Hypertension is a risk factor for a large number of vision-threatening eye disorders.In this study,we investigated for the first time the retinal neural structure of the hypertensive BPH/2J mouse(Schlager mouse)and compared it to its control counterpart,the normotensive BPN/3J strain.The BPH/2J mouse is a selectively inbred mouse strain that develops chronic hypertension due to elevated sympathetic nervous system activity.When compared to the BPN/3J strain,the hypertensive BPH/2J mice showed a complete loss of outer layers of the neural retina at 21 weeks of age,which was indicative of a severe vision-threatening disease potentially caused by hypertension.To elucidate whether the retinal neural phenotype in the BPH/2J strain was attributed to increased BP,we investigated the neural retina of both BPN/3J and BPH/2J mice at 4 weeks of age.Our preliminary results showed for the first time that the BPH/2J strain develops severe retinal neural damage at a young age.Our findings suggest that the retinal phenotype in the BPH/2J mouse is possibly due to elevated blood pressure and may be contributed by an early onset spontaneous mutation which is yet to be identified or a congenital defect occurring in this strain.Further characterization of the BPH/2J mouse strain is likely to i)elucidate gene defects underlying retinal disease;ii)understand mechanisms leading to neural retinal disease and iii)permit testing of molecules for translational research to interfere with the progression of retinal disease.The animal experiments were performed with the approval of the Royal Perth Hospital Animal Ethics Committee(R535/17-18)on June 1,2017.

Association between the rate of the morning surge in blood pressure and cardiovascular events and stroke

Background The exaggerated surge in morning blood pressure （BP） that many patients experience upon awakening may be closely related to target organ damage and may be a predictor of cardiovascular complications. However, no previous studies have evaluated the rate of this surge independently of the evening period. It remains unclear whether the rate of increase experienced during the surge is a significant or independent determinant of cardiovascular events. Methods We randomly selected 340 ambulatory BP monitoring （ABPM） patients. All subjects without type 2 diabetes mellitus were divided into two groups： hypertensive group （n=170） and normotensive group （n=170）. We analyzed ambulatory blood pressure recordings using a double logistic curve-fitting procedure to determine whether the magnitude of the surge in BP and heart rate （HR） in the morning is related to the level of BP in hypertensive individuals. We evaluated the association between the rate of the morning surge in systolic BP （SBP） and the incidence of myocardial infarction and stroke in normotensive and hypertensive subjects. Results Comparisons between hypertensive and normotensive subjects showed that the rates of the morning surges in SBP, mean BP （MBP）, and diastolic BP （DBP） were greater in the hypertensive group （P 〈0.05） than in the normotensive group. The rate of morning surge in BP was found to be correlated with the daytime SBP （r=0.236, P 〈0.01）, the difference between the day and night plateau （r=0.249, P 〈0.01）, and the night SBP （r---0.160, P 〈0.05）, respectively. After controlling for age, sex, and mean systolic pressure within 24 hours （24 h SBP）, the rate of morning surge in SBP was closely correlated with daytime SBP （r=0.463, P 〈0.001）, night SBP （r=-0.173, P 〈0.05）, and the difference between the day and night plateau （r=0.267, P 〈0.001）. Logistic regression analysis revealed that the rate of morning surge in SBP was an independent determinant of myocardial infarction （OR=1.266, 95% C1=1.153-1.389, P 〈0.001） and stroke （OR=1.367, 95%

Helicoverpa zea(Boddie)is a destructive agricultural pest species that is targeted by both Bacillus thuringiensis(Bt)maize and cotton in the United States.Cry1A.105 and Cry2Ab2 are two Bt proteins expressed in a widely planted maize event MON 89034.In this study,two tests(Test-Ⅰ and Test-Ⅱ)were conducted to evaluate the relative fitness of Bt-susceptible and-resistant H.zea on non-Bt diet(Test-Ⅰ and Test-Ⅱ)and a diet containing a mix of Cry1A.105 and Cry2Ab2 at a low concentration(Test-Ⅱ only).Insect populations evaluated in Test-Ⅰ were two Bt-susceptible strains and three Bt-resistant strains(a single-protein Cry1A.105-,a single-protein Cry2Ab2-,and a dual-protein Cry1A.105/Cry2Ab2-resistant strains).Test-Ⅱ analyzed the same two susceptible strains,three backcrossed-and-reselected Cry1A.105/Cry2Ab2-single-/dual-protein-resistant strains,and three F1 heterozygous strains.Measurements of life table parameters showed that neither the single-nor dual-protein Cry1A.105/Cry2Ab2 resistance in H.zea was associated with fitness costs under the test conditions.The single Cry protein resistances at a concentration of a mix of Cry1A.105 and Cry2Ab2 that resulted in a zero net reproductive rate for the two susceptible strains were functionally incomplete recessive or codominant,and the dual-protein resistance was completely dominant.The lack of fitness costs could be a factor contributing to the rapid revolution of resistance to the Cry proteins in this species.Data generated from this study should aid our understanding of Cry protein resistance evolution and help in refining IRM programs for H.zea.

