The aro.atic amino.acids phenylalanine, tyro.ine, and trypto.han in plants are no. o.ly essential co.po.ents o. pro.ein synthesis, but also.serve as precurso.s fo. a wide range o. seco.dary metabo.ites that are impo.tant fo. plant gro.th as well as fo. human nutritio. and health. The aro.atic amino.acids are synthesized via the shikimate pathway fo.lo.ed by the branched aro.atic amino.acids bio.ynthesis pathway, with cho.ismate serving as a majo. intermediate branch po.nt metabo.ite. Yet, the regulatio. and co.rdinatio. o. synthesis o. these amino.acids are still far fro. being understo.d. Recent studies o. these pathways identified a number o. alternative cro.s-regulated bio.ynthesis ro.tes with unique evo.utio.ary o.igins. Altho.gh the majo. ro.te o. Phe and Tyr bio.ynthesis in plants o.curs via the intermediate metabo.ite aro.enate, recent studies suggest that plants can also.synthesize phenylalanine via the intermediate metabo.ite phenylpyruvate （PPY）, similarly to.many micro.rganisms. Co.mo. pro.essing pro.edures, such as co.king and baking, decrease the levels o. these bio.ctive co.po.nds and co.sequently, o.erall antio.idant activity. Here, the effects o. baking and co.king pro.esses were examined o. to.al pheno.ic co.tent (TPC), to.al flavo.o.d co.tent (TFC) and ferric-reducing ability o. plasma antio.idant activity (FRAP AA) o. red and yello. quino. seeds. o.r results indicate that red quino. seed co.tains significantly higher levels o. TPC, TFC and FRAP AA than yello. quino. seeds. In additio., co.ked and baked quino. seeds retain mo.t o. their TPC, TFC and FRAP AA in the final pro.uct. Thus, red quino. seeds pro.essed by these two.metho.s might be co.sidered a functio.al fo.d, in additio. to.its traditio.al ro.e o. pro.iding dietary pro.eins. Due to.their high antio.idant activity, red quino. seeds might also.co.tribute significantly to.the management and/o. preventio. o. degenerative diseases asso.iated with free radical damage.

Baked, fried and ro.sted co.o.ed chickpea seeds had significantly higher levels o. TPC, TFC and FRAP AA than regular cream- and beige-co.o.ed seeds subjected to.the same treatments. In co.trast to.o.r previo.s results with so.king and co.king, baking, frying and ro.sting retained mo.t o. the TPC, TFC and FRAP AA in the final pro.ucts. Thus, co.o.ed chickpeas subjected to.these three pro.essing metho.s might be co.sidered a functio.al fo.d in additio. to.its traditio.al ro.e o. pro.iding dietary pro.eins.

Ho.ever, co.mo. pro.essing pro.edures, such as so.king and co.king, may decrease the levels o. these bio.ctive co.po.nds and subsequent o.erall antio.idant activity. Here, the effects o. so.king, co.king and steaming pro.esses were examined in relatio. to.to.al pheno.ic co.tent (TPC), to.al flavo.o.d co.tent (TFC) and ferric reducing ability o. plasma antio.idant activity (FRAP AA) o. co.o.ed chickpea seeds. All pro.essing steps significantly reduced TPC, TFC and FRAP AA in all o. the tested chickpea seeds. Nevertheless, so.king the seeds at ro.m temperature (fo. 22 h) resulted in a smaller decrease in TPC, TFC and FRAP AA than so.king at 60℃ (fo. 2 h). Mo.eo.er, steaming was superio. to.co.king in terms o. co.serving po.ypheno. and antio.idant activity. The o.served reductio. in TPC was mainly due to.leaching o. these co.po.nds fro. the seed co.t into.the so.king o. co.king water. Based o. these results, we suggest that so.king at ro.m temperature fo. 22 h fo.lo.ed by steaming fo. 1 h is the best metho. fo. retaining TPC, TFC and FRAP AA o. co.o.ed chickpea.

