Neural progenitor cells derived from fibroblasts induced by small molecule compounds under hypoxia for treatment of Parkinson’s disease in rats

Neural progenitor cells(NPCs) capable of self-renewal and differentiation into neural cell lineages offer broad prospects for cell therapy for neurodegenerative diseases. However, cell therapy based on NPC transplantation is limited by the inability to acquire sufficient quantities of NPCs. Previous studies have found that a chemical cocktail of valproic acid, CHIR99021, and Repsox(VCR) promotes mouse fibroblasts to differentiate into NPCs under hypoxic conditions. Therefore, we used VCR(0.5 mM valproic acid, 3 μM CHIR99021, and 1 μM Repsox) to induce the reprogramming of rat embryonic fibroblasts into NPCs under a hypoxic condition(5%). These NPCs exhibited typical neurosphere-like structures that can express NPC markers, such as Nestin, SRY-box transcription factor 2, and paired box 6(Pax6), and could also differentiate into multiple types of functional neurons and astrocytes in vitro. They had similar gene expression profiles to those of rat brain-derived neural stem cells. Subsequently, the chemically-induced NPCs(ciNPCs) were stereotactically transplanted into the substantia nigra of 6-hydroxydopamine-lesioned parkinsonian rats. We found that the ciNPCs exhibited long-term survival, migrated long distances, and differentiated into multiple types of functional neurons and glial cells in vivo. Moreover, the parkinsonian behavioral defects of the parkinsonian model rats grafted with ciNPCs showed remarkable functional recovery. These findings suggest that rat fibroblasts can be directly transformed into NPCs using a chemical cocktail of VCR without introducing exogenous factors, which may be an attractive donor material for transplantation therapy for Parkinson’s disease.

Near-bed sediment transport in a heavily modified coastal plain estuary

Numerous estuaries of the world have been strongly modified by human activities.These interferences can make great adjustments of not only sediment transport processes,but also the collective behavior of the estuary.This paper provides a typical case of a heavily modified coastal plain estuary of Sheyang on the China coast,where a sluice barrage was built in 1956 to stop the intrusions of storm surges and saline water.Four sets of instrumented tripods were simultaneously deployed along a cross-shore transect to continuously observe near-bed flow currents and sediment transport.The in-situ surveys lasted over a spring and neap tide cycle when a strong wind event occurred in the neap tide.Comparisons of flows and sediment transport between tide-dominated and wind-dominated conditions demonstrated the important role of episodic wind events in flows and sediment transport.The wind-induced currents,bottom stresses,and sediment transport rates were significantly greater when wind was present than corresponding quantities induced by the tides.The long-shore sediment transport induced by winds exceeds the cross-shore component,especially near the river mouth bar.These results indicate the noticeable importance of wave-dominated coastal processes in shaping topographic features.A regime shift of estuarine evolution under highly intense human forcing occurs from fluvial to marine processes.This finding suggests that the management strategy of the estuarine system should focus on the restoration of estuarine processes,rather than the present focus on inhibition of marine dynamics.

Deformation behavior and microstructural evolution during hot compression of an a+b Ti-6.5Al-3.5Mo-1.5Zr-0.3Si alloy

The effect of processing parameters on the flow response and microstructural evolution of the a＋b titanium alloy Ti-6.5Al-3.5Mo-1.5Zr-0.3Si has been studied by conducting isothermal hot compressive tests at a strain rate of 0.01-10 s-1 at 860-1100°C. The true stress-true strain curves of the sample hot-compressed in the a＋b phase region exhibit a peak stress followed by continuous flow softening, whereas in the b region, the flow stress attains a steady-state regime. At a strain rate of 10 s-1, the alloy exhibits plastic flow insta-bilities. According to the kinetic rate equation, the apparent activation energies are estimated to be about 674-705 kJ/mol in the a＋b region and 308-335 kJ/mol in the b region, respectively. When deformed in the a＋b region, the globularization process of the a colony structure occurs, and a dynamic recrystallized microstructures are observed to show bimodal. Dynamic recrystallization can take place in the b region irrespective of starting deformed structures.

