessel and cardioembolic stroke, suggesting a mechanism of accelerated *** so ught to replicate this association in ischemic stroke as a whole, and individual stroke subtypes, in a non-Icelandic European population. To assess a role in e arly atherosclerosis,we also sought associations with underlying asymptomatic at herosclerosis itself, assessed by carotid ultrasound in a community population. Methods -A total of 737 consecutive white patients with stroke and 933 white co mmunity controls free of symptomatic cerebrovascular disease were examined using a case control methodology. For association with atherosclerosis,intima-media thickness (IMT) in a community population (n=1000) was assessed using carotid ultrasound. Nineteen single nucleotide polymorphisms (sNPs) and 1 minisatellite in the PDE4D gene were determined, with haplotyping undertaken using Phase 2.0. Results -No association with ischemic stroke overall was identified. six of the 19 sNPs were associated with cardioembolic stroke and 2 different sNPs with large vessel disease. There was no association with carotid artery IMT or carotid plaque in the asymptomatic community *** -The PDE4D gene is not a major risk factor for ischemic stroke, or early atherosclerosis, within the 2 European population samples studied. On analysis of individual stroke subtypes, there is a possible association with cardioembolic stroke, but the lack of association with carotid IMT and plaque would suggest that this is via a mechanism other than accelerated atherosclerosis.

A high angular resolution near-infrared image that shows the intensity of polarization for the GG Tau A binary system was obtained with the subaru Telescope. The image shows a circumbinary disk scattering the light from the central binary. The azimuthal profile of the intensity of polarization for the circumbinary disk is roughly reproduced by a simple disk model with the Henyey-Greenstein phase function and the Rayleigh function, indicating there are small dust grains at the surface of the disk. Combined with a previous observation of the circumbinary disk, our image indicates that the gap structure in the circumbinary disk orbits counterclockwise, but material in the disk orbits clockwise. We propose that there is a shadow caused by material located between the central binary and the circumbinary disk. The separations and position angles of the stellar components of the binary in the past 20 yr are consistent with the binary orbit with a = 33.4 AU and e = 0.34.

地点:南非夸祖鲁纳塔尔赫拉比萨卫生服务分区。目的:描述纳入区级结核病防治规划的以社区为基础的耐多药结核病(MDR-TB)治疗管理模式的建立,并评价此模式下患者早期治疗转归是否可以与传统的住院治疗关怀模式相比。设计:将2008年3—12月间在社区管理模式(CM)下开始接受耐多药结核病治疗的患者与2001年1月至2008年2月间在传统住院治疗关怀模式(TM)下开始接受耐多药结核病治疗的患者进行对比。运用Mantel-Cox long-rank检验在Kaplan-Merier生存曲线中比较2组患者开始治疗的时间、痰涂片和痰培养阴转的时间。摘要结果:共纳入50例CM患者和57例TM患者;50例CM患者中有39例(78.0%)为人类免疫缺陷病毒(HIV)阳性。CM患者开始治疗的中位时间是84 d,TM患者开始治疗的中位时间是106.5 d(P=0.002)。CM患者痰涂片阴转的中位时间比TM患者短(59 d对92 d,P=0.055),痰培养阴转的中位时间也是如此(85 d对119 d,P=0.002)。结论:以社区为基础的耐多药结核病治疗管理模式在南非农村地区现有的结核病防治规划中是可行的,应在资源允许的地区进行推广。

*** York City(NYC)experienced an initial surge and gradual decline in the number of sARs-CoV-2-confirmed cases in 2020.A change in the pattern of laboratory test results in COVID-19 patients over this time has not been reported or correlated with patient *** performed a retrospective study of routine laboratory and sARs-CoV-2 RT-PCR test results from 5,785 patients evaluated in a NYC hospital emergency department from March to June employing machine learning ***.A COVID-19 high-risk laboratory test result profile(COVID19-HRP),consisting of 21 routine blood tests,was identified to characterize the sARs-CoV-2 *** half of the sARs-CoV-2 positive patients had the distinct COVID19-HRP that separated them from sARs-CoV-2 negative ***-CoV-2 patients with the COVID19-HRP had higher sARs-CoV-2 viral loads,determined by cycle threshold values from the RT-PCR,and poorer clinical outcome compared to other positive patients without the ***,the percentage of sARs-CoV-2 patients with the COVID19-HRP has significantly decreased from March/April to May/***,viral load in the sARs-CoV-2 patients declined,and their laboratory profile became less distinguishable from sARs-CoV-2 negative patients in the later *** longitudinal analysis illustrates the temporal change of laboratory test result profile in sARs-CoV-2 patients and the COVID-19 evolvement in a Us *** analysis could become an important tool in COVID-19 population disease severity tracking and *** addition,this analysis may play an important role in prioritizing high-risk patients,assisting in patient triaging and optimizing the usage of resources.
