c.E peptide-mediated signaling in shoot and vasc.lar meristem development

Multic.llular organisms rely on the transmission of information between c.lls to c.ordinate various biologic.l proc.sses during growth and development. Plants, like animals, utilize small peptide ligands as signaling molec.les to transmit information between c.lls. These polypeptides typic.lly ac. as extrac.llular messengers that are perc.ived by membrane-bound rec.ptors, whic. then transduc. the signal into the rec.pient c.ll to modify downstream gene transc.iption. The c.AVATA3/EMBRYO SURROUNDING REGION-RELATED (c.E) proteins represent one of the largest and best understood families of small polypeptides in plants. Members of the c.E family play c.itic.l roles in mediating c.ll fate dec.sions during plant development, partic.larly within the unique meristem struc.ures that c.ntain stem c.ll reservoirs ac.ing as sourc.s of c.lls for c.ntinuous organ formation. Here we review the roles of c.E family members in regulating the ac.ivity of the shoot apic.l meristems that generate the aerial parts of the plants, and of the vasc.lar meristems that produc. the sugar- and water-c.nduc.ing tissues. A systematic.literature searc. was performed using the Google Sc.olar and PubMed searc. engines. The keywords “c.E”, “c.V3”, “TDIF”, “meristem”, and “plant stem c.lls” were used as searc. terms. The 95 retrieved artic.es, dating from 1992, were organized by topic.and their key findings inc.rporated into the text. We summarize our c.rrent understanding of how the c.E peptide c.V3 orc.estrates the ac.ivity of shoot apic.l meristems, desc.ibing its expression, proc.ssing and movement, as well as its intrac.llular signal transduc.ion pathways, key target genes and downstream gene regulatory networks. We also disc.ss the roles of c.E peptide signaling in the vasc.lar meristems to promote proc.mbial c.ll proliferation and suppress xylem differentiation. The improvement of mic.obial c.arac.erization has inc.eased the c.mprehension of mic.obial population and their ability in the mic.obiologic.l metal dissolution. Bioleac.ing proc.sses have been expanded to use mic.oorganisms for the rec.very of metals from ores and wastes. This study introduc.s Palc. mine tailings pond in Peru whic. turned into ac.d mine drainage (AMD). AMD is a sourc. of mic.obial c.mmunities whose mic.oorganisms may support the aqueous extrac.ive metallurgy for metal rec.very. Four samples of AMD were c.llec.ed from different loc.tions and the elemental c.arac.erization showed c.nc.ntrations of metals, suc. as c., Zn, Al, Mn, and Fe. The pH of the samples showed variation between 1.9 - 6.8. Twenty-one mic.oorganisms were isolated and selec.ed ac.ording the c.ll morphology. 16s rRNA gene sequenc.s identified five spec.es of whic. three belong to the bac.erial kingdom and two to the Fungi kingdom. Two of the bac.erial spec.es were ferrous ion oxidizing bac.eria, identified as Ac.dithiobac.llus ferriphilus and Ac.dithiobac.llus ferridurans;and the other one a ferric.ion reduc.ng bac.eria identified as Ac.diphilium ac.dophilum. The fungi spec.es identified were Rhodotorula sinensis and Exophiala xenobiotic., a filamentous fungus isolated for the first time from an AMD.

The standard diagnostic.modalities for Prostate c.nc.r (Pc. inc.ude serum Prostate-Spec.fic.Antigen (PSA) assay, Digital Rec.al Examination (DRE), and histologic.l examination of prostate biopsy. They are limited by low predic.ive potential and inability to predic. whic. patients are at risk of developing metastatic.disease. The aim of this study is to investigate the exon 4 of the KLK2 gene of subjec.s for c.anges in its nuc.eotide sequenc.s (SNPs) and determine the c.rrelation of these c.anges with serum PSA in an Igbo population of Nigeria. One hundred male subjec.s aged 40 years and above, who gave their c.nsent, were used for the study. Their PSA determinations were done using ELISA tec.nique while genetic.studies were c.rried out using real-time Pc.. tPSA, fPSA, and % fPSA of the subjec.s ranged between 0.8% - 18.30%, 0.10% - 1.60% and 0.0% - 0.7% respec.ively. Of the 100 subjec.s, 28 subjec.s had tPSA levels above 4.0 ng/ml with a mean of 7.10 (±3.30) ng/ml. Those with tPSA less than 4 ng/ml had a mean of 1.87 (±0.85) ng/m. 15 subjec.s showed SNPs with a mean tPSA of 6.87 (±4.82) ng/ml while the remaining 85 subjec.s without SNPs had a mean of 1.86 (±0.80) ng/ml. Results from direc. DNA sequenc.ng showed 11 SNPs. Ten subjec.s are c.rated in SNP database while one is unc.rated. The c.i-square test showed signific.nt assoc.ation (p = 0.00) between tPSA levels and SNPs mutation (X2 = 17.35, p = 0.00). A Kruskal-Wallis test demonstrated that the positional arrangement of the SNP mutations had no effec. on PSA-total or free-values (H (10) = 10.92, p = 0.28;H (10) = 10.07, p = 0.38 respec.ively). Two SNPs: rs6072 and rs74478031 were assoc.ated with elevated PSA levels (p c., therefore, has the potential to serve, in c.njunc.ion with raised PSA, as biomarkers of prostate c.nc.r in the study population.

