Background:The incidence of chronic obstructive pulmonary disease(COPD)complicated with invasive pulmonary aspergillosis(IPA)has increased in the last two decades.The mechanism underpinning susceptibility to and high mortality of COPD complicated with IPA is unclear,and the role of T helper cells 17(Th17 cells)in the compound disease remains unknown.Therefore,this study aimed to assess the function of Th17 cells in COPD combined with IPA.Methods:COPD,IPA,and COPD+IPA mouse models were established in male wild type C57/BL6 mice.The amounts of Th17 cells and retinoic acid-related orphan receptorsγt(RORyt)were tested by flow cytometry.Then,serum interleukin(IL)-17 and IL-23.levels were detected by enzyme-linked immunosorbent assay(ELISA)in the control,COPD,IPA and COPD+IPA groups.In addition,COPD+IPA was induced in IL-17 knockout(KO)mice,for determining the role of Th17 cells in COPD+IPA.Results:Compared with the COPD group,the COPD+IPA group showed higher amounts of blood RORyt([35.09±16.12]%vs.[17.92±4.91]%,P=0.02)and serum IL-17(17.96±9.59 pg/mL vs.8.05±4.44 pg/mL,P=0.02),but blood([5.18±1.09]%vs.[4.15±0.87]%,P=0.28)and lung levels of Th17 cells(1.98±0.83]%vs.[2.03±0.98]%,P=0.91),lung levels of RORyt([9.58±6.93]%vs.[9.63±5.98]%,P=0.49)and serum IL-23(51.55±27.82 pg/mL us.68.70±15.20 pg/mL,P=0.15)showed no significant differences.Compared with the IPA group,the COPD+IPA group displayed lower amounts of blood([5.18±1.09]%vs.[9.21±3.56]%,P=0.01)and lung Th17 cells([1.98±0.83]%vs.[6.29±1.11]%,P=0.01)and serum IL-23(51.55±27.82 pg/mL vs.154.90±64.60 pg/mL,P=0.01)and IL-17(17.96±9.59 pg/mL uUs.39.81±2.37 pg/mL,P=0.02),while comparable blood([35.09±16.12]%Vs.[29.86±15.42]%,P=0.25)and lung levels of RORγt(9.58±6.93]%VUS,[15.10±2.95]%,P=0.18)were found in these two groups.Finally,Aspergillus load in IL-17 KO COPD+IPA mice was almost 2 times that of COPD+IPA mice(1,851,687.69±944,480.43"vs.892,958.10±686,808.80,t=2.32,P=0.02).Conclusion:These findings indicate that Th17 cells might be involved in the pathogenesis of COPD c

During deepwater managed pressure drilling(MPD),the gas kick may occur in abnormally high-pressure formations.If the traditional well control method is adopted,the treatment time is long and the advantage of early gas kick detection of MPD is lost.The dynamic managed pressure well-control(MPWC)method can be used to rapidly treat gas kick in deepwater MPD.In this paper,considering the effect of large-variable-diameter annulus and complex wellbore temperature in deepwater drilling,a simplified model of non-isothermal gas-liquid two-phase flow was established for dynamic deepwater MPWC simulation.Using this model,the response characteristics of outlet flow and wellhead backpressure were investigated.The results indicated that the gas fraction,outlet liquid flow rate,pit gain and wellhead backpressure presented complex alternating characteristics when gas moved upwards in the wellbore due to the large-variable-diameter annulus.The outlet liquid flow rate would be lower than the inlet flow rate and the pit gain would decrease before the gas moved to the wellhead.The variation trend of the wellhead backpressure was consistent with that of the pit gain.When the gas-liquid mixture passed through the choke,the expansion or compression of the gas caused part of the choke pressure drop to be supplemented or unloaded,delaying the response rate of the wellhead backpressure.The wellbore temperature,borehole diameter and seawater depth had different effects on outlet flow rate,pit gain and wellhead backpressure.This research could provide a new idea for well control methods in deepwater managed pressure drilling.

The multiphase flow characteristic is one of the most concerning problems during solid fluidization exploitation of marine natural gas hydrate reservoirs.In this research,a new transient gas-liquid-solid multiphase flow model with hydrate phase transition was developed.Meanwhile,this model considered the coupling relationship among convective heat transfer,hydrate dynamic decomposition,and multi-phase flow.The model can simulate the change of flow pattern from solid-liquid to gas-liquid-solid flow,and describe the distribution character of volume fraction of phase,wellbore temperature and pressure,and hydrate decomposition rate during transportation.The simulation results indicate that the hydrate decomposition region in the wellbore gradually expands,but the hydrate decomposition rate gradually decreases during the solid fluidization exploitation of hydrate.When mining time lasts for 4 h,and the bottom hole pressure decreases by about 0.4 MPa.Increasing NaCl concentration in seawater helps expand hydrate decomposition regions and improves the wellbore hydrate decomposition rate.When the Nacl mass fraction in seawater reaches 15%,it will raise the hydrate decomposition regions to the whole wellbore.In addition,the higher the wellhead backpressure,the lower the decomposition area and decomposition rate of hydrate in the wellbore.When wellhead backpressure reaches 2 MPa,the volume fraction of gas near the wellhead will reduce to about 12%.This work is expected to provide a theoretical basis for the development of marine hydrate reservoirs.

