LINE-1 family member GCRG123 gene is up-regulated in human gastric signet-ring cell carcinoma

AIM： To analyze the expression profiles of a human gastriccancer-related gene, GCRG123, in human gastric signetring cell carcinoma tissues, and to perform bioinformatics analysis on GCRG123.METHODS： In situ hybridization was used to explore the GCRG123 expression pattern in paraffin-embedded gastric tissues, including 15 cases of signet-ring cell carcinoma, 15 of intestinal-type adenocarcinoma, and 15 of normal gastric mucosa. Northern blotting was used to analyze the differences in GCRG123 expression between stomach signet-ring cell carcinoma and intestinal-type adenocarcinoma tissues. Online software, including BLAST, Multalin and BLAT, were applied for bioinformatics analysis. National Center for Biotechnology Information （NCBI） and the University of California Santa Cruz （UCSC） databases were used for the analyses.RESULTS： The in situ hybridization signal appeared as blue precipitates restricted to the cytoplasm. Ten out of 15 cases of gastric signet ring cell carcinoma, normal gastric mucosal epithelium and pyloric glands showed high GCRG123 expression. Low GCRG123 expression was observed in gastric intestinal-type adenocarcinoma and normal gastric glands. Northern blotting revealed that GCRG123 was up-regulated in signet-ring cell carcinoma tissue but down-regulated in intestinal-type adenocarcinoma tissue. BLAST and Multalin analyses revealed that the GCRG123 sequence had 92% similarity with the ORF2 sequence of human long interspersed nuclear element retrotransposons （LINE-1, L1）. BLAT analysis indicated that GCRG123 mapped to all chromosomes. GCRG123 was found to integrate in the intron-17 and -23 of Rb, 5＇ flanking region of IL-2 and clotting factor Ⅸ genes.CONCLUSION： GCRG123, an active member of the Lt family, was up-regulated in human gastric signet-ring cell carcinoma.

A novel gene,GCRG224,is differentially expressed in human gastric mucosa

AIM: To clone genes that may predispose us to humangastric cancer and to analyze it's expression in gastric ***: Specimens of paired tumor, paratumor andnormal gastric mucosa tissues collected from fifteen patientswho suffered from stomach antrum adenocarcinoma wereused for analysis. Seven out of the fifteen cases were firststudied by fluorescent differential display reversetranscription polymerase chain reaction (DDTR-PCR) *** differentially expressed bands of interest were cloned,analyzed by Northern blot, sequencing and RT-PCR. ThroughBLAST, the sequencing results were compared with GenBankdatabase for homology analysis. In situ hybridization withdiG-labeled cRNA probes was used to analyze the expressionof interesting cDNA bands in paraffin embedded pairednormal gastric mucosa and cancer tissues isolated from 30gastric adenocarcinoma ***: DDRT-PCR showed that one of the interestingcDNA bands, which was named W2, expressed much higherin all seven tested tumor and paratumor samples than intheir normal counterparts, it was sub-cloned into a pGEM-TEasy vector. Two subclones were subsequently *** of the subclone, GCRG224, was studied further. Thesequencing result showed that GCRG224 consisted of 1 159base pairs and had one open reading frame (ORF). It locatedat human chromosome 11q14. No homologue was found inGenBank database with GCRG224-ORF. This nucleotidesequence data were submitted to GenBank with accessionNo. AF438406. RT-PCR showed that GCRG224 expressedhigher in 11/15 gastric cancer tissues than in non-tumortissues. However, the result of Northern blot analysis showeda higher GCRG224 expression in the non-tumor tissue thanin the tumor one. Human multiple tissue Northern blot analysisrevealed that GCRG224 also expressed in human normal colontissue, and peripheral blood leukocyte. A Multi-UCAV cooperative occupation method based on weapon engagement zones for beyond-visual-range air combat