Effect of recombinant human endostatin onradiotherapy for esophagus cancer

Objective:To investigate the effect of radiotherapy plus recombinant human endostatin（RHendostatin） on esophageal cancer and its mechanism.Methods:A total of SO nudemice were equally randomized into control group,radiotherapy group,and combined therapy group Ⅰ,Ⅱ,and Ⅲ after inoculating with Ecal09 cell suspension（1×107 cells/mL）.On the day of grouping,control group and radiotherapy group were injected normal saline,while radiotherapy group and 3 combined therapy groups received radiotherapy;besides,combined therapy group Ⅰ,Ⅱ,and Ⅲ was injected RH-endostatin of 2.5,5,10 mg/kg respectively.After 3-week therapy,the tumors of each group were collected and microvessel density and VEGF expression in tumors were determined.In vitro,Eca109 cells were divided into control group,radiotherapy group,and combined therapy group.Forty-eight hours after treatment cell cycle distribution and apoptosis rate were detected,and the activity of VEGF signal paths was semiquantitatively analyzed.Results:Since the 6th day of treatment,the relative tumor proliferation rate of combined therapy group Ⅱ was lower than radiotherapy group（P<0.05） and 40%since the 15 th day.Average microvessel density and EGFR expression in combined therapy group Ⅱ were lower than radiotherapy group（P<0.05）.In vitro,the cell percentage in S and G2/M phase of combined therapy group cells was lower than that in radiotherapy group cells,while the apoptosis rate and the expression of VEGF,AKT,p-AKT,ERK1/2 and p-ERKl/2 in combined group were higher than that in radiotherapy group（P<0.05）.Conclusions:RH-endostatin promotes the efficacy of radiotherapy on esophageal cancer,which may be partly realized by inhibiting the activity of VEGF related signal paths.

Non-heading Chinese cabbage(NHCC)is an important leafy vegetable cultivated worldwide.Here,we report the first high-quality,chromosome-level genome of NHCC001 based on PacBio,Hi-C,and Illumina sequencing data.The assembled NHCC001 genome is 405.33 Mb in size with a contig N50 of 2.83 Mb and a scaffold N50 of 38.13 Mb.Approximately 53%of the assembled genome is composed of repetitive sequences,among which long terminal repeats(LTRs,20.42%of the genome)are the most abundant.Using Hi-C data,97.9%(396.83 Mb)of the sequences were assigned to 10 pseudochromosomes.Genome assessment showed that this B.rapa NHCC001 genome assembly is of better quality than other currently available B.rapa assemblies and that it contains 48,158 protein-coding genes,99.56%of which are annotated in at least one functional database.Comparative genomic analysis confirmed that B.rapa NHCC001 underwent a whole-genome triplication(WGT)event shared with other Brassica species that occurred after the WGD events shared with Arabidopsis.Genes related to ascorbic acid metabolism showed little variation among the three B.rapa subspecies.The numbers of genes involved in glucosinolate biosynthesis and catabolism were higher in NHCC001 than in Chiifu and Z1,due primarily to tandem duplication.The newly assembled genome will provide an important resource for research on B.rapa,especially B.rapa ssp.chinensis.

In recent years,computing art has developed rapidly with the in-depth cross study of artificial intelligence generated con-tent(AIGC)and the main features of artworks.Audio-visual content generation has gradually been applied to various practical tasks,including video or game score,assisting artists in creation,art education and other aspects,which demonstrates a broad application pro-spect.In this paper,we introduce innovative achievements in audio-visual content generation from the perspective of visual art genera-tion and auditory art generation based on artificial intelligence(Al).We outline the development tendency of image and music datasets,visual and auditory content modelling,and related automatic generation systems.The objective and subjective evaluation of generated samples plays an important role in the measurement of algorithm performance.We provide a cogeneration mechanism of audio-visual content in multimodal tasks from image to music and display the construction of specific stylized datasets.There are still many new op-portunities and challenges in the field of audio-visual synesthesia generation,and we provide a comprehensive discussion on them.

Hybrid pedestrian positioning system using wearable inertial sensors and ultrasonic ranging

Pedestrian positioning system(PPS)using wearable inertial sensors has wide applications towards various emerging fields such as smart healthcare,emergency rescue,soldier positioning,etc.The performance of traditional PPS is limited by the cumulative error of inertial sensors,complex motion modes of pedestrians,and the low robustness of the multi-sensor collaboration structure.This paper presents a hybrid pedestrian positioning system using the combination of wearable inertial sensors and ultrasonic ranging(H-PPS).A robust two nodes integration structure is developed to adaptively combine the motion data acquired from the single waist-mounted and foot-mounted node,and enhanced by a novel ellipsoid constraint model.In addition,a deep-learning-based walking speed estimator is proposed by considering all the motion features provided by different nodes,which effectively reduces the cumulative error originating from inertial sensors.Finally,a comprehensive data and model dual-driven model is presented to effectively combine the motion data provided by different sensor nodes and walking speed estimator,and multi-level constraints are extracted to further improve the performance of the overall system.Experimental results indicate that the proposed H-PPS significantly improves the performance of the single PPS and outperforms existing algorithms in accuracy index under complex indoor scenarios.

