Objective To evaluate the chemical composition and effects of Artemisia vulgaris(AV)hydroalcoholic extract(HEAV)on breast cancer cells(MCf-7 and SKBR-3),chronic myeloid leukemia(K562)and NIH/3T3 *** Phytochemical analysis of HEAV was done by high-performance liquid chromatography-mass(HPLC)*** and cell death studies were performed using trypan blue and Annexin/fITC-7AAD,***-1(fer-1)and necrostatin-1(Nec-1)were used to assess the mode of HEAV-induced cell death and acetoxymethylester(BAPTA-AM)was used to verify the involvement of cytosolic calcium in this *** calcium measurements were made using *** HEAV decreased the viability of MCf-7,SKBR-3 and K562 cells(Pf HEAV-treated K562 cells was reduced compared to HEAV-exposed fibroblasts(Pf K562 cells with HEAV induced cell death primarily by late apoptosis and necrosis in assays using annexin V-fITC/7-AAD(Pf Nec-1 and fer-1 increased the viability of K562 cells treated with HEAV relative to cells exposed to HEAV alone(Pfrom lysosomes in K562 cells(Pfurthermore,BAPTA-AM,an intracellular Ca^(2+)chelator,decreased the number of non-viable cells treated with HEAV(Pf cell death,which are partially dependent on lysosomal release of Ca^(2+).These effects may be related to artemisinin and caffeoylquinic acids,the main compounds identified in HEAV.

Improving wood volume predictions in dry tropical forest in the semi-arid Brazil

The volumetric variability of dry tropical forests in Brazil and the scarcity of studies on the subject show the need for the development of techniques that make it possible to obtain adequate and accurate wood volume *** this study,we analyzed a database of thinning trees from a forest management plan in the Contendas de SincoráNational forest,southwestern Bahia State,*** data set included a total of 300 trees with a trunk diameter ranging from 5 to 52 ***,validation and statistical selection of four volumetric models were *** to the difference in height values for the same diameter and the low correlation between both variables,we do not suggest models which only use the diameter at breast height(DBH)variable as a predictor because they accommodate the largest estimation *** comparing the best single entry model(Hohenald-Krenn)with the Spurr model(best fit model),it is noted that the exclusion of height as a predictor causes the values of 136.44 and 0.93 for Akaike information criterion(AIC)and adjusted determination coefficient(R2 adj),which are poorer than the second best model(Schumacher-Hall).Regarding the minimum sample size,errors in estimation(root mean square error(RMSE)and bias)of the best model decrease as the sample size increases,especially when a larger number of trees with DBH≥15.0 cm are randomly *** sampling by diameter class produces smaller volume prediction errors than random sampling,especially when considering all *** summary,the Spurr and Schumacher-Hall models perform *** models suggest that the total variance explained in the estimates is not less than 95%,producing reliable forecasts of the total volume with *** estimates indicate that the bias around the average is not greater than 7%.Our results support the decision to use regression methods to build models and estimate their parameters,seeking stratification strategies in diameter classes for the sample trees.

应用近红外光谱分析技术（NIR）测定土壤参数具有快速、方便的特点。文章分析了不同含水率、不同颗粒大小的土壤样本在不同测试角、不同测试高度对土壤光谱的影响，并得到了不同含水率和不同粒径土壤的含氮量预测模型。研究了这些因素对含氮率测量的影响，分析了NIR技术在田间实地应用预测的可能性。研究表明，光谱仪在距土壤高度为100mm，测试角为45。时，具有最大的吸光度。土壤粒径和含水率这2个参数明显影响，当粒径在0．5～5mm变化时，含氮量预测相关系数r为0．81左右，当土壤粒径在〈o．25和〉5mm模型的预测能力变差。当土壤样品呈天然潮湿状态时，氮的预测结果较差。而样品干燥以后，预测相关系数较高。为土壤原位光谱测试提供了依据。

Ammonium Ion Adsorption and Settleability Improvement Achieved in a Synthetic Zeolite-Amended Activated Sludge

Municipal wastewater treatment plants typically exhibit two classic problems： high ammonium concen- tration in water after conventional biological treatment and, in some cases, poor activated sludge sediment ability. Potential solutions to these problems were investigated by adding a synthetic zeolite obtained from coal fly ash to different steps of activated sludge treatment. The experimental results for ammonium removal fit well with the theoretical adsorption isotherms of the freundlich model with a maximum adsorption capacity of 13.72 mg.g-＇. Utiliza- tion of this kind of zeolite to improve activated sludge sediment ability is studied for the first time in this work. It is found that the addition of the zeolite （1 g. L-1） to an activated sludge with settling problems significantly enhances its sediment ability and comoact ability. This is confirmed by the sludge volume index （SVI）, which was reduced from 163 ml.g-1 to 70 ml.g-r, the V60 value, which was reduced from 894 ml.L-1 to 427 ml.L-1, and the zeta poten- tial （0, which was reduced from -19.81 mV to -14.29 mV. The results indicate that the addition of this synthetic zeolite to activated sludge, as an additional waste management practice, has a positive impact on both ammonium removal and sludge settleability.

