Protein profiles of enzymatically isolated rumen epithelium in sheep fed a fibrous diet

Background: The rumen wall plays a major role in efficient transfer of digested nutrients in the rumen to peripheral tissues through the portal venous system. Some of these substrates are metabolised in the epithelium during this process. To identify the specific proteins involved in these processes, we used proteomic technologies. Protein extracts were prepared from ventral rumen tissue of six sheep fed a fibrous diet at 1.5× maintenance energy *** a newly developed method, we were able to enzymatically isolate the epithelial cells from underlying tissue layers, thus allowing cytosol and membrane fractions to be independently analysed using liquid chromatography tandem mass spectrometry(lC MS/MS).Results: Using our procedure we identified 570 epithelial proteins in the Ovis aries sequence database. Subcellular locations were largely cytosolic(n = 221) and extracellular(n = 85). However, a quarter of the proteins identified were assigned to the plasma membrane or organelle membranes, some of which transport nutrients and metabolites. Of these 91 were transmembrane proteins(TMHMM), 27 had an N-terminal signal peptide(signalP) and TMHMM motif,13 had a glycosylphosphatidylinositol(GPI) anchor and signalP sequence, 67 had beta(β) strands or 17 β strands and a transit peptide sequence, indicating the identified proteins were integral or peripheral membrane proteins. Subunits of the 5 protein complexes involved in mitochondrial cellular energy production were well represented. Structural proteins(15%), proteins involved in the metabolism of lipids and proteins(26%) and those with steroid or cytokine action were a feature of the ***: Our research has developed a procedure to isolate rumen epithelium proteins from the underlying tissue layers so that they may be profiled using proteomic technologies. The approach improves the number of proteins that can be profiled that are specific to the epithelium of the rumen wall. It provides new insights

Objective:To determine the relationship between asymptomatic malaria parasitemia and some oxidative stress parameters in pregnant Nigerian ***:This is a cross-sectional study involving 130 normal pregnant women at various trimesters,who were attending antenatal clinic at the University of Nigeria Teaching Hospital(UNTH) and Kenechukwu Specialist Hospital in Enugu.A comparable group(control),made of 30 non pregnant women was also *** a 24 hour dietary recall,serum levels of vitamin A,C and malondialdehyde(MDA) were determined by colorimetric method,while vitamin E was determined by absorptiometric ***:There were no statistically significant differences in age,parity,estimated calorie,vitamins A,C and E intake between the pregnant and non pregnant groups(P>0.05).The serum level of the vitamins(umol/l) and MDA(umol/l) in control,1st,2nd and 3rd trimesters respectively were:(l)Vitamin A:1.6±0.36 vs 0.6±0.26 vs 0.62±0.33 vs 0.46±0.21(P<0.0001);(2) Vitamin C:75.65±14.15 vs 62.97±24.4 vs 37.85±15.19 vs 28.94±8.52(P<0.0001);(3) Vitamin E:3.01±1.32 vs 3.45±2.01 vs 9.36±2.75 vs 9.82±2.97(P<0.0001);(4) MDA:1.42±0.02 vs 1.61±0.02 vs 1.79±0.02 vs 2.03±0.05(P<0.0001).However,there were no significant changes in the serum level of the vitamins and MDA between the positive and the negative parasitemia subjects(P>0.05). Conclusions:Asymptomatic malaria parasitemia does not induce additional oxidative stress on pregnant women in *** enormity of acute and complicated attack should be further investigated.