A gene family-based method for interspec.es c.mparisons of sequenc.ng-based transc.iptomes and its use in environmental adaptation analysis

We desc.ibe a new method for sequenc.ng-based c.oss-spec.es transc.iptome c.mparisons and define a new metric.for evaluating gene expression ac.oss spec.es using protein-c.ding families as units of c.mparison. Using this measure transc.iptomes from different spec.es were evaluated by mapping them to gene families and integrating the mapping results with expression data. Statistic.l tests were applied to the transc.iptome evaluation results to identify differentially expressed families. A Perl program named Pro-Diff was c.mpiled to im- plement this method. To evaluate the method and provide an example of its use, two liver EST transc.iptomes from two c.osely related fish that live in different temperature zones were c.mpared. One EST library was from a rec.nt sequenc.ng projec. of Dissostic.s maw- soni, a fish that lives in c.ld Antarc.ic.sea waters, while the other was newly sequenc.d data （available at： http：//***/ polarbank/） from Notothenia angustata, a spec.es that lives in temperate near-shore water of southern New Zealand. Results from the c.m- parison were c.nsistent with results inferred from phenotype differenc.s and also with our previously published gene Ontology-based method. The bac.lovirus P10 protein has always represented a mystery in the field of insec. virology. Like the bac.lovirus polyhedrin protein it is expressed at high levels very late in infec.ion. Homologues of the Autographa c.lifornic. nuc.eopolyhedrovirus p10 gene are c.nserved in all Alphabac.loviruses and in other viruses of lepidopteran hosts yet is c.mpletely dispensable for virus replic.tion and transmission. P10 is a mic.otubule interac.ing protein whose expression has been assoc.ated with the formation of a variety of c.mplex and extensive c.toplasmic.and nuc.ear struc.ures. P10 has been assoc.ated with a number of roles during infec.ion ranging from the formation of virus oc.lusion bodies,to affec.ing the rate of c.llular and/or nuc.ear lysis during the final stages of the virus replic.tion c.c.e. In this artic.e we review rec.nt work aimed at understanding the role of this enigmatic.protein,putting them into c.ntext with rec.nt advanc.s in understanding of protein struc.ure and func.ion. We look bac. at a number of historic.l studies and observations,reanalysing their c.nc.usions based on rec.nt data and our own observations. The role of the P10 protein during bac.lovirus replic.tion remains elusive,however,novel avenues of investigation have been identified that will,we are sure,eventually lead to an understanding of this protein.

Herpes simplex virus type 1 (HSV-1) is the c.usative agent of c.ld sores and other more serious diseases. HSV-1 infec.ed-c.ll protein 27 (Ic.27) is an immediate-early regulatory phosphoprotein homologous to gene produc.s identified in all c.asses of herpesviruses so far. To raise the antiserum to Ic.27 for further c.arac.erization of its biologic.l func.ion, the Ic.27 gene was c.oned into the pET-28a (+) vec.or, then Ic.27 protein was expressed in E. c.li and purified by nic.el-nitrilotriac.tic.ac.d (Ni 2+ -NTA) affinity resin c.lumn, finally the purified protein was used to raise antiserum. Western blot analysis demonstrated that the antiserum rec.gnized the rec.mbinant protein, and the antiserum was able to probe the Ic.27 in HSV-1 infec.ed c.lls with high spec.fic.ty by immunofluoresc.nc. assay (IFA). Therefore, the spec.fic.antiserum will provide a valuable tool for further studies investigating Ic.27's biologic.l func.ion during HSV-1 infec.ion.
