As a dioxygenase. Ten-Eleven Translocation 2 （TET2） catalyzes subsequent steps of 5-methylcytosine （5mC） oxidation. TET2 plays a critical role in the self-renewal, proliferation, and differentiation of hei-natopoietic stem cells, but its impact on mature hematopoietic cells is not well-characterized. Here we show that Tet2 plays an essential role in osteoclastogenesis. Dele- tion of Tet2 impairs the differentiation of osteoclast precursor cells （macrophages） and their matu- ration into bone-resorbing osteoclasts in vitro. Furthermore, Tet2 / mice exhibit mild osteopetrosis, accompanied by decreased number of osteoclasts in vivo. Tet2 loss in macrophages results in the altered expression of a set of genes implicated in osteoclast differentiation, such as Cehpa, Mafb, and Nfkbiz. Tet2 deletion also leads to a genome-wide alteration in the level of 5-hydroxymethylcytosine （ShmC） and altered expression of a specific subset of macrophage genes associated with osteoclast differentiation. Furthermore, Tet2 interacts with Runxl and negatively modulates its transcriptional activity. Our studies demonstrate a novel molecular mechanism controlling osteoclast differentiation and function by Tet2, that is, through interactions with Runxl and the maintenance of genomie 5hmC. Targeting Tet2 and its pathway could be a potential therapeutic strategy for the prevention and t,＇eatment of abnormal bone mass caused by the deregulation of osteoclast activities.

The apoptosis of glomerular mesangial cells(GMC)in rat Thy-1 nephritis(Thy-1N),a model of human mesangioproliferative glomerulonephritis(MsPGN),is accompanied by sublytic C5b-9 *** the mechanism by which sublytic C5b-9 induces GMC apoptosis is *** the present studies,the effect of X-linked inhibitor of apoptosisassociated factor 1(XAF1)expression on GMC apoptosis and the role of p300 and interferon regulatory factor-1(IRF-1)in mediating XAF1 gene activation were determined both in the GMC induced by sublytic C5b-9(in vitro)and in the renal tissues of rats with Thy-1N(in vivo).The in vitro studies demonstrated that IRF-1-enhanced XAF1 gene activation and its regulation by p300-mediated IRF-1 acetylation were involved in GMC apoptosis induced by sublytic *** element of IRF-1 binding to XAF1 promoter and two acetylated sites of IRF-1 protein were also *** vivo,silence of p300,IRF-1 or XAF1 genes in the renal tissues diminished GMC apoptosis and secondary GMC proliferation as well as urinary protein secretion in Thy-1N ***,these data implicate that sublytic C5b-9 induces the expression of both p300 and IRF-1,as well as p300-dependent IRF-1 acetylation,which may contribute to XAF1 gene activation and subsequent GMC apoptosis in Thy-1N rats.

Exploring carbon emission effects based on the evolution of residents’ dietary structure to achieve the carbon neutrality goal and mitigate climate change is an important *** study took China as the research object(data excluding Hong Kong,Macao and Taiwan) and used the carbon emission coefficient method to quantitatively measure the food carbon emissions from 1987–2020,then analyzed the carbon emission effects under the evolution of dietary *** results showed that during the study period,the Chinese dietary structure gradually changed to a high-carbon consumption *** dietary structure of urban residents developed to a balanced one,while that of rural residents developed to a high-quality *** the study period,the per capita food carbon emissions and total food consumption of Chinese showed an increasing *** per capita food carbon emissions of residents in urban and rural showed an overall upward *** total food carbon emissions in urban increased significantly,while that in rural increased first and then *** influence of beef and mutton on carbon emissions is the highest in dietary *** with the balanced dietary pattern,the food carbon emissions of Chinese residents had not yet reached the peak,but were evolving to a high-carbon consumption pattern.

Trees progress through various growth stages, each marked by specific responses and adaptation strategies to environmental conditions. Despite the importance of age-related growth responses on overall forest health and management policies, limited knowledge exists regarding age-related effects on dendroclimatic relationships in key subtropical tree species. In this study, we employed a dendrochronological method to examine the impact of rapid warming on growth dynamics and climatic sensitivity of young (40-60 years) and old (100-180 years) Pinus massoniana forests across six sites in central-southern China. The normalized log basal area increment of trees in both age groups increased significantly following rapid warming in 1984. Trees in young forests further showed a distinct growth decline during a prolonged severe drought (2004-2013), whereas those in old forests maintained growth increases. Tree growth was more strongly influenced by temperature than by moisture, particularly in old forests. Spring temperatures strongly and positively impacted the growth of old trees but had a weaker effect on young ones. Old forests had a significantly lower resistance to extreme drought but faster recovery compared to young forests. The "divergence problem" was more pronounced in younger forests due to their heightened sensitivity to warming-induced drought and heat stress. With ongoing warming, young forests also may initially experience a growth decline due to their heightened sensitivity to winter drought. Our findings underscore the importance of considering age-dependent changes in forest/tree growth response to warming in subtropical forest management, particularly in the context of achieving "Carbon Peak & Carbon Neutrality" goals in China.

Synergic catalytic effect between active sites and supports greatly determines the catalytic activity for the aerobic oxidative desulfurization of fuel *** this work,Ni-doped Co-based bimetallic metal-organic framework(CoNi-MOF)is fabricated to disperse N-hydroxyphthalimide(NHPI),in which the whole catalyst provides plentiful synergic catalytic effect to improve the performance of oxidative desulfurization(ODS).As a bimetallic MOF,the second metal Ni doping results in the flower-like morphology and the modification of electronic properties,which ensure the exposure of NHPI and strengthen the synergistic effect of the overall *** with the monometallic Co-MOF and naked NHPI,the NHPI@CoNi-MOF triggers the efficient activation of molecular oxygen and improves the ODS performance without an *** sulfur removal of dibenzothiophene-based model oil reaches 96.4%over the NHPI@CoNi-MOF catalyst in 8 h of ***,the catalytic product of this aerobic ODS reaction is sulfone,which is adsorbed on the catalyst surface due to the difference in *** work provides new insight and strategy for the design of a strong synergic catalytic effect between NHPI and bimetallic supports toward high-activity aerobic ODS materials.
