Objective:In order to study if Trichostatin A facilitate Arsenic Trioxide to induce differentiation of HL-60 cells and its underlying ***:the alteration in the cell proliferation was determined by MTT *** reduction assay and CD11b analysis by FACS were used to detect the differentiation of HL60 *** cycle arrest was measured by *** expressions of m RNA of P21,P27 and Cyclin D3,Cyclin E in HL60 cells after exposure to TSA and(or)Arsenic Trioxide were evaluated by semi-quantitative ***:It was found that the combinationof TSAand Arsenic Trioxide could inhibit proliferation and induce differentiation of HL60 cells obviously than Arsenic Trioxide *** up-regulationof the m RNA expression of P21,P27 but down-regulation of the expression of Cyclin D3,Cyclin E were related to the differentiation of ***:TSA has synergistic effect with Arsenic Trioxide in inducing differentiation of HL60 and it’s associated with the change of P21,P27 and Cyclin D3,Cyclin E expression.

CD44 is highly expressed in human acute myeloid leukemia （AML） cells. Some experiments had shown that it was possible to reverse differentiation blockage in AML cells by CD44 ligation with specific antibodies, indicating that CD44 was closely related to the differentiation of leukemia cells. The differentiation of acute promyelocytic leukemia cell line HL-60 cells could be induced by all trans-retinoic acid （ATRA） and hexamethylene bisacetamide （HMBA）, but so far the mechanism was not demonstrated clearly. In the present study, we investigated whether ATRA or HMBA induced the growth arrest of HL-60 cells by down-regulating the expression of CD44. The results indicated that the proliferation of HL-60 cells was obviously inhibited and the differentiation was induced by both ATRA and HMBA. The decreased expression of CD44 and cyclin E mRNA, and the increased expression of p27 and p21 at mRNA levels were observed. Furthermore, there was a negative correlation between the expression of CD44 and p27. It was concluded that ATRA and HMBA played a role in the differentiation induction of HL-60 cells, which was mediated by the down-regulation of CD44, accompanied by down-regulation of cyclin E, and up-regulation of p27 and p21 mRNA. 目的探讨Notch信号通路抑制药物r分泌酶抑制剂（γ-secretase inhibtor，GSI）对小鼠异基因骨髓移植（allo-BMT）后急性移植物抗宿主病（aGVHD）发生发展的影响。方法用C57BL／6小鼠作为供鼠，BALB／c小鼠作为受鼠，建立小鼠同种allo-BMT模型。BALB／c小鼠随机被分为6组：（1）实验组1：受者经60Co射线照射后输注供者的骨髓细胞与脾细胞，并于移植前第1天，移植后第1、3天分别给受鼠腹腔注射GSI 5μmol／kg；（2）实验组2：受者经。Co射线照射后输注供者的骨髓细胞与脾细胞，并于移植后第1、2、3天分别给受鼠腹腔注射GSI5μmol／妇；（3）移植对照组：受者经。co射线照射后输注供者的骨髓细胞与脾细胞，并分别于移植前第1天和移植后第1、3天给受鼠腹腔注射二甲基亚砜；（4）单纯移植组：受者经60Co射线照射后输注供者的骨髓细胞与脾细胞；（5）单纯照射组：小鼠经60Co射线照射后不进行移植；（6）空白对照组：小鼠未作任何处理。观察各组小鼠的生长情况，监测外周血白细胞数和血小板数，测定移植后第7天时血白细胞介素4（IL-4）和7干扰素（IFN-γ）水平，观察小鼠的肝脏和小肠组织的病理变化。结果实验组1及实验组2的aGVHD发生率分别为60％（6／10）、50％（5／10），均明显低于移植对照组的90％（9／10，P％0．01）。实验组1及实验组2的小鼠存活时间分别为（25．4±3．1）d、（25．2±3．4）d，均明显长于移植对照组的（17．4±2．3）d（P〈0．01）。实验组1及实验组2发生aGVHD的受鼠小肠及肝脏病理改变明显轻于移植对照组。实验组1及实验组2受鼠移植后第7天时的血清IL-4浓度分别为（31．71±1．60）ng／L、（29．27±1．86）ng／L，均高于移植对照组的（23．04±1．82）ng／L（P〈0．01，P〈0．05）；实验组1及实验组2血清IFN-γ浓度分别为（25．97±1．15）ng／L、（22．69±3．01）ng／L，均明显低于移植对照组的（36．77±2．62）ng／L（P〈0．01）。接受移植的小鼠移植后第7天白细胞和血小板降至最低，然后缓慢回升，至研究结束，实验组1及实验组2小鼠的白细胞和血小板仍明显低于正常值（P〈0．01，P〈0．05）。结论GSI能够抑制小鼠allo-BMT后致死性aGVHD的发生，其机制可能与GSI抑制IFN-γ分泌，同时促进IL-4分泌有关。

