The effect of 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR) on fatty acid oxidation in hepatocytes isolated from neonatal piglets

In the present study, the effect of 5-aminoimidazole-4-carboxamide ribonucleoside （AICAR） on long-chain fatty acid oxidation by hepatocytes isolated from suckled neonatal pig liver （a low ketogenic and lipogenic tissue） was tested Incubation of hepatocytes with AICAR （0.5 raM） in the presence of ] mM of carnitine and 10 mM of glucose for 1 hour at 37℃ had no significant effect on total [1-14C]-palrnitate （0.5 mM） oxidation （14CO2 and 14C-Acid soluble products （ASP））. Consistent with the fatty acid oxidation, carnitine palmitoyltransferase I activity and inhibition of its activity by malonyI-CoA （10 MM） assayed in cell homogenate also remained constant. However, addition of AICAR to the hepatocytes decreased 14CO2 production by 18% compared to control （p 〈 0.06）. The reduction of labeled carboxylic carbon accumulated in C02 caused a significant difference in distribution of oxidative products between 14C02 and 14C-ASP （p 〈 0.03） compared with the control. It was also noticed that acetyI-CoA carboxylase （ACC） was increased by AICAR （p 〈 0.03）, indicating that ACC might drive acetyI-CoA toward fatty acid synthesis pathway and induce an increase in distribution of fatty acid carbon to 14C-ASP. Addition of insulin to hepatocyte incubations with AICAR did not change the oxidative product distribution between CO2 and ASP, but further promoted ACC activity. The increased ACC activity was 70% higher than in the control group when citrate was absent in the reaction medium and was 30% higher when citrate was present in the medium. Our results suggest that AICAR may affect the distribution of metabolic products from fatty acid oxidation by changing ACC activity in hepatocyte isolated from suckled neonatal piglets; however, the basis for the increase in ACC activity elicited by AICAR is not apparent.

This article is part exposition of a recent rather technical paper of the last two authors on matrix pavings related to the 1959 Kadison-Singer Extension Problem and part a report on further computational results providing new bounds on the paving parameters for classes of small matrices investigated there and subsequently. A website maintained by the authors provides to all interested the matrices experimentally discovered that yield these bounds along with the proprietary MATLAB software with simple operational directions to load them, pave them, and perform paving searches. The convergence to 1 or not of the infinite sequences of these paving parameters in most cases is equivalent to the Kadison-Singer Extension Problem, and in all cases convergence to 1 negates the problem. The last two sections describe the search process and an interpretation of the data integrated with the results of the precursor to this paper.

背景:在与关节软骨有关的组织工程研究中,应用未脱钙和未染色的样本对于保留关节软骨的三维结构十分有利。当分析大量样本时,应用未脱钙的组织块将减少实验时间和复杂性。目的:检验在未脱钙、未染色的组织块上研究关节软骨形态的可能性,这样可以避免切割并染色薄切片。方法:取新西兰白兔和Sardinian绵羊的股骨髁,在多聚甲醛溶液中固定,乙醇中脱水,包埋于多异丁烯酸甲酯中,切成组织块,并染色。将金钯溅镀于标本上,然后用jeol jSM 6310电子显微镜进行观察。将背向散射和二次发射扫描电镜对组织块相同部位的观察结果进行合并分析。结果与结论:在兔的关节软骨组织块上,可明显区分未钙化与钙化软骨的传代,但较难辨认位于Ⅱ区和Ⅲ区的小软骨陷窝。在绵羊的软骨组织块中,则能较为容易地区分关节软骨各区和细胞结构。应用背向散射电镜能容易分辨出钙化软骨结构和潮标。合并二次发射扫描电镜和背向散射电镜,可在样本上观察到大的腔道。这些腔道穿过软骨下骨和钙化软骨,终止于未钙化的软骨。结果表明,应用背向散射和二次发射扫描电镜能在未脱钙和未染色的包埋组织块上观察到关节软骨的三维结构。

Three-dimensional(3D)printing is a useful technique that allows the creation of objects with complex structures by deposition of successive layers of *** materials are often from fossil ***,efforts are being made to produce environmentally friendly materials for 3D *** addition of lignocellulosic fibres to a polymer matrix is one of the alternatives to replace,for instance,glass fibres in composites as reinforcing *** fields of biocom-posites and 3D printing open innovative application areas for pulp fibres from the pulp and paper *** this work,biocomposites of poly(lactic acid)(PLA),poly(hydroxyalkanoate)(PHA)and kraft pulp fibres were prepared in order to find a suitable formulation for filaments for 3D *** effect of two different types of kraft fibres(bleached(B)and unbleached(U))and of PHA on the mechanical and thermal properties of the biocomposites was *** addition of 30%kraft fibres to PLA resulted in an increase of the tensile modulus from 3074 to∼4800 *** the case of biocomposites containing PHA(50%PLA/20%PHA/30%kraft)the increase in modulus was more moderate(PLA+PHA+U:3838 MPa,and PLA+PHA+B:3312 MPa).The tensile strength of PLA(66 MPa)increased to 77 MPa in PLA+kraft biocomposites,while a reduc-tion in strength was observed for PLA+PHA+U(43 MPa)and PLA+PHA+B(32 MPa).Filaments prepared with PLA,PHA and bleached and unbleached pulp fibres showed similar printability of complex geometries,demonstrating that unbleached pulp fibres could also be utilized in the preparation of biocomposites with good mechanical performance and 3D printing properties.

Myogenic contractions of the heart of the female blood-feeding insect, Rhodnius prolixus （Stal）, are inhibited by crude extracts of testes applied directly to isolated dorsal vessels. Dorsal vessels were observed with a stereo microscope and heart beats timed with a stopwatch. In normal Rhodnius saline, hearts contract at 14.8 4- 7.1 beats per minute （n = 45）. Crude extracts of the testes and the two male reproductive accessory organs （the opaque and transparent accessory glands） were prepared from previously frozen tissue by homogenizing 5-20 glands in a small glass homogenizer containing Rhodnius saline, centrifuging for 5 rain at 2 000 g, and collecting the supernatant. Testes extract as low as 1.0 glands per mL inhibit contractions whereas crude extracts of the opaque or transparent accessory glands have no consistent effect. We refer to this cardiac inhibitor as rhodtestolin （Rhodnius testis inhibitory factor）, and discuss its possible effects on the female during copulation.

The advances in research and treatment of autism spectrum disorders(ASD) over the past three decades have focused largely on early childhood and school-age years. Although ASD is a lifelong condition, there has been relatively little attention paid to ASD during the adolescent and adulthood periods. As the population of those with ASD continues to rise and age, the need to provide research and treatment for this group has become increasingly evident. This paper reviews the current literature available on symptoms, functioning, and treatment of adolescents and adults with ASD, as well as the unique issues that arise for individuals with ASD after childhood. Adulthood outcomes for ASD are generally poor, even for those with average to above average cognitive ability. Further research and additional clinical resources are needed for this rapidly increasing group.