Recent advances in on-board radar and missile capabilities,combined with individual payload limitations,have led to increased interest in the use of unmanned combat aerial vehicles(UCAVs)for cooperative occupation during beyond-visual-range(BVR)air ***,prior research on occupational decision-making in BVR air combat has mostly been limited to one-on-one *** such,this study presents a practical cooperative occupation decision-making methodology for use with multiple *** weapon engagement zone(WEZ)and combat geometry were first used to develop an advantage function for situational assessment of one-on-one *** encircling advantage function was then designed to represent the cooperation of UCAVs,thereby establishing a cooperative occupation *** corresponding objective function was derived from the one-on-one engagement advantage function and the encircling advantage *** resulting model exhibited similarities to a mixed-integer nonlinear programming(MINLP)*** such,an improved discrete particle swarm optimization(DPSO)algorithm was used to identify a *** occupation process was then converted into a formation switching task as part of the cooperative occupation model.A series of simulations were conducted to verify occupational solutions in varying situations,including two-on-two *** results showed these solutions varied with initial conditions and weighting *** occupation process,based on formation switching,effectively demonstrates the viability of the proposed *** cooperative occupation results could provide a theoretical framework for subsequent research in cooperative BVR air combat.

Effects of different types of slow-and controlled-release fertilizers on rice yield

This experiment explored the effects of single application of seven types of slow-and controlled-release fertilizers on rice yield and various population *** on a study of the nitrogen(N)release characteristics of these fertilizers,pot experiments were conducted in 2018 and 2019 with split fertilization(CK,urea applied split equally at basal and panicle initiation stages,respectively)as control,which assessed the effects on SPAD value,yield and yield components,dynamic changes of rice tillers and dry matter *** results showed that the N release characteristics of different types of slow-and controlled-release fertilizers were significantly ***-coated urea(PCU)showed a controlledrelease mode and provided sustained release throughout the whole growth ***-coated urea(SCU)exhibited a slow-release mode,providing insufficient release at the middle and late *** inhibitor urea(AHA)and ureaformaldehyde(UF)yielded a rapid-release mode,with an explosive N release at the early stage and no release at the middle and late *** results showed that PCU delayed the peak seedling *** with CK,dry matter accumulation and SPAD showed no significant differences,and due to the continuous release of N throughout the growth stages,rice yield,spikelets per panicle,seed setting rate,and 1000-grain weight were all *** to the lack of N supply at the late stage and the low number of spikelets,SCU led to a reduction of rice yield,which is nevertheless not statistically *** and UF were susceptible to environmental factors and had varying effects on rice *** results of this experiment indicated that given a fixed amount of N applied in a pot,the stronger the N supply capacity and the longer the effective duration time of the fertilizer,the higher the dry matter accumulation at the late growth stage,and the higher the rice yield.

High mobility group-box 3 overexpression is associated with poor prognosis of resected gastric adenocarcinoma

I_κB kinase-beta inhibitor attenuates hepatic fibrosis in mice

AIM: To investigate the anti-fibrosis effect of IκB kinase-beta inhibitor (IKK2 inhibitor IMD0354) in liver fibrosis. METHODS: Twenty male C57BL6 mice were divided into four groups. Five high-fat fed mice were injected with lipopolysaccharide (LPS, 10 mg/kg) intraperitoneally and five high-fat fed mice were without LPS injection to build models of liver injury, and the intervention group (five mice) was injected intraperitoneally with IKK2 inhibitor (IMD 30 mg/kg for 14 d), while the remaining five mice received a normal diet as controls. Hepatic function, pathological evaluation and liver interleukin-6 (IL-6) expression were examined. Western blotting and real-time polymerase chain reaction were used to detect the expressions of nuclear factor-κB (NF-κB), alpha-smooth muscle actin (α-SMA), tumor growth factor-beta1 (TGF-β1), tumor necrosis factor-alpha (TNF-α), typeⅠand type Ⅲ collagen proteins and mRNA. RESULTS: A mouse model of liver injury was successfully established, and IMD decreased nuclear transloca-tion of NF-κB p65 in liver cells.
