Self-reported participation restrictions in normal hearing individuals in thyroid ontogeny: Evidence of subclinical changes

Purpose: To verify evidence of subclinical alterations through self-reported participation restrictions in normal hearing individuals and congenital hypothyroidism patients.Methods: An analytical, quantitative, cross-sectional exploratory study with a 1:1 ratio, consisting of a convenience sample of 86 normal hearing individuals with(n = 42) and without(n = 44) congenital hypothyroidism diagnosis. All participants underwent tonal and speech audiometry, immitance and distortion product otoacoustic emissions. The researchers excluded people with hearing loss, genetic syndromes and metabolic diseases. The instrument used for evaluate of self-reported participation restrictions was the HHIE-adapted questionnaire, composed of 25 questions, 12 of which were social domain and 13 emotional domain. Student's ttest and chi-square test were used for statistical analysis at a significance level of 5%.Results: There was a significant(p < 0.001) self-reported participation restrictions in CH(61.9%), with a greater relevance for the social domain(p = 0.002). There was a greater frequency of mild/moderate(40.5%)and higher prevalence of association with clinical factors and adherence to treatment.Conclusion: The findings indicate that self-reported participation restrictions in normal hearing individuals with congenital hypothyroidism was more significant than in the non-exposed group, suggesting evidence of subclinical auditory abnormalities in this population.

The production of plastics has exponentially increased over time such that the observation of microplastics in the oceans has become a major concern given how frequently these particles and marine biota may interact.Microplastics can be ingested by diverse species,including invertebrates,which can lead to physical damage due to their small size and potentially chemical damage related to the ingestion of monomers,polymeric additives,or adsorbed chemicals.This study reports on the first evidence of microplastics in Mnemiopsis leidyi.Microplastics were described by type,quantified,and had their chemical composition identified.In total,328 samples of microplastic(MP)candidates were observed:blue fibers were the most abundant,followed by black,red,and green fibers as well as blue fragments.Three types of microplastics were chemically identified as polyurethane,unplasticized polyvinylchloride,and polyethylene polylauryllactam.Overall,this study demonstrated that Mnemiopsis leidyi have the capacity to incorporate microplastics.Although more studies are needed,Mnemiopsis leidyi is relatively abundant on the Atlantic coast and should be recommended for use as a bioindicator for microplastics.

Detection and determination of undeclared synthetic caffeine in weight loss formulations using HPLC-DAD and UHPLC-MS/MS

Caffeine is present in products marketed for weight loss, with the purpose of increasing thermogenesis and lipid metabolism. The dosage declared by the product manufacturer, or even its presence, is not always correctly described on the label. This work aimed to investigate the undeclared synthetic caffeine in weight loss formulations by a high-performance liquid chromatography with diode array detection(HPLC-DAD) method. From one hundred products purchased through Brazilian e-commerce, seventeen contained caffeine, either naturally or synthetically added to formulation. The caffeine-containing samples were confirmed by an ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS) method, and adulteration was clearly proven in five products. The content highest caffeine contained 448.8 mg per dose. Other irregularities were also found; nevertheless, the most serious was the addition of synthetic drugs without asking the consumers. Additional drugs expose the consumer to more possible side-effects as well as deleterious drug interactions. Intentional adulteration with any unlabeled substance is typically motivated by a desire to increase or alter the claimed effect of the marketed product to gain a commercial advantage.

Formation and Evolution of Wakes in the Spacetime Generated by a Cosmic String in f(R) Theory of Gravity

The formation and evolution of cosmic string wakes in the framework of a f(R) theory of gravity are investigated in this work. We consider a simple model in which baryonic matter flows past a cosmic string. We treat this problem in the Zel’dovich approximation. We compare our results with previous results obtained in the context of General Relativity and Scalar-Theories of Gravity.

Leukocytoclastic vasculitis caused by hepatitis C virus in a liver transplant recipient: A case report

BACKGROUND Infection by the hepatitis C virus(HCV) is currently considered to be a global health issue, with a high worldwide prevalence and causing chronic disease in afflicted individuals. The disease largely involves the liver but it can affect other organs, including the skin. While leukocytoclastic vasculitis has been reported as one of the dermatologic manifestations of HCV infection, there are no reports of this condition as the first symptom of HCV recurrence after liver transplantation.CASE SUMMARY We report here a case of leukocytoclastic vasculitis in a liver transplant recipient on maintenance immunosuppression. The condition presented as a palpable purpura in both lower extremities. Blood and urine cultures were negative and all biochemical tests were normal, excepting evidence of anemia and hypocomplementemia. Imaging examination by computed tomography showed a small volume of ascites, diffuse thickening of bowel walls, and a small bilateral pleural effusion. Skin biopsy showed leukocytoclasia and fibrinoid necrosis.Liver biopsy was suggestive of HCV recurrence in the graft, and HCV polymerase chain reaction yielded 11460 copies/mL and identified the genotype as 1 A. Treatment of the virus with a 12-wk direct-acting antiviral regimen of ribavirin, sofosbuvir and daclatasvir led to regression of the symptoms within the first 10 d and subsequent complete resolution of the symptoms.CONCLUSION This case highlights the difficulties of diagnosing skin lesions caused by HCVinfection in immunosuppressed patients.