Status of Cry1Ac and Cry2Ab2 resistance in field populations of Helicoverpa zea in Texas, USA

Helicoverpa zea is a major target pest of Bt corn and Bt cotton.Field-evolved resistance of H.zea to Cry1 and Cry2 proteins has been widely reported in the United States.Understanding the frequency of resistance alleles in a target insect is critical for Bt resistance management.Despite multiple cases of practical resistance to Cry proteins having been documented in H.zea,there are no data on the current status of alleles conferring resistance to Cry1Ac and Cry2Ab2 in field populations of this pest.During 2018‒2019,a total of 106 F2 families for Cry1Ac and 120 F2 families for Cry2Ab2 were established using mass mating and light trap strategy.We screened 13,568 and 15,360 neonates using a discriminatory dose of Cry1Ac and Cry2Ab2,respectively.The results showed that 93.4%and 35.0%of the F2 families could survive on the discriminatory dose of Cry1Ac and Cry2Ab2,respectively.The estimated resistance allele frequency for Cry1Ac in H.zea ranged from 0.4150 to 0.4975 and for Cry2Ab2 ranged from 0.1097 and 0.1228.These data indicate that the frequency of alleles conferring resistance to Cry1 and Cry2 proteins in H.zea in Texas are high.In addition,our data suggest the resistance to Cry1Ac and Cry2Ab2 in the screened families of H.zea varies from recessive to dominant.The information in this study provides precise estimates of Cry resistance allele frequencies in H.zea and increases our understanding of the risks to the sustainability of Bt crops.

气管支气管异物是儿童常见的急重症之一。该病起病急、病情重，甚至可危及生命。尽早诊断和取出异物是减少并发症和降低病死率的关键。虽然近年由于防范意识逐渐增强，气管支气管异物发病率有所下降，但由于该病临床表现的多样性，在诊断和鉴别诊断上仍有一定的难度，因此漏诊、误诊时有发生，并可导致严重后果。针对该病提出系统规范的指导性建议十分必要。制订本专家共识的目的是规范气管支气管异物的诊断和治疗流程，指导快速确诊、准确鉴别、及时取出异物以及有效处理围手术期并发症从而保障患儿的生命安全。同时对儿童气管支气管异物需要进行宣传和科普教育，提高全民的预防意识，降低气管支气管异物给儿童造成的意外伤害。

Performance of Bt-susceptible and -heterozygous dual-gene resistant genotypes of Spodoptera frugiperda (J.E.Smith) (Lepidoptera:Noctuidae) in seed blends of non-Bt and pyramided Bt maize

A seed blend refuge has been implemented in the U.S.Corn Belt for Bt maize resistance management.The fall armyworm,Spodoptera frugiperda(J.E.Smith),is a target pest of Bt maize in the Americas.The larvae of this pest are mobile,which may affect the efficacy of seed blend refuges.In this study,field and greenhouse trials were conducted to determine the performance of Bt-susceptible(aabb)and-heterozygous dual-gene-resistant(AaBb)genotypes of S.frugiperda in seed blends of non-Bt and pyramided Bt maize.Three field trials evaluated larval survival,larval growth,and plant injury with aabb in seed blends of Bt maize expressing Cry1A.105/Cry2Ab2/Vip3A with 0–30%non-Bt seeds.Greenhouse tests investigated the performance of aabb and AaBb in seed blends of Cry1A.105/Cry2Ab2 with 0–30% non-Bt seeds.In pure non-Bt maize plots,after 9–13 d of neonates being released on the plants,0.39 and 0.65 larvae/plant survived with leaf injury ratings of 4.7 and 5.9(Davis's 1–9 scale)in the field and greenhouse,respectively.In contrast,live larvae and plant injury were virtually not observed on Bt plants across all planting patterns.Larval occurrence and plant injury by aabb on non-Bt plants were similar between seed blends and pure non-Bt plantings,suggesting that the blended refuges could provide an equivalent susceptible population as structured refuge under the test conditions.In the greenhouse,the two insect genotypes in seed blends performed similarly,indicating that the seed blends did not provide more favorable conditions for AaBb over aabb.The information generated from this study should be useful in managing S.frugiperda and evaluating if send blends could be suitable refuge options for Bt resistance management in the regions where the insect is a primary target pest.