目的探讨Notch1 siRNA对体内外人骨髓瘤细胞RPMI-8226细胞硼替佐米药物敏感性的影响。方法体外采用Notch1 siRNA转染RPMI-8226细胞,CCK-8实验检测细胞增殖及硼替佐米药物的敏感性;Western blot检测各组细胞Notch1蛋白表达变化;将RPMI-8226细胞皮下注射于NOD/SCID小鼠,建立人多发性骨髓瘤(MM)小鼠移植瘤模型,将成瘤小鼠分为三组:NS+bortezomib(Notch1 siRNA转染联合硼替佐米)组;CS+bortezomib(Control siRNA转染联合硼替佐米)组;UN+bortezomib(硼替佐米)组,观察各组肿瘤体积变化,免疫组化染色法观察Notch1变化,TUNEL法检测细胞凋亡。结果 Notch1 siRNA有效下调骨髓瘤细胞RPMI-8226细胞Notch1蛋白表达;Notch1 siRNA在96 h抑制细胞增殖作用明显增加,与CS及UN组比较对细胞增殖的作用可见明显差异(P<0.01);Notch1 siRNA转染组细胞对硼替佐米IC50值为1.21μmol/L,与Control siRNA转染组及未转染组相比均存在统计学差异(P<0.01);Notch1 siRNA降低移植瘤Notch1蛋白表达,Notch1 siRNA转染组细胞的AI明显高于Control siRNA转染组及未转染组(P<0.01);Notch1 siRNA转染联合硼替佐米组肿瘤体积明显减小,13、17及21 d与Control siRNA转染联合硼替佐米组比较均有统计学差异(P<0.05)。结论体外实验Notch siRNA抑制人骨髓瘤细胞RPMI-8226细胞增殖增加硼替佐米的敏感性,体内试验证实Notch siRNA联合硼替佐米可以明显减小荷瘤MM小鼠肿瘤体积、增加凋亡,提示Notchl是治疗MM的有效分子靶点。

目的筛选影响接受诱导化疗急性髓系白血病(非M3)患者预后情况的主要风险因素,根据风险因素产生数学模型,预测其预后情况。方法对108例急性髓系白血病(非M3)患者进行回顾性研究,筛选年龄、细胞毒(药物)治疗史及全身状况等影响因素,构建Logistic回归模型,用该模型进行预测,与实际缓解情况比较,对缓解率做卡方检验,验证模型预测的准确性。结果 (1)单因素分析发现对急性髓系白血病(非M3)患者缓解有影响的变量为FLT3表达(χ2=23.336,P=0.000),CD34、HLADR共表达(χ2=6.783,P=0.000),NPM1表达(χ2=23.156,P=0.014),是否有骨髓增生异常综合征(MDS)前期病变(χ2=6.000,P=0.014),染色体核型(χ2=3.458,P=0.045)及乳酸脱氢酶(LDH)水平(χ2=2.472,P=0.015)。(2)多因素分析发现对急性髓系白血病(非M3)患者缓解有影响的因素为FLT3及NPM1表达[OR分别为0.042(95%CI 0.007~0.245)和9.287(95%CI 3.028~28.484),P值分别为0.000和0.000]。结论本试验中Logistic回归模型拟合较好,且具有一定的诊断价值。

The incidence of obesity has been rapidly increasing, and this condition has become a major public health threat. A substantial shift in environmental factors and lifestyle, such as unhealthy diet, is among the major driving forces of the global obesity pandemic. Longitudinal studies and randomized intervention trials have shown that genetic susceptibility to obesity may interact with dietary factors in relation to the body mass index and risk of obesity. This review summarized data from recent longitudinal studies and intervention studies on variations and diets and discussed the challenges and future prospects related to this area and public health implications.
