Boron Isotopic Fractionation and Trace element Incorporation in Various Species of Modern Corals in Sanya Bay, South China Sea

The boron isotope paleo-pH proxy has been extensively studied due to its potential for understanding past climate change, and further calibrations were considered for accurate applications of the proxy because of significant variability related to biocarbonate microstructure. In this work, we studied the boron isotopic fractionation between modern marine corals and their coexisting seawater collected along shallow area in Sanya Bay, South China Sea. The apparent partition coefficient of boron（KD） ranged from 0.83×10-3 to 1.69×10-3, which are in good agreement with previous studies. As the analyzed coral skeleton（~5 g） spanned the growth time period of 1–2 years, we discussed the boron isotopic fractionation between pristine corals and modern seawater using the annual mean seawater pH of 8.12 in this sea area. Without taking the vital effect into account,（11B/10B）coral values of all living corals spread over the curves of（11B/10B）borate vs.（11B/10B）sw with the α4-3 values ranging from 0.974 to 0.982. After calibrating the biological effect on the calcifying fluid pH, the field-based calcification on calcifying fluid pH（i.e., Δ（pHbiol-pHsw）） for coral species of Acropora, Pavona, Pocillopora, Faviidae, and others including Proites are 0.42, 0.33, 0.36, 0.19, respectively, and it is necessary to be validated by coral culturing experiment in the future. Correlations in B/Ca vs. Sr/Ca and B/Ca vs. pHbiol approve temperature and calcifying fluid pH influence on skeletal B/Ca. Fundamental understanding of the thermodynamic basis of the boron isotopes in marine carbonates and seawater will strengthen the confidence in the use of paleo-pH proxy as a powerful tool to monitor atmospheric CO2 variations in the past.

Temperature profiles down to 1500m(CTD) collected by Academia Sinica from 1986 to 1990 are used and discussed in relation to the dynamic heights at130 e across the North equatorial Current (NeC). An extremely high correlation between subsurface (say at 400 m depth) temperature and dynamic height relative to 1500 db is found, and the corresponding regression relationships suggest a method to estimate gpostrophic circulation from subsurface temperature alone. These suggest that the conclusions from extensive studies on this topic in Australian waters also apply to the NeC region, at least at130 e , thus making the subsurface thermal structure an excellent indicator of the variation of the NeC.

The Turtleback Diagram for Conditional Probability

We elaborate on an alternative representation of conditional probability to the usual tree diagram. We term the representation “turtleback diagram” for its resemblance to the pattern on turtle shells. Adopting the set theoretic view of events and the sample space, the turtleback diagram uses elements from Venn diagrams—set intersection, complement and partition—for conditioning, with the additional notion that the area of a set indicates probability whereas the ratio of areas for conditional probability. Once parts of the diagram are drawn and properly labeled, the calculation of conditional probability involves only simple arithmetic on the area of relevant sets. We discuss turtleback diagrams in relation to other visual representations of conditional probability, and detail several scenarios in which turtleback diagrams prove useful. By the equivalence of recursive space partition and the tree, the turtleback diagram is seen to be equally expressive as the tree diagram for abstract concepts. We also provide empirical data on the use of turtleback diagrams with undergraduate students in elementary statistics or probability courses.

Validation of a radioimmunoassay-based fecal corticosteroid assay for Richardson＇s ground squirrels Urocitellus richardsonii and behavioural correlates of stress

We validated a radioimmunoassay-based method quantifying fecal glucocorticoid metabolites (FGMs) from captive male and female Richardson’s ground squirrelsUrocitellus richardsonii. Blood samples were drawn to explore the correlation between plasma cortisol and FGM concentrations. We also injected groups of squirrels with normal saline (CTL; control), adre-nocorticotropic hormone (ACTH; stimulating adrenal activity), or dexamethasone (DeX; suppressing adrenal activity). Potential correlations between stress and behaviour were explored through quantification of fecal pellet production and the intervention necessary to elicit defecation, as well as the behaviour of subjects in the context of handling. Changes in plasma cortisol concentration between capture (baseline), and following handling (stress-induced) were also quantified for free-living squirrels. While glucocorticoid concentrations recovered from feces during our captive-animal study were not well correlated with plasma cortisol concentrations, and uncorrelated with defecation or behaviour, FGM concentrations did reflect the activation of the hypothalamic-pituitary-adrenal (HPA) axis. FGM concentrations increased significantly during initial captivity, but declined to baseline level as individuals acclimated to the novel environment. Injection of subjects with ACTH increased FGMs above baseline, confirming activation of the HPA axis. Plasma cortisol concentrations increased significantly with induced stress, indicating that capture and handling activated the glucocorticoid stress response even among previously handled, free-living subjects. Our findings validate a non-invasive tool that will afford new insight into the physiological processes underlying social, reproductive and antipredator behaviour of Richardson’s ground squirrels.

Correlating Bladder Cancer Risk Genes with Their Targeting MicroRNAs Using MMiRNA-Tar

The Cancer Genome Arias （TCGA）（http：//***） is a valuable data resource focused on an increasing number of well-characterized cancer genomes. In part, TCGA provides detailed information about cancer-dependent gene expression changes, including changes in the expression of transcription-regulating microRNAs. We developed a web interface tool MMiRNA-Tar （http：f/***-Tar） that can calculate and plot the correlation of expression for mRNA-microRNA pairs across samples or over a time course for a list of pairs under different prediction confidence cutoff criteria. Prediction confidence was estab- lished by requiring that the proposed mRNA-microRNA pair appears in at least one of three target prediction databases： TargetProfiler, TargetScan, or miRanda. We have tested our MMiRNA-Tar tool through analyzing 53 tumor and 11 normal samples of bladder urothelial carcinoma （BLCA） datasets obtained from TCGA and identified 204 microRNAs. These microRNAs were correlated with the mRNAs of five previously-reported bladder cancer risk genes and these selected pairs exhib- ited correlations in opposite direction between the tumor and normal samples based on the cus- tomized cutoff criterion of prediction. Furthermore, we have identified additional 496 genes （830 pairs） potentially targeted by 79 significant microRNAs out of 204 using three cutoff criteria, i.e.,false discovery rate （FDR）〈 0.1, opposite correlation coefficient between the tumor and normal samples, and predicted by at least one of three target prediction databases. Therefore, MMiRNA- Tar provides researchers a convenient tool to visualize the co-relationship between microRNAs and mRNAs and to predict their targeting relationship. We believe that correlating expression profiles for microRNAs and mRNAs offers a complementary approach for elucidating their interactions.
