Microbiota modification by probiotic supplementation reduces colitis associated colon cancer in mice

AIM To investigate the effect of probiotic supplementation during the development of an experimental model of colitis associated colon cancer(CAC). METHODS C57 BL/6 mice received an intraperitoneal injection of azoxymethane(10 mg/kg), followed by three cycles of sodium dextran sulphate diluted in water(5% w/v). Probiotic group received daily a mixture of Lactobacillus acidophilus, Lactobacil us rhamnosus and Bifidobacterium bifidum. Microbiota composition was assessed by 16 Sr RNA Illumina Hi Seq sequencing. Colon samples were collected for histological analysis. Tumor cytokines was assessed by Real Time-PCR(Polymerase Chain Reaction); and serum cytokines by Multiplex assay. All tests were two-sided. The level of significance was set at P graphs were generated and statistical analysis performed using the software graph Pad Prism 5.0. The project was approved by the institutional review board committee. RESULTS At day 60 after azoxymethane injection, the mean number of tumours in the probiotic group was 40% lower than that in the control group, and the probiotic group exhibited tumours of smaller size(groups. However, there was a significant difference in beta diversity in the multidimensional scaling analysis. The abundance of the genera Lactobacillus, Bifidobacterium, Allobaculum, Clostridium XI and Clostridium XVⅢ increased in the probiotic group(P ge was accompanied by reduced colitis, demonstrated by a 46% reduction in the colon inflammatory index; reduced expression of the serum chemokines RANTES and Eotaxin; decreased p-IKK and TNF-α and increased IL-10 expression in the colon. CONCLUSION Our results suggest a potential chemopreventive effect of probiotic on CAC. Probiotic supplementation changes microbiota structure and regulates the inflammatory response, reducing colitis and preventing CAC.

Objective: To investigate the effects of perorally administered juice on tetrachloromethane(CCl4)-induced hepatotoxicity model in rats. Methods: Male Wistar rats were tubeadministrated silymarin, Ecballium juice at 0.2 mL/kg and 0.7 mL/kg daily for 3 consequent days, ie. 3.28 μg and 11.48 μg of cucurbitacin B per kg of body weight respectively. On the third day, liver damage was induced by intraperitoneal application of CCl4. On the fourth day, abdominal cavity was macroscopically examined and liver samples were taken for histopathological and immunochemical evaluation. HPLC was used to determine the content of the active substance cucurbitacin B. Results: The experiment revealed that 0.7 ml/kg juice concentration expressed the highest pro-apoptotic activity, but with prevailing negative effects. Compared with the lower concentration, there was an observable vasodilatation with consequent interstitial hemorrhages and a larger scope of inflammatory damage, which suppressed the hepatoprotective effect. In the 0.2 mL/kg concentration, there was a smaller pro-apoptotic activity but other parameters had better results, and the liver parenchyma damage was reversible. Conclusions: No reactions confirming the potentially allergic effect on laboratory rats were observed; its hepatoprotective and anti-inflammatory effect was confirmed on a model of acute liver damage.

Factoring quadratics over Z is a staple of introductory algebra and textbooks tend to create the impression that doable factorizations are fairly common. To the contrary, if coefficients of a general quadratic are selected randomly without restriction, the probability that a factorization exists is zero. We achieve a specific quantification of the probability of factoring quadratics by taking a new approach that considers the absolute size of coefficients to be a parameter n. This restriction allows us to make relative likelihood estimates based on finite sample spaces. Our probability estimates are then conditioned on the size parameter n and the behavior of the conditional estimates may be studied as the parameter is varied. Specifically, we enumerate how many formal factored expressions could possibly correspond to a quadratic for a given size parameter. The conditional probability of factorization as a function of n is just the ratio of this enumeration to the total number of possible quadratics consistent with n. This approach is patterned after the well-known case where factorizations are carried out over a finite field. We review the finite field method as background for our method of dealing with Z [x]. The monic case is developed independently of the general case because it is simpler and the resulting probability estimating formula is more accurate. We conclude with a comparison of our theoretical probability estimates with exact data generated by a computer search for factorable quadratics corresponding to various parameter values.

This viral outbreak of the novel SARS-CoV-2 has profoundly disrupted lives globally and has placed unparalleled burdens on individual physical and psychological *** is imperative to comprehend the psychological journey of COVID-19 patients to provide them with effective *** study aimed to explore the psychological illness experience of severe COVID-19 survivors from the first symptoms,to hospitalization,recovery,and post-discharge ***-structured interviews were conducted from November 2022 to April 2023,with 30 Romanian former COVID-19 *** analysis of transcribed interviews identified four major themes:(1)a distant threat;(2)the tipping point;(3)the turning point;and(4)readjustment and *** participants faced denial or uncertainty early on,misattributing symptoms and delaying seeking *** as a last resort brought anxiety,isolation,and closer confrontations with mortality *** symptoms improved,and participants received supportive care from both healthcare professionals and family members,hope and optimism *** health improvements and feelings of safety,lingering anxiety ***-discharge adjustment involved managing lasting effects and selectively resuming *** some of the participants,reflection afterward prompted greater self-understanding,empathy and connection,and motivation for *** results have implications for healthcare practitioners,policymakers,and researchers.