Effect of FeSi additive in dual-chamber sample cup on thermal analysis characteristic values and vermiculating rate of compacted graphite iron

Thermal analysis plays a key role in the online inspection of molten iron quality.Different solidification process of molten iron can be reflected by thermal analysis curves,and silicon is one of important elements affecting the solidification of molten iron.In this study,FeSi75 was added in one chamber of the dual-chamber sample cup,and the influences of FeSi75 additive on the characteristic values of thermal analysis curves and vermiculating rate were investigated.The results show that with the increase of FeSi75,the start temperature of austenite formation TALfirstly decreases and then increases,but the start temperature of eutectic growth TSEF,the lowest eutectic temperature TEU,temperature at maximum eutectic reaction rate TEM,and highest eutectic temperature TERkeep always an increase.The temperature at final solidification point TEShas little change.The FeSi75 additive has different influences on the vermiculating rate of molten iron with different vermiculation,and the vermiculating rate increases for lower vermiculation molten iron while decreases for higher one.According to the thermal analysis curves obtained by a dual-chamber sample cup with 0.30wt.%FeSi75 additive in one chamber,the vermiculating rate of molten iron can be evaluated by comparing the characteristic values of these curves.The time differenceΔtERcorresponding to the highest eutectic temperature TERhas a closer relationship with the vermiculating rate,and a parabolic regression curve between the time differenceΔtERand vermiculating rateηhas been obtained within the range of 65%to 95%,which is suitable for the qualified melt.

Meiosis is the process of producing haploid gametes through a series of complex chromosomal events and the coordinated action of various proteins.The mitochondrial protease complex(ClpXP),which consists of caseinolytic mitochondrial matrix peptidase X(ClpX)and caseinolytic protease P(ClpP)and mediates the degradation of misfolded,damaged,and oxidized proteins,is essential for maintaining mitochondrial homeostasis.ClpXP has been implicated in meiosis regulation,but its precise role is currently unknown.In this study,we engineered an inducible male germ cell-specific knockout caseinolytic mitochondrial matrix peptidase X(Clpx^(cKO))mouse model to investigate the function of ClpX in meiosis.We found that disrupting Clpx in male mice induced germ cell apoptosis and led to an absence of sperm in the epididymis.Specifically,it caused asynapsis of homologous chromosomes and impaired meiotic recombination,resulting in meiotic arrest in the zygotene-to-pachytene transition phase.The loss of ClpX compromised the double-strand break(DSB)repair machinery by markedly reducing the recruitment of DNA repair protein RAD51 homolog 1(RAD51)to DSB sites.This dysfunction may be due to an insufficient supply of energy from the aberrant mitochondria in Clpx^(cKO) spermatocytes,as discerned by electron microscopy.Furthermore,ubiquitination signals on chromosomes and the expression of oxidative phosphorylation subunits were both significantly attenuated in Clpx^(cKO) spermatocytes.Taken together,we propose that ClpX is essential for maintaining mitochondrial protein homeostasis and ensuring homologous chromosome pairing,synapsis,and recombination in spermatocytes during meiotic prophase I.

Scattered Co-anchored MoS2 synergistically boosting photothermal capture and storage of phase change materials

Pristine phase change materials（PCMs） suffer from inherent deficiencies of poor solar absorption and photothermal conversion.Herein,we proposed a strategy of co-incorporation of zero-dimensional（OD）metal nanoparticles and two-dimensional（2D） photothermal materials in PCMs for efficient capture and conversion of solar energy into thermal energy.Highly scattered Co-anchored MoS2nanoflower cluster serving as photon and phonon triggers was prepared by in-situ hydrothermal growth of ZIF67 polyhedron on 2D MoS2and subsequent high-temperature carbonization.After encapsulating thermal storage unit（paraffin wax）,the obtained composite PCMs integrated high-performance photothermal conversion and thermal energy storage capability.Benefiting from the synergistic enhancement of OD Co nanoparticles with localized surface plasmon resonance effect,carbon layer with the conjugation effect and 2D MoS2with strong solar absorption,composite PCMs exhibited a high photothermal conversion efficiency of 95.19%,Additionally,the resulting composite PCMs also demonstrated long-term thermal sto rage stability and durable structu ral stability after 300 thermal cycles.The proposed collabo rative co-incorporation strategy provides some innovative references for developing next-generation photothermal PCMs in solar energy utilization.