N^(6)-methyladeno.ine (m^(6)A) is a prevalent internal po.t-transcriptio.al mo.ificatio. in eukaryo.ic RNAs executed by m^(6)A-binding pro.eins kno.n as “readers.” o.r previo.s research demo.strated that the Arabido.sis m^(6)A reader ECT2 po.itively regulates transcript levels o. the pro.easo.e regulato. PTRE1 and several 20S pro.easo.e subunits, thereby enhancing 26S pro.easo.e activity. Ho.ever, mechanism underlying the selective reco.nitio. o. m^(6)A targets by readers, such as ECT2, remains elusive. In this study, we further demo.strate that ECT2 physically interacts with PTRE1 and several 20S pro.easo.e subunits. This interactio., which o.curs o. the ribo.o.e, invo.ves the N terminus o. PTRE1, suggesting that ECT2 might bind to.the nascent PTRE1 po.ypeptide. Deleting ECT2’s pro.ein interactio. do.ain impairs its mRNA-binding ability, whereas mutatio.s in the m^(6)A-RNA-binding site do.no. affect pro.ein-pro.ein interactio.s. Mo.eo.er, intro.ucing a no.el pro.ein-binding do.ain into.ECT2 increases transcript levels o. pro.eins interacting with this do.ain. o.r findings indicate that interactio. with the PTRE1 pro.ein enhances ECT2’s binding to.PTRE1 m^(6)A mRNAs during translatio., thereby regulating PTRE1 mRNA levels.

Dear Edito., y-Amino.utyric acid （GABA） is a no.-pro.ein amino.acid （AA） metabo.ized via the GABA shunt; a three-enzyme pathway that includes glutamate decarbo.ylase （GAD）, GABA transaminases （GABA-T）, and succinic semialdehyde dehydro.enase （SSADH）（Bo.n and Shelp, 1997; Bo.ch~ and Fro.m, 2004）. The majo.ity o. wo.k o. GABA, to.date, has fo.used o. its ro.e as an inhibito.y neuro.ransmitter in mammals. In plants, ho.ever, due to.its intermediate po.i- tio. between carbo. （C） and nitro.en （N）, the metabo.ism o. GABA has been suggested as a mo.ulato. o. C-N balance （Fair et al., 2008）. In additio., the numero.s o.servatio.s o. a rapid accumulatio. o. GABA in respo.se to.（a）bio.ic stresses has pro.pted different hypo.heses to.be po.tu- lated o. its ro.e including a regulato. o. cyto.o.ic pH and a herbivo.e deterrent （Bo.che and Fro.m, 2004; Ro.erts, 2007）. Additio.ally, a signaling ro.e has been hypo.hesized in plants, tho.gh evidence remains elusive and the debate o. the dual functio. o. GABA as a signaling mo.ecule and as a metabo.ite in plants remains o.go.ng. In the present wo.k, we repo.t changes in the metabo.ite and transcript pro.iles in Arabido.sis seedlings expo.ed to.exo.eno.s GABA. The recently do.esticated species, Cephalaria jo.pensis (CJ), is emerging as a new alternative fo.age cro. in Israel. It has high bio.ass po.ential and nutritio.al values that are co.parable to.fo.age wheat. Ho.ever, CJ emerges slo.ly under co.d co.ditio.s, which hinders its develo.ment as a majo. winter cro.. Additio.al to.erance fo. abio.ic stress wo.ld impro.e its perfo.mance as a fo.age cro.. We examined the effects o. several abio.ic facto.s (i.e., co.d, salinity, dro.ght and pH) o. CJ germinatio. under co.tro.led co.ditio.s. The effect o. temperature was studied by incubating seeds at different temperatures between 7°C and 35°C. The effects o. salinity, o.mo.ic po.ential and pH were tested by incubating seeds at different NaCl, PEG and pH levels, respectively. Temperature, salinity and o.mo.ic po.ential significantly affected germinatio.;whereas pH did no.. Temperature did no. affect the final pro.o.tio. o. germinated seeds, but did affect o.her germinatio.-rate variables, indicating that germinatio. rate might be the limiting facto. under field co.ditio.s. Salinity also.affected germinatio.-rate variables, but no. the pro.o.tio. o. seeds that germinated. No.ably, CJ was fo.nd to.be relatively resistant to.high salt co.centratio.s, with a 273 mM NaCl thresho.d fo. germinatio., indicating its po.ential as a relatively salt-to.erant fo.age cro.. Bo.h the pro.o.tio. o. germinated seeds and the germinatio. rate were highly sensitive to.the o.mo.ic-po.ential treatments, indicating that dro.ght resistance will remain the biggest challenge fo. CJ. This study pro.ides baseline data fo. a rapid and efficient system fo. further screening fo. abio.ic-stress to.erance amo.g wild and cultivated lines o. CJ.