Image sequences processing and video encoding are extremely time consuming problems. The time complexity of them depends on image contents. This paper presents an estimation of a block motion method for video coding with edge alignment. This method uses blocks of size 4 × 4 and its basic idea is to find motion vector using the edge position in each video coding block. The method finds the motion vectors more accurately and faster than any known classical method that calculates all the possibilities. Our presented algorithm is compared with known classical algorithms using the evaluation function of the peak signal-to-noise ratio. For comparison of the methods we are using parameters such as time, CPU usage, and size of compressed data. The comparison is made on benchmark data in color format YUV. Results of our proposed method are comparable and in some cases better than results of standard classical algorithms.

Survivin activity in normal human osteoblasts and osteosarcoma cells

PURPOSE: This study was designed to investigate and compare in vitro survivin activity in normal human osteoblast and Mg-63 osteosarcoma cell cultures with and without vitamin D3. METHODS: Normal human alveolar bone explants were recovered from extraction sites of non-carious teeth of 9 healthy donors and cultured to the 2nd passage. Mg-63 osteosarcoma cells were obtained from ATCC. To compare the survivin activities in these two types of cells and to determine the effect of vitamin D3 on survivin expression and associated activities of cell proliferation and differentiation, levels of survivin, osteocalcin and alkaline phosphatase were measured at 7-20 day cultures with and without vitamin D3 in both cultures of normal osteoblasts and osteosarcoma cells. Matched pair t-test and two sample independent t-test were applied for statistical analysis of the data. P values of 0.05 or less were considered statistically significant. RESULTS: A significantly higher level of survivin was detected in normal osteoblasts compared to osteosarcoma cells at 7 days without vitamin D3 treatment (Pgnificantly decreased after vitamin D3 treatment at 7 days (Pgher expression of survivin and less differentiated osteoblasts that still retain their proliferative ability. Vitamin D3 has significant negative effect on expression of survivin in normal human osteoblasts but not on that in osteosarcoma cells.

In the course, FTV 201A： MEDIA INDUSTRIES and CULTURES of PRODUCTION： FOUNDATIONS, we discussed how to think the philosophy and thoughts that surround people who produce and work in the culture of production and media. That way, the purpose of this work is to think the use of post-horror label as a marketing strategy in the context of Brazilian contemporary literature and cinema. For that, I map the profile of three Brazilian writers： Santiago Nazarian, Antonio Xerxenesky, and Raphael Montes. They are published now by Companhia das Letras, the largest publisher in Brazil, which has an extensive work with the horror literature, having the exclusivity of Stephen King’s books. In addition, all three circulate differently in the media space, standing between an acceptance of academic criticism and the prestige of large-scale consumer audiences. I analyzed what are the writer strategies to construct their image and establish themselves in the literary market using the Post-horror label and its relation with the production company, RT Features, which is financing them. In the end, I did a critical analyzes of the post-horror label as a transmedia strategy to construct an author’s name.

The present study is intended to be an analysis of the main Romanian legal provision in what concerns the impact of foreign exchange differences and potential consequences at the level of financial statements for companies. In Romanian legislation, the issue of the foreign exchange differences is treated at a general level： these are recognized as a profit and loss item and as a consequence, they are included in the taxable base when booked in the accounting. Our legislation does not provide for specific treatments depending on the ＇realization momenf＇ of these differences or depending on the conditions for recognizing a gain/loss from foreign exchange differences. For the unrealized exchange differences arising from the application of accounting rules on monthly assessment of foreign currency monetary items, there is not a specific event or transaction to determine income or expenditure. This monthly review depending on the National Bank of Romania [BNR] foreign exchange rate valid on the last day of the month aims to bring closer to reality the financial position of an entity. The exchange rate is a monetary policy item set by the central bank and may be influenced by various factors such as： monetary policy of the BNR; inflation target objectives and rate of exchange stabilization or reduction efforts, as is the case of BNR in the last years to fulfill EU criteria for adoption of the EURO; periodical influences of speculative capitals on the exchange rate level; economic status and especially exports and imports. However, a taxpayer should not be charged unless there is evidence of the ＂economic enrichment＂ thereof. However, this enrichment, seen as a rising economic value of the company, depends on the perspective from which is seen： the owner of the company or the tax authorities. The exchange differences impact also the value of the owners＇ equity for which a minimum level is requested under the commercial company law. In case of negative values, this triggers