Melon is as an alternative model to understand fruit ripening due to the coexistence of climacteric and non-climacteric varieties within the same species,allowing the study of the processes that regulate this complex trait with genetic *** phenotyped a population of recombinant inbred lines(RILs),obtained by crossing a climacteric(Védrantais,cantalupensis type)and a non-climcteric variety(Piel de Sapo T111,inodorus type),for traits related to climacteric maturation and ethylene *** in the RIL population exhibited various combinations of phenotypes that differed in the amount of ethylene produced,the early onset of ethylene production,and other phenotypes associated with *** characterized a major QTL on chromosome 8,ETHQV8.1,which is sufficient to activate climacteric ripening,and other minor QTLs that may modulate the climacteric *** ETHQV8.1 allele was validated by using two reciprocal introgression line populations generated by crossing Védrantais and Piel de Sapo and analyzing the ETHQV8.1 region in each of the genetic backgrounds.A Genome-wide association study(GWAS)using 211 accessions of the *** further identified two regions on chromosome 8 associated with the production of aromas,one of these regions overlapping with the 154.1 kb interval containing *** ETHQV8.1 region contains several candidate genes that may be related to fruit *** work sheds light into the regulation mechanisms of a complex trait such as fruit ripening.

Objective: Although Angelica archangelica is a medicinal and aromatic plant with a long history of use for both medicinal and food purposes, there are no studies regarding the antineoplastic activity of its *** study aimed to evaluate the cytotoxicity and antitumor effects of the crude extract of A. archangelica root(CEAA) on breast ***: The cytotoxicity of CEAA against breast adenocarcinoma cells(4T1 and MCf-7) was evaluated by a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay. Morphological and biochemical changes were detected by Hoechst 33342/propidium iodide(PI) and annexin V/PI staining. Cytosolic calcium mobilization was evaluated in cells staining with fURA-4 NW. Immunoblotting was used to determine the effect of CEAA on anti-and pro-apoptotic proteins(Bcl-2 and Bax, respectively).The 4 T1 cell-challenged mice were used for in vivo ***: Using ultra-high-performance liquid chromatography–mass spectrometry analysis, angelicin, a constituent of the roots and leaves of A. archangelica, was found to be the major constituent of the CEAA evaluated in this study(73 mg/mL). The CEAA was cytotoxic for both breast cancer cell lines studied but not for human fibroblasts. Treatment of 4 T1 cells with the CEAA increased Bax protein levels accompanied by decreased Bcl-2 expression, in the presence of cleaved caspase-3 and cytosolic calcium mobilization, suggesting mitochondrial involvement in breast cancer cell death induced by the CEAA in this cell line. No changes on the Bcl-2/Bax ratio were observed in CEAA-treated MCf7 cells. Gavage administration of the CEAA(500 mg/kg) to 4T1 cell-challenged mice significantly decreased tumor growth when compared with untreated ***: Altogether, our data show the antitumor potential of the CEAA against breast cancer cells in vitro and in vivo. further research is necessary to better elucidate the pharmacological application of the CEAA in breast cancer therapy.

High-throughput phenotyping of two plant-size traits of Eucalyptus species using neural networks

In forest modeling to estimate the volume of wood,artificial intelligence has been shown to be quite effi-cient,especially using artificial neural networks(ANNs).Here we tested whether diameter at breast height(DBH)and the total plant height(Ht)of eucalyptus can be pre-dicted at the stand level using spectral bands measured by an unmanned aerial vehicle(UAV)multispectral sensor and vegetation *** do so,using the data obtained by the UAV as input variables,we tested different configurations(number of hidden layers and number of neurons in each layer)of ANNs for predicting DBH and Ht at stand level for different Eucalyptus *** experimental design was randomized blocks with four replicates,with 20 trees in each experimental *** treatments comprised five Eucalyptus species(***,***,***,***-dis,and ***)and Corymbria *** and Ht for each plot at the stand level were measured seven times in separate overflights by the UAV,so that the multispectral sensor could obtain spectral bands to calculate vegetation indices(VIs).ANNs were then constructed using spectral bands and VIs as input layers,in addition to the categorical variable (species), to predict DBH and Ht at the stand level simultaneously. This report represents one of the first appli-cations of high-throughput phenotyping for plant size traits in Eucalyptus species. In general, ANNs containing three hidden layers gave better statistical performance (higher esti-mated r, lower estimated root mean squared error-RMSE) due to their greater capacity for self-learning. Among these ANNs, the best contained eight neurons in the first layer, seven in the second, and five in the third (8 − 7 − 5). The results reported here reveal the potential of using the gener-ated models to perform accurate forest inventories based on spectral bands and VIs obtained with a UAV multispectral sensor and ANNs, reducing labor and time.
