Water security is a widely concerned issue in the world nowadays.A new method,water poverty index(WPI),was applied to evaluate the regional water *** state farms in Heilongjiang Province,Northeastern China were selected to evaluate water security status b.sed on the data of 2006 using WPI and mean deviation grading *** method of WPI includes five key indices:resources(R),access(A),capacity(C),utilization(U)and environment(E).Each key index further consists of several *** to the results of WPI,the grade of each farm was calculated b. using the method of mean deviation ***,the radar images can b. protracted of each *** the radar images,the conclusions can b. drawn that the WPI values of Farm 853 and Hongqiling are under very safe status,while that of Farm Raohe is under safe status,those of Farms Youyi,597,852,291 and Jiangchuan are under moderate safe status,that of Farm b.ixing is under low safe status and those of Farm Shuangyashan,Shuguang and b.oshan are under unsafe *** results from this study can provide b.sic information for decision making on rational utilization of water resources and regulations for regional water safety guarantee system.

b.ckground:Early neurologic deterioration(END)may occur in patients with anterior circulation ischemic stroke(ACIS)after receiving endovascular treatment(EVT).Hemodynamic insufficiency,re-occlusion,and post-re-canalization hyper-perfusion are likely to play a critical role in *** hypothesized that hemodynamic changes can predict END in patients with ACIS postsuccessful EVT using trans-cranial Doppler(TCD).Methods:We utilized a prospectively maintained datab.se of ACIS patients treated with EVT b.tween Septemb.r 2016 and June 2018 in the Xuanwu Hospital,Capital Medical *** parameters including peak systolic velocity(PSV),b.lateral mean flow velocity(MFV),and pulse index(PI)were determined via the middle cereb.al arteries within 72 h post-EVT.A logistic regression model was applied to detect independent predictors for ***:Totally,112 EVT patients were included in this study and 80/112 patients experienced successful re-canalization withb.al edema(11/17)was considered as a leading role and followed b. symptomatic intra-cranial hemorrhage(4/17)and ischemia progression(2/17).For the 80 patients,the PSV(median:127 cm/s vs.116 cm/s,P=0.039),the ratio of ipsilateral-MFV/contra-lateral-MFV(iMFV/cMFV)(median:1.29 vs.1.02,P=0.036)and iMFV/mean b.ood pressure(Mb.)(median:0.97 vs.0.79,P=0.008)in END patients were higher than those of *** the receiver-operating characteristic curve to ob.ain cut-off values for PSV,PI,iMFV/cMFV,and iMFV/Mb. for END,we found that PI≥0.85(odds ratio:11.03,95%confidence interval:1.92–63.46,P=0.007)and iMFV/Mb.≥0.84(odds ratio:9.20,95%confidence interval:2.07–40.84,P=0.004)were independent predictors of END in a multivariate logistic regression model,with a sensitivity of 82.4%and 76.5%and a specificity of 42.9%and 66.7%,respectively,and had the positive predictive values of 29.0%and 38.2%,and negative predictive values of 90.0%and 91.

【目的】优化稳定期慢性阻塞性肺疾病(chronic ob.tructive pulmonary diseases,COPD)患者支气管肺泡灌洗液(b.onchoalveolar lavage fluids,b.LF)细菌宏基因组DNA的提取方法,以便于高效提取微量的细菌DNA进行后续的PCR反应和测序。【方法】取稳定期COPD患者的b.LF 5mL,离心收集细胞。为了有效提取样品中革兰氏阳性菌的基因组,对QIAGEN的DNA提取试剂盒的操作步骤进行优化:加入裂解缓冲液ATL后首先运用研磨珠和多功能生物样品匀质器破碎菌壁,再加入蛋白酶K孵育,然后加入裂解缓冲液AL振荡混匀。无水乙醇沉淀DNA后,将全部溶液过柱,用洗液AW1和AW2各洗柱一次,最后加50μL洗脱液洗脱DNA。提取的DNA定量后,运用PCR方法检测样本中的细菌16S rDNA量,并按照测序要求构建DNA文库进一步验证。【结果】试剂盒优化法提取的b.LF的DNA总量为467.5(135.0-1697.5)ng,明显高于按照传统酚-氯仿法提取的DNA总量95.0(0-612.5)ng,并且所提取的DNA可以很好的扩增细菌的16S rDNA以及构建DNA文库,改良后的扩增产物明显增多(P=0.002)。【结论】使用DNA提取试剂盒结合研磨珠和多功能生物样品匀质器破菌壁的方法能够更高效的提取b.LF中微量的宏基因组DNA,为进一步的测序和菌群分析打下基础。
