RNA binding proteins in spermatogenesis： an in depth focus on the musashi family

Controlled gene regulation during gamete development is vital for maintaining reproductive potential. During the complex process of mammalian spermatogenesis, male germ cells experience extended periods of the inactive transcription despite heavy translational requirements for continued growth and differentiation. Hence, spermatogenesis is highly reliant on mechanisms of posttranscriptional regulation of gene expression, facilitated by RNA binding proteins （RBPs）, which remain abundantly expressed throughout this process. One such group of proteins is the musashi family, previously identified as critical regulators of testis germ cell development and meiosis in Drosophila, and also shown to be vital to sperm development and reproductive potential in the mouse. This review describes the role and function of RBPs our recent knowledge of the musashi proteins in spermatogenesis. within the scope of male germ cell development, focusing on The functional mechanisms utilized by RBPs within the cell are outlined in depth, and the significance of sub-cellular localization and stage-specific expression in relation to the mode and impact of posttranscriptional regulation is also highlighted. We emphasize the historical role of the musashi family of RBPs in stem cell function and cell fate determination, as originally characterized in Drosophila and Xenopus, and conclude with our current understanding of the differential roles and functions of the mammalian musashi proteins, musashi-1 and musashi-2, with a primary focus on our findings in spermatogenesis. This review highlights both the essential contribution of RBPs to posttranscriptional regulation and the importance of the musashi family as master regulators of male gamete development.

AIm： To investigate genetic diversity of Helicobacter pylori （H. pylorl） cell division-related gene A （cdrA） and its effect on the host ***： Inactivation of H. py/ori cdrA, which is involved in ceil division and morphological elonga- tion, has a role in chronic persistent infections. Ge- netic property of H. pylori cdrA was evaluated using polymerase chain reaction and sequencing in 128 （77 American and 51 Japanese） clinical isolates obtained from 48 and 51 patients, respectively. Enzyme-linked immunosorbent assay was performed to measure in- terleukin-8 （IL-8） secretion with gastric biopsy speci- mens obtained from American patients colonized with cdrA-positive or -negative strains and AGS cells co- cultured with wild-type HPK5 （cdrA-positive） or its de- rivative HPKT510 （cdrA-disruptant）. Furthermore, the cytotoxin-associated gene A （cagA） status （transloca- tion and phosphorylation） and kinetics of transcription factors [nuclear factor-kappa B （NF-~：B） and inhibition kappa B] were investigated in AGS cells co-cultured with HPK5, HPKT510 and its derivative HPKSCA （cagA- disruptant） by western blotting analysis with immuno- precipitation. RESULTS： Genetic diversity of the H. pylori cdrA gene demonstrated that the cdrA status segregated into two categories including four allele types, cdrA-positive （al- lele types, I and 11 ） and cdrA-negative （allele types; 111 and IV） categories, respectively. Almost all Japanese isolates were cdrA-positive （ 1 ： 7.8% and 11 ： 90.2%）, whereas 16.9% of American isolates were cdrA-positive （11） and 83.1% were cdrA-negative （nl： 37.7% and IV： 45.5%）, indicating extended diversity of cdrA in individual American isolates. Comparison of each isolate from different regions （antrum and corpus） in the stomach of 29 Americans revealed that cdrA status was identical in both isolates from different regions in 17 cases. The digestive gland of the apple snail Pomacea canaliculata lodges two types of pigmented corpuscles(identified as C and K corpuscles)which has been proposed as endosymbiont/*** corpuscular types are always present in the digestive gland of adult snails,they are released into the tubuloacinar lumen and are later expelled in the *** their part,hatchlings lack any C or K corpuscles in the digestive gland as well as in their feces,whereas C corpuscles appear in both the gland and feces within one week after ***,it is possible that the detritivorous hatchlings acquire the putative C-endosymbiont from feces in the sediments where they live,***‘lateral’or‘horizontal’*** possibility was put to test in an experiment in which we prevented any lateral transmission,by a 7-days aseptic culture,with no food,of aseptically obtained *** the end of the experiment,we observed that most juveniles had survived the culture period,and hence the digestive glands and feces of survivors were studied by light microscopy of resin embedded,toluidine blue-stained *** studied glands and fecal samples showed C *** is concluded that lateral transmission of the endosymbiont,if any,is not indispensable for the acquisition of the endosymbiont by hatchlings.

An Ecological Evaluation of Vinyl Chloride Exposure and Liver Cancer Incidence and mortality in Texas

The goal of this analysis was to evaluate the association between county-level ambient vinyl chloride(VC)and county-level liver cancer incidence and mortality rates in *** county-level ambient VC data were obtained from the National Air Toxics ***-adjusted county-level liver cancer incidence rates were abstracted from the Texas Cancer Registry and age-standardized county-level liver cancer mortality rates were obtained from the peer-reviewed *** imputation was utilized to impute incidence rates in counties with suppressed liver cancer incidence *** binomial and Poisson regression models were utilized to evaluate the association between county-level ambient VC and county-level liver cancer incidence and mortality rates,respectively,adjusted for county-level heavy drinking prevalence,hepatitis mortality rates,median income,and race(percent Hispanic).County-level ambient VC was not associated with county-level liver cancer incidence or mortality ***,when compared to the lowest tertile of ambient VC,the middle(relative risk[RR]:1.06,95%confidence interval[CI]:0.95-1.19)and highest(RR:1.03,95%CI:0.90-1.17)tertiles of ambient VC were not associated with liver cancer ***,county-level ambient VC in the middle(RR:0.95,95%CI:0.85-1.05)and highest(RR:0.93,95%CI:0.82-1.05)tertiles were not associated with liver cancer *** analysis suggests that county-level ambient VC is not associated with liver cancer incidence or mortality in *** study provides novel results regarding liver cancer risk from low-level non-occupational exposure to ambient VC.

Holographic imaging offers a reliable and fast method to capture the complete three-dimensional （3D） information of the scene from a single perspective. We review our recently proposed single-channel optical system for generating digital Fresnel holograms of 3D real-existing objects illuminated by incoherent light. In this motionless holographic technique, light is reflected, or emitted from a 3D object, propagates through a spatial light modulator （SLm）, and is recorded by a digital camera. The SLm is used as a beamsplitter of the single-channel incoherent interferometer, such that each spherical beam originated from each object point is split into two spherical beams with two different curve radii. Incoherent sum of the entire interferences between all the couples of spherical beams creates the Fresnel hologram of the observed 3D object. When this hologram is reconstructed in the computer, the 3D properties of the object are revealed.

In spray atomization and codeposition, a molten stream of metal is disintegrated into a fine dispersion of droplets by high velocity gas jets. The resulting semi-solidified droplets are directed towards a substrate where they impact and collect as rapidly solidified splats. Relatively high rates of solidification are achieved as a result of the thinness of the splats and the rapid heat extraction during flight and upon impacting with the substrate. The processing method uses codeposition of the metallic semi-solidified droplets (metallic matrix) with the injected reinforcement ceramic particles. In the present paper, the microstructures, mechanical properties, interfacial properties, thermal stability and aging behaviour of spray atomized and codeposited Al-Li-X mmC's (injected X=SiC, Al2O3) are reported and correlated to the processing conditions.