Single-hole hollow polymer nanospheres were fabricated by raspberry-like template method using ＂graft-from＂ strategy through atom transfer radical polymerization （ATRP）. Nanometer-sized silica spheres were covalently attached onto the surfaces of micrometer-sized silica spheres. Crosslinked polymer shells on the nano-sized spheres outside the attached area were formed by ＂graft-from＂ strategy through ATRP. After removal of the silica cores, single-hole hollow crosslinked polymer nanospheres were obtained. In this strategy, most of ATRP monomers may be used and thus many functional groups can be easily incorporated into the single-hole hollow crosslinked polymer nanospheres.

Phlorizin(PHL)is a natural compound with strong antioxidant properties mainly found in apples.In this paper,the interaction mechanism of PHL with pepsin and trypsin was comparatively evaluated by computer simulation,fluorescence spectra,circular dichroism(CD),and Fourier transform infrared(FT-IR)spectra at a molecular level.Fluorescence spectra showed that PHL quenches the pepsin/trypsin by static quenching.Thermodynamic parameters indicated that PHL binds to pepsin mainly through hydrogen bonds and van der Waals forces,and that of trypsin was electrostatic forces.The ground state complexes PHL and protease have a moderate affinity of 105 L/mol PHL binds more strongly to trypsin than to pepsin.CD and FT-IR spectra results showed that pepsin/trypsin decreased theβ-sheet content and slightly changed its secondary structure upon PHL.These experimental results are mutually verified with the predicted computer-aid simulation results.Upon PHL and trypsin binding,the antioxidant capacity of PHL was elevated.Nevertheless,the antioxidant capacity of PHL was decreased after binding to pepsin.This work elucidates the binding of PHL binding mechanisms to pepsin/trypsin and provides useful information for the digestion of PHL to improve the application of PHL in food processing.

Continuous cooling transformation diagrams in synthetic weld heat-affected zone(SH-CCT diagrams)show the phase transition temperature and hardness at different cooling rates,which is an important basis for formulating the welding process or predicting the performance of welding heat-affected zone.However,the experimental determination of SH-CCT diagrams is a time-consuming and costly process,which does not conform to the development trend of new materials.In addition,the prediction of SHCCT diagrams using metallurgical models remains a challenge due to the complexity of alloying elements and welding processes.So,in this study,a hybrid machine learning model consisting of multilayer perceptron classifier,k-Nearest Neighbors and random forest is established to predict the phase transformation temperature and hardness of low alloy steel using chemical composition and cooling rate.Then the SH-CCT diagrams of 6 kinds of steels are calculated by the hybrid machine learning model.The results show that the accuracy of the classification model is up to 100%,the predicted values of the regression models are in good agreement with the experimental results,with high correlation coefficient and low error value.Moreover,the mathematical expressions of hardness in welding heat-affected zone of low alloy steel are calculated by symbolic regression,which can quantitatively express the relationship between alloy composition,cooling time and hardness.This study demonstrates the great potential of the material informatics in the field of welding technology.

Regenerating spent graphite(SG)from retired lithium-ion batteries(LIBs)can effectively avoid resource waste.However,the technology is challenged by the impurity content and energy consumption.In this study,micro-expanded graphite(MEG)was synthesized by one-step oxidation method using waste LIBs anode graphite as material and perchloric acid as intercalation and oxidant agent.Then,its performance as a LIBs anode material were investigated as well as the greenhouse gas(GHG)emissions of the whole process were calculated.Perchloric acid was successfully embedded in the SG during the reaction,which effectively removed the impurities in the graphite.Defects introduced during intercalation and delamination,such as nanopores and intercrystalline cracks.Both provide additional space for Li ions during charging and discharging,thereby promoting capacity enhancement.The prepared MEG expresses a rate capability as high as 340.32 m Ah/g at a current density of 0.1 C and still retains 81.73%of the capacity after 100 cycles at a current density of 1 C.Additionally,the GHG emissions of the synthesis process of this article and other literatures are compared.The results demonstrated that perchloric acid treatment process provides a low-carbon,time-and energy-saving approach for regenerated SG as battery grade material.

Objective:To investigate the protective effects and underlying mechanisms of Xuebijing Injection(XBJ)on the lung endothelial barrier in hydrogen sulfide(H2S)-induced acute respiratory distress syndrome(ARDS).Methods:Sprague-Dawley rats were exposed to H2S(300 ppm)to establish ARDS model,while human pulmonary microvascular endothelial cells(HPMECs)were incubated with NaHS(a H2S donor,500μmol/L)to establish cell model.H2S and XBJ were concurrently administered to the rat and cell models.Lung hematoxylin and eosin staining,immunohistochemistry,transmission electron microscopy and wet/dry ratio measurement were used to confirm ARDS induced by H2S in vivo.The expression levels of claudin-5,phosphorylated protein kinase B(p-AKT)/t-AKT and p-forkhead box transcription factor O1(FoxO1)/t-FoxO1 in vivo and in vitro were also assessed.Paracellular permeability and transepithelial electrical resistance(TEER)were measured to evaluate endothelial barrier function in the cell model.Results:The morphological investigation showed that XBJ attenuated H2S-induced ARDS in rats.XBJ significantly ameliorated both the reduction in TEER and the increased paracellular permeability observed in NaHS-treated HPMECs(P<0.05).The protective effects of XBJ were blocked by LY294002,a phosphatidylinositol 3-kinase(PI3K)/AKT/FoxO1 pathway antagonist(P<0.05).Furthermore,XBJ promoted the expression of claudin-5 and increased the levels of p-AKT and p-FoxO1 in vivo and in vitro(P<0.05).Conclusion:XBJ ameliorated H2S-induced ARDS by promoting claudin-5 expression via the PI3K/AKT/FoxO1 signaling pathway.
