A m.chanism.for Inhaled Anesthetic-Induced Solid Organ Injury: Inflam.ation

Background: Inhaled anesthetics, including halothane, iso- and sevoflurane induce proinflam.atory cytokine release. Halothane is an inhaled anesthetic agent that is m.tabolized by the liver into a highly reactive product, trifluoroacetyl chloride, which can react endogenously to form.a trifluoroacetyl-adduct (TFA-adduct). The m.A-adduct is form.d by acetaldehyde and m.londialdehyde reacting with endogenousproteins and is found in both patients and anim.ls post-consum.tion of alcohol. These TFA and m.A-adducts have been shown to cause the release of proinflam.atorycytokines by endogenous inflam.atory cells. If both adducts share a sim.lar m.chanism.of cell activation, receiving general anesthesia following alcohol ingestion could exacerbate the inflam.atory response caused by the inhaled general anesthetic halothane and lead to solid organ (including liver and brain) injury. m.thods: Control diet and alcohol-fed rats were random.zed to receive halothane pretreatm.nts by intraperitoneal injection m.xed in sesam. oil. Following the intraperitoneal injections, the intact heart was rem.ved, HECs were isolated and stim.lated with unm.dified bovine serum.album.n (Alb), m.A-m.dified Alb (m.A-Alb), Hexyl-m.A, or lipopolysaccharide (LPS), and supernatant concentrations of TNF-α were determ.ned. Results: Halothane pre-treated rat HECs dem.nstrated significantly greater TNF-α concentration following m.A-adduct and LPS stim.lation than the non-halothane pre-treated in both pair and alcohol-fed rats, but was significantly greater in the alcohol-fed groups. Conclusion: These results dem.nstrate that halothane and m.A-adduct pre-treatm.nt will increase the inflam.atory response (TNF-α release) in rat HECs following LPS and m.A stim.lation in vitro. Also, these results suggest that halothane exposure m.y increase the risk of alcohol-induced solid organ injury secondary to TNF-induced inflam.ation. The developm.nt of alcoholic liver disease (ALD) can be attributed to m.ny factors that cause dam.ge to the liver and alter its functions. Data collected over the last 30 years strongly suggests that an im.une com.onent m.y be involved in the onset of this disease. This is best evidenced by the detection of circulating autoantibodies, infiltration of im.une cells in the liver, and the detection of hepatic aldehyde m.dified proteins in patients with ALD. Experim.ntally, there are num.rous im.une responses that occur when proteins are m.dified with the m.tabolites of ethanol. These products are form.d in response to the high oxidative state of the liver during ethanol m.tabolism. causing the release of m.ny inflam.atory processes and potential of necrosis or apoptosis of liver cells. Should cellular proteins becom. m.dified with these reactive alcohol m.tabolites and be recognized by the im.une system. then im.une responses m.y be initiated. Therefore, it was the purpose of this article to shed som. insight into how the im.une system.is involved in the developm.nt and/or progression of ALD.

The onset of alcoholic liver disease (ALD) is initiated by different cell types in the liver and a num.er of different factors including: products derived from.ethanol-induced inflam.ation, ethanol m.tabolites, and the indirect reactions from.those m.tabolites. Ethanol oxidation results in the production of m.tabolites that have been shown to bind and form.protein adducts, and to increase inflam.atory, fibrotic and cirrhotic responses. Lipopolysaccharide (LPS) has m.ny deleterious effects and plays a significant role in a num.er of disease processes by increasing inflam.atory cytokine release. In ALD, LPS is thought to be derived from.a breakdown in the intestinal wall enabling LPS from.resident gut bacterial cell walls to leak into the blood stream. The ability of adducts and LPS to independently stim.late the various cells of the liver provides for a two-hit m.chanism.by which various biological responses are induced and result in liver injury. Therefore, the purpose of this article is to evaluate the effects of a two-hit com.ination of ethanol m.tabolites and LPS on the cells of the liver to increase inflam.a-tion and fi brosis, and play a role in the developm.nt and/or progression of ALD.

The oxygen reduction reaction （ORR） in the cathode catalyst layer （CCL） of polym.r electrolyte fuel cells （PEFC） is one of the m.jor causes of perform.nce loss during operation. In addition, the CCL is the m.st expensive com.onent due to the use of a Pt catalyst. Apart from.the ORR itself, the species transport to and from.the reactive sites determ.nes the perform.nce of the PEFC. The effective transport properties of the species in the CCL depend on its nanostructure. Therefore a three-dim.nsional reconstruction of the CCL is required. A series of two-dim.nsional im.ges was obtained from.focused ion beam. scanning electron m.croscope （FIB-SEm. im.ging and a segm.ntation m.thod for the two-dim.nsional im.ges has been developed. The pore size distribution （PSD） was calculated for the three-dim.nsional geom.try. The influence of the alignm.nt and the anisotropic pixel size on the PSD has been investigated. Pores were found in the range between 5 nm.and 205 nm. Evaluation of the Knudsen num.er showed that gas transport in the CCL is governed by the transition flow regim.. The liquid water transport can be described within continuum.hydrodynam.cs by including suitable slip flow boundary conditions.

类风湿关节炎是一种系统性自身免疫病,其特点之一为心血管疾病的高发病率和高病死率。类风湿关节炎和心血管疾病相关联的机制包括共有的炎症介质、肽/蛋白质的翻译后修饰,以及随后的免疫应答、脂蛋白组成和功能的改变、氧化应激增加和内皮细胞功能障碍。尽管人们对这些机制及其与传统心血管危险因素的复杂相互作用有了越来越多的了解,但对于在类风湿关节炎背景下进行最佳的危险分层、预防和治疗方法仍然未知。减少心血管疾病所致负担的综合性措施,除需对传统危险因素把控外,还要对类风湿关节炎固有的危险因素(如疾病活动度增加)进行优化管理。对类风湿关节炎的治疗似乎对心血管风险以及关联这些疾病的机制产生了不同的影响。需要更多的研究来确定是否存在优先的类风湿关节炎治疗方法可以预防心血管疾病。最终,对于在类风湿关节炎中心血管疾病的独特机制的了解将有助于危险分层,以及在该患者群体中确定有效降低心血管风险的新靶点。

Effects of undigested protein-rich ingredients on polarised sm.ll intestinal organoid m.nolayers

Here, we describe the use of m.nolayers of intestinal epithelial cells derived from.intestinal organoids and transcriptom.cs to investigate the direct effects of dietary protein sources on epithelial function. m.chanically dissociated 3 D organoids of m.use duodenum.were used to generate a polarized epithelium.containing all cell types found in the tissue of origin. The organoid-derived cell m.nolayers were exposed to 4%(w/v) of ‘undigested(non-hydrolysed)-soluble' fraction of protein sources used as feed ingredients [soybean m.al(SBm. and casein], or alternative protein sources(spray dried plasm. protein, and yellow m.al worm., or controls for 6 h prior to RNA isolation and transcriptom.cs. All protein sources altered expression of unique biological processes in the epithelial cells. Exposure of intestinal organoids to SBm.downregulated expression of retinol and retinoid m.tabolic processes as well as cholesterol and lipid biosynthetic pathways, consistent with the reported hypotriglyceridaem.c effect of soy protein in vivo. These findings support the use of intestinal organoids as m.dels to evaluate com.lex interactions between dietary ingredients and the intestinal epithelium.and highlights som. unique host effects of alternative protein sources in anim.l feed and potentially hum.n food.
