Ventricular tachycardia(VT) is a crucial cause of sudden cardiac death(SCD) and a primary cause of mortality and morbidity in patients with structural cardiac disease. VT includes clinical disorders varying from benign to lifethreatening. Most life-threatening episodes are correlated with coronary artery disease, but the risk of SCD varies in certain populations, with various underlying heart conditions, specific family history, and genetic variants. The targets of VT management are symptom alleviation, improved quality of life, reduced implantable cardioverter defibrillator shocks, prevention of reduction of left ventricular function, reduced risk of SCD, and improved overall survival. Antiarrhythmic drug therapy and endocardial catheter ablation remains the cornerstone of guideline-endorsed VT treatment strategies in patients with structural cardiac abnormalities. novel strategies such as epicardial ablation, surgical cryoablation, transcoronary alcohol ablation, pre-procedural imaging, and stereotactic ablative radiotherapy are an appealing area of res-earch. In this review, we gathered all recent advances in innovative therapies as well as experimental evidence focusing on different aspects of VT treatment that could be significant for future favorable clinical applications.

Background Multiple cerebral venous sinus thrombosis(CVT)registries from various geographical regions indicate that female gender,the use of contraceptive pills,pregnancy and puerperium are important risk *** this study,we report the changes in the epidemiology of patients with CVT managed over the past 26 *** The CMC Vellore CVT registry is a prospectively maintained database at the Christian Medical College,Vellore since January *** software was used to analyse the data and assess the changes in the incidence,age and gender distribution over the previous 26 *** Among 1701 patients treated during the study period,908(53%)were women and 793(47%)were *** mean incidence of CVT was 49 per 100000 admissions before 2010,which increased to 96 per 100000 after *** gender had a higher odds of developing CVT OR–2.07(CI 1.68 to 2.55,pning incidence of postpartum CVT after 2010 compared with the decade before 2010(50%vs 20%).The mean age at presentation had increased from 24.5 to 33.2 years in the last *** There was a clear change in the gender pattern from being a condition with female preponderance,to one where equal or more men are being *** incidence of postpartum CVT cases could be the driving *** increase in the overall incidence of CVT cases was noted,probably due to a higher index of clinical suspicion and better diagnostic imaging modalities.

Seasonal flu,primarily caused by influenza A H1n1 and H3n2 subtype viruses or influenza B viruses,is the most prevalent respiratory tract infection globally and leads to substantial morbidity andmortality *** the influenza virus being initially recognized as a respiratory pathogenwithwell-characterized transmission through respiratory droplets,its impact on the ocular epithelium and associated gene expression remains relatively *** this study,we investigated the transcriptional profiles of immortalized human corneal epithelial cells(HCE-S)and A549 human lung epithelial cells infected with H1n1 and H3n2 influenza *** comparison with A549 cells,a reduced number of differentially expressed geneswas observed in HCE-S upon influenza virus ***,there was a significant upregulation of the genes IFI44L and OAS1,along with lower release of the CCL5/RAnTES ***,our findings revealed uniquely upregulated LGALS9(encoding galectin-9)in HCE-S following infection with the 2009 pandemic H1n1 ***,targeted knockdown of LGALS9 in these cells resulted in a measurable decrease in viral infection,highlighting its role in the cellular responses to influenza virus and suggesting a novel avenue for antiviral ***,our findings provide insight into the distinct mechanisms of influenza virus interactions with different epithelial cells and underscore the importance of studying the ocular surface in understanding influenza pathogenesis.

Enhanced potency of an IgM-like nanobody targeting conserved epitope in SARS-CoV-2 spike n-terminal domain

Almost all the neutralizing antibodies targeting the receptor-binding domain(RBD)of spike(S)protein show weakened or lost efficacy against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)emerged or emerging variants,such as Omicron and its *** suggests that highly conserved epitopes are crucial for the development of neutralizing ***,we present one nanobody,n235,displaying broad neutralization against the SARS-CoV-2 prototype and multiple variants,including the newly emerged Omicron and its ***-electron microscopy demonstrates n235 binds a novel,conserved,cryptic epitope in the n-terminal domain(nTD)of the S protein,which interferes with the RBD in the neighboring S *** neutralization mechanism interpreted via flow cytometry and Western blot shows that n235 appears to induce the S1 subunit shedding from the trimeric S ***,a nano-IgM construct(Mn235),engineered by fusing n235 with the human IgM Fc region,displays prevention via inducing S1 shedding and cross-linking virus *** to n235,Mn235 exhibits varied enhancement in neutralization against pseudotyped and authentic viruses in *** intranasal administration of Mn235 in low doses can effectively prevent the infection of Omicron sub-variant BA.1 and XBB in vivo,suggesting that it can be developed as a promising prophylactic antibody to cope with the ongoing and future infection.