Human CD4 ＋ effector T lymphocytes generated upon TCR engagement with self-peptides respond defectively to IL-7 in their transition to memory cells

The peripheral repertoire of CD4＋ T lymphocytes contains autoreactive cells that remain tolerant through several mechanisms. However, nonspecific CD4＋ T cells can be activated in physiological conditions as in the course of an ongoing immune response, and their outcome is not yet fully understood. Here, we investigate the fate of human naive CD4＋ lymphocytes activated by dendritic cells （DCs） presenting endogenous self-peptides in comparison with lymphocytes involved in alloresponses. We generated memory cells （Tmem） from primary effectors activated with mature autologous DCs plus interleukin （IL）-2 （Tmauto）, simulating the circumstances of an active immune response, or allogeneic DCs （Tmallo）. Tmem were generated from effector cells that were rested in the absence of antigenic stimuli, with or without IL-7. Tmem were less activated than effectors （demonstrated by CD25 downregulation） particularly with IL-7, suggesting that this cytokine may favour the transition to quiescence. Tmauto and TmaHo showed an effector memory phenotype, and responded similarly to polyclonal and antigen-specific stimuli. Biochemically, IL-7-treated Tma^o were closely related to conventional memory lymphocytes based on Erk-l/2 activation, whereas Tmauto were more similar to effectors. Autologous effectors exhibited lower responses to IL-7 than allogeneic cells, which were reflected in their reduced proliferation and higher cell death. This was not related to IL-7 receptor expression but rather to signalling deficiencies, according to STAT5 activation These results suggest that ineffective responses to IL-7 could impair the transition to memory cells of naive CD4＋ T lymphocytes recognizing self-peptides in the setting of strong costimulation.

Cytomegalovirus is the most frequent agent of congenital viral infections,affecting approximately 0.2%-2.4%of live births in different countries[1-7].Universal screening for congenital cytomegalovirus infection in newborns is not a standard practice considering cost-benefit issues and viabil-ity of diagnostic tests.Besides,most newborns are asympto-matic and there is no effective treatment for this population[2-5].

Schistosoma mansoni: Phytochemical Effect on Aquatic Life Cycle

Background: Two aspects need to be considered for schistosomiasis control: morbidity and transmission. In this context, many soluble substances have been tested and Euphorbia milii latex is one of the most promising Brazilian molluscicides. Phytochemical studies involving simulation of the applicability of this latex on all aquatic forms of the S. mansoni life cycle are rare in the literature. The aim of this work was to evaluate the effect of E. milii latex on S. mansoni in the egg, miracidium and different developmental stages in Biomphalaria glabrata. Methods: The laboratory study was designed to simulate the different forms of exposure of the life cycle stages of S. mansoni to the LC50 of E. milii latex;we tested the exposition from four situations of S. mansoni contact with the latex and observed the exposure on different snails’ infection stage too. All snails were analyzed weekly for cercarial shedding and reproductive biology. Results: The results showed that contact of S. mansoni eggs and miracidia with the LC50 of E. milii negatively influenced the development of the parasite life cycle in the intermediate host, with consequent reduction of cercarial shedding. The exposure of infected snails affected the reproductive biology and cercarial shedding in all intra-mollusk development stages of S. mansoni, but the reduction was greater in the first, fourth, fifth, sixth, seventh and eighth weeks of infection. The LC50 of E. milii latex had toxic action on eggs and miracidia, and the number of cercariae shed by snails during the study period declined by about 80%. Conclusions: We can conclude that the use of natural biodegradable compounds containing low concentrations of substances already characterized as having eco-toxico- logical potential can be an important tool to reduce the transmission of Schistosomiasis.

Sward Structural Characteristics and Performance of Beef Heifers Reared under Rotational Grazing Management on Campos Grassland

This study aimed to evaluate the effect of two rest intervals on structural sward characteristics and productive performance of beef heifers reared on Campos grassland managed on rotational grazing. The treatments were two intervals between grazing of 375 and 750 DD (degree days), based on thermal cumulative sum for leaf expansion of native grasses belonging to two functional groups. The experimental design was a randomized complete block design with two treatments and three replications. The tested animals were beef heifers with initial age of 12 months and average weight of 185.2 ± 17.4 kg. Measures in the pasture were: herbage mass, mass of green leaf blades, stem mass, dead material mass and green leaf allowance. The 750 DD rest interval presented higher herbage mass (24%), higher green herbage mass and 19% more leaves in the canopy. This rest interval also presents a high proportion of dead material and stems in the sward structure. In the other way, the 375 DD rest interval presents better chemical characteristics, with 20% more crude protein in the hand plucking samples. Heifers’ dry matter intake was similar between the rest intervals (2.04% of live weight) and the bite mass was also similar (0.22 g DM per bite) but the animal performance was higher in the 375 DD rest interval. The stocking density showed similarity between treatments with an average of 875 kg/LW/ha. The live weight gain per area was higher in 40 kg/LW/ha (P = 0.117) for 375 DD rest interval, reaching 251 kg/LW/ha produced over the 149 experimental days. Based on these results, we can conclude that the treatment of shortest interval between defoliation gave the highest gain individual animals in warm seasons.