为明确呋虫胺和唑虫酰胺微乳剂在茶叶种植、烘干、浸泡过程中的残留特征,在山东、浙江等10地开展规范农药登记残留田间试验,并进行加工因子校正膳食风险评估模型比较研究。样品经体积分数5%甲酸-乙腈提取、盐析分层、分散固相萃取净化后,经超高效液相色谱-串联质谱法检测,外标法定量。结果表明:在0.005~0.500μg/ml范围内,呋虫胺及其代谢物、唑虫酰胺质量浓度与其响应峰面积呈良好线性关系(R^(2)>0.99),在0.05、1.00、20.00、50.00 mg/kg(茶鲜叶、茶干叶),0.05、0.10、5.00 mg/kg(茶汤)添加水平下,呋虫胺及其代谢物1-甲基-3-[(3-四氢呋喃)甲基]脲、1-甲基-3-[(3-四氢呋喃)甲基]二氢胍盐的平均加标回收率为74%~103%,相对标准偏差为1%~9%;唑虫酰胺的平均加标回收率为75%~95%,相对标准偏差为1%~6%,方法定量限均为0.05 mg/kg;20%呋虫胺·唑虫酰胺微乳剂按推荐剂量施药后,呋虫胺(含代谢物)在茶鲜叶中的规范残留试验中值(supervised trials median residue,STMR)经烘干因子校正后与茶干叶中STMR相应的国家估算每日摄入量(national estimated daily intake,NEDI)均为4.08 mg,占每日允许摄入量(allowable daily intake,ADI)的32.4%;唑虫酰胺在茶鲜叶中的STMR经烘干因子校正后的NEDI为0.17581 mg,占ADI的46.5%,茶干叶中STMR对应的NEDI为0.18925 mg,占ADI的50.1%,呋虫胺和唑虫酰胺在不同评估模型下获得的长期膳食暴露风险均低于100%;呋虫胺(含代谢物)经加工因子校正后的国际短期膳食摄入量(international estimation of short-term dietary intake,IESTI)与校正前处于同一风险水平,低于急性参考剂量(acute reference dose,ARfD)(1 mg/kg);唑虫酰胺的IESTI为3.46×10^(-4) mg/kg(采收间隔7 d),低于ARf D(0.01 mg/kg),本研究方法在茶汤中未检测到唑虫酰胺残留,故未得到浸泡因子及相应的IESTI。综上,按推荐剂量在茶树上使用20%呋虫胺·唑虫酰胺微乳剂是安全的,最终残留量对应的长期及急性膳食摄入风险均在可接受范围,利用加工因子校正评估模型更切合实际风险水平。

The United Nation’s Strategic Approach to International Chemicals Management (SAICM)’s vision states that"...by 2020,chemicals are produced and used in ways that minimize significant adverse impacts on human health and the environment."The chemical industry endeavors to conduct safety evaluations of its products and make the information available to the public in relatively simple *** an industry toxicologist,I intend to introduce the principles of chemical management,step-by-step approach on risk characterization of chemicals,and technologies and concepts involved in the *** Global Product Strategy(GPS) will be introduced,which was developed by the International Council of Chemical Associations(ICCA) as part of its commitment to *** is part of the chemical industry’s voluntary Responsible Care Global Charter,to help companies to promote the safe use of chemical products and enhance product stewardship throughout the value chain.A key component of GPS is a Risk Assessment Guidance document,which provides a step-by-step process for companies to conduct risk assessments of *** summary,we first select chemicals,gather information,allocate to tiers based on exposure and hazard information,and then close data gaps by conducting additional testing or evaluations of chemicals to characterize hazards and *** hazard characterization information is then integrated with exposure assessments in order to derive a risk characterization,the outcome of which is documented in the GPS Safety ***(intelligent) Testing Strategy and animal alternative testing methods and Klimisch system for study rating are also *** is considered as a potential risk assessment and communication tool for chemicals,especially for developing economies and small to medium sized companies.

Early Triassic microbialites from the Changxing Region of Zhejiang Province,South China

Microbialites, often considered as a signal of extreme marine environment, are common in the lower Triassic strata of South China where they flourished in the aftermath of the end-Permian mass extinction. Early Triassic microbialite facies are known to vary palaeogeographically, perhaps due to differing climates, ocean chemistry, and water depths. This paper provides the first record of a brief, but spectacular development of microbialites in the aftermath of the end-Permian mass extinction at Panjiazhuang section, Changxing Region of Zhejiang Province (eastern South China). Here, the Upper Permian Changxing Formation comprises typical shallow platform facies rich in calcareous algae and foraminifera, the development of which was terminated by the major end-Permian regression. A 3.4-m-thick microbialite began to form at the onset of the transgression in the earliest Triassic. The microbialite at Panjiazhuang section is composed of thrombolite that contains abundant calcified cyanobacteria, small gastropods, microconchid tubes and ostracods, representing a low-diversity shallow marine community in the aftermath of the end-Permian crisis. The microbialites are succeeded by thin-bedded micrites bearing thin-shelled bivalves, which record a rapid sea-level rise in the Early Triassic. Abundant populations of small pyrite framboids are observed in the upper part of the microbialites and the overlying thin-bedded micrites, suggesting that dysoxic water conditions developed at that time. The appearance of microbialites near the Permian-Triassic boundary (PTB) at Panjiazhuang section was the result of peculiar marine conditions following the end-Permian regression, whilst their disappearance was due to the increasing water depth and the development of dysoxia.
