RNA interference(RNAi)is a powerful tool that post-transcriptionally silences target genes in eukaryotic ***,silencing efficacy varies greatly among different insect ***,we met with little success when attempting to knock down genes in the mirid bug Apolygus lucorum via dsRNA *** disappearance of double-stranded RNA(dsRNA)could be a potential factor that restricts RNAi ***,we found that dsRNA can be degraded in midgut fluids,and a dsRNase of ***(AldsRNase)was identified and *** alignment indicated that its 6 key amino acid residues and the Mg2+-binding site were similar to those of other insects’*** signal peptide and endonuclease non-specific domain shared high sequence identity with the brown-winged green stinkbug Plautia stali *** showed high salivary gland and midgut expression and was continuously expressed through the whole life cycle,with peaks at the 4th instar ecdysis in the whole *** purified AldsRNase protein obtained by heterologously expressed can rapidly degrade *** comparing the substrate specificity of AldsRNase,3 specific substrates(dsRNA,small interfering RNA,and dsDNA)were all degraded,and the most efficient degradation is ***,immunofluorescence revealed that AldsRNase was expressed in the cytoplasm of midgut *** cloning and functional study of AldsRNase,the enzyme activity and substrate specificity of the recombinant protein,as well as the subcellular localization of nuclease,the reason for the disappearance of dsRNA was explained,which was useful in improving RNAi efficiency in *** and related species.

Owing to the broad energy coverage of Insight-HXMT in the hard X-ray band,we detected the highest energy of pulsation exceeding 200 keV around the 2017–2018 outburst peak of the first Galactic pulsating ultraluminous X-ray source (PULX) Swift J0243.6+6124,which is the highest energy detected from PULXs to *** also obtained the highest energy of pulsation of every exposure during the outburst in 2017–2018,and found the highest energy is roughly positively correlated with *** our newly developed method,we identified the critical luminosity being 4×10^(38)erg s^(-1) when the main peaks of the low and high energy pulse profiles became aligned,which separates the fan-beam dominated and pencil-beam dominated accretion *** the critical luminosity,the phase of the main peak shifted gradually from 0.5 to 0.8 until the outburst peak in al energy bands is reached,which is in agreement with the phase shift found previously at low *** result is consistent with what is derived from spectral analysis.

Haemaphysalis longicornis ticks,commonly found in East Asia,can transmit various pathogenic viruses,includingthe severe fever with thrombocytopenia syndrome virus(SFTSV)that has caused febrile diseases among humansin Hubei ***,understanding of the viromes of *** was limited,and the prevalence ofviruses among *** ticks in Hubei was not well *** study investigates the viromes of bothengorged(fed)and free(unfed)*** ticks across three mountainous regions in Hubei Province from 2019to ***-sequencing analysis identified viral sequences that were related to 39 reference viruses belonging tounclassified viruses and seven RNA viral families,namely Chuviridae,Nairoviridae,Orthomyxoviridae,Parvoviridae,Phenuiviridae,Rhabdoviridae,and *** abundance and diversity in these ticks were analysed,andphylogenetic characteristics of the Henan tick virus(HNTV),Dabieshan tick virus(DBSTV),Okutama tick virus(OKTV),and Jingmen tick virus(JMTV)were elucidated based on their full genomic *** demonstrated that DBSTV was the most common virus found in individual *** ticks(12.59%),followed by HNTV(0.35%),whereas JMTV and OKTV were not *** results improve our understanding of *** tick viromes in central China and highlight the role of tick feeding status and geographyin shaping the viral *** findings of new viral strains and their potential impact on public health raisethe need to strengthen surveillance efforts for comprehensively assessing their spillover potentials.

Coptis chinensis Franch.,also named Chinese goldthread is a member of Ranunculaceae in the order Ranunculales and represents an important lineage of early eudicots with traditional medicinal *** our study,by using syntenic analysis combined with phylogenomic analysis of *** and four other representative genomes from basal and core eudicots,we confirmed that the WGD event in *** was shared by Aquilegia coerulea and Papaver somniferum *** quickly occurred after Ranunculales diverged from other eudicots,likely a Ranunculales common tetraploidization(RCT).The synonymous nucleotide substitutions at synonymous sites distribution of syntenic blocks across these genomes showed that the evolutionary rate of the *** genome is faster than that of the *** genome by approximately 13.7%,possibly due to Papaveraceaes having an additional special tetraploidization event(PST).After Ks correction,the RCT dated to 115—130 million years ago(MYA),which was close to the divergence of Ranunculaceaes and Papaveraceaes approximately115.45—130.51 ***,we identified homologous genes related to polyploidization and speciation and constructed multiple sequence alignments with different reference ***,the event-related subgenomes in the basal genomes all showed genomic fractionation bias,suggesting a likely allopolyploid nature of the RCT,PST and T-Alpha and T-Beta events in Tetracentron *** addition,we detected that the sixteen P450 subfamilies were markedly expanded in the genomes of Ranunculales,and most of them were related to the RCT and PST *** constructed a new platform for Early Eudicot Comparative genomic Research(http://***/***)to store more *** summary,our findings support the WGD of *** shared by Ranunculales,which is likely an allotetraploidization *** present effort offered new insights into the evolution of key polyploidization events and the genes related to second

电弧增材制造具有沉积效率高、成本低、沉积形状和尺寸不受限制等优点,而目前电弧增材制造成型件的成型精度难以精确保证。沉积层尺寸作为评价构件成型质量的标准之一,对判断加工质量以及缺陷补偿至关重要。实时监测电弧增材制造过程中沉积层尺寸的变化状态,对于优化工艺参数,确保增材制造构件的成型质量具有重要意义。电弧光谱信息可以反映电弧状态,电弧状态与成型质量密切相关,因此研究电弧光谱与沉积层尺寸的关系具有重要意义。以钛合金(TC4)材料作为基板和焊丝,电弧等离子体光谱信号为研究对象,研究GTAW增材电弧光谱特性与沉积层尺寸的相关性。搭建光谱采集系统,采集熔池上方、熔池外围、钨极下方不同位置的电弧光谱信号。基于谱线分离性高原则,分别选取波长为404.20 nm的TiⅠ谱线、波长为416.36 nm的TiⅡ谱线、波长为420.20、434.81、480.50和487.98 nm的ArⅡ谱线以及波长为696.54和794.82 nm的ArⅠ谱线,提取其谱线的峰强特征,结合ReliefF算法分别挖掘不同谱线强度特征与沉积层尺寸的相关性。结果表明,三组位置的所有谱线中熔池上方的波长为404.03 nm的TiⅠ元素谱线、416.36 nm的TiⅡ元素谱线以及794.82 nm的ArⅠ元素谱线谱峰强度特征与沉积层尺寸具有较强的相关性。分别研究相同位置的不同谱线峰强特征与沉积层尺寸的相关性差异,结果表明熔池上方与沉积层尺寸相关性最大特征谱线为波长696.54 nm的ArⅠ谱线、熔池外围和钨极下方与沉积层尺寸相关性最大的特征谱线为波长794.82的ArⅠ谱线。为减小随机误差,采用PCA算法将三个电弧光谱采集位置上与沉积层尺寸相关性最大的谱线对应的强度特征进行融合,获得新的融合特征,结合K近邻算法建立沉积层尺寸预测模型,分别计算这四个特征预测样本类别的准确率,发现融合特征预测样本所属的沉积层尺寸的准确率更高。基于此新特征结合阈值分割法实现动态监测沉积层尺寸变化。
