Petiveria alliacea Suppresses Airway Inflammation and Allergen-Specific Th2 Responses in Ovalbumin-Sensitized Murine Model of Asthma

Objective： To examine the effect of metanol extract of Petiveria alliacea （PM） on airway inflammation in a murine model of chronic asthma. Methods： Two-month-old male BALB/c mice （n=6-8/group） were sensitized on days 0 and 14 by intraperitoneal injection of 20μg ovalbumin （OVA）. On day 25, the mice received an airway challenge with OVA （3%, w/v, in phosphate buffered saline）. PM was administered orally by oral gavage to mice at doses of 100, 200 and 400 mg/kg body weight once daily from days 18 to 23. Control mice were orally administered phosphate buffered saline （PBS） to induce a model of asthma. At the end of the test, respiratory reactivity was assayed, the total cell number, interleukin-4 （IL-4）, IL-5, IL-13, tumor necrosis factor-alpha （TnF-α） and reactive oxygen species （ROS） in the bronchoalveolar lavage fluid （BALF） were determined and the levels of serum IgE, intercellular cell adhesion molecule 1 （ICAM-1） and eotoxin were measured. In addition, lung tissue was used to qualify the IL-4, IL-5, IL-13, TnF-α and transforming growth factor beta 1 （TGF-β1）. Histologic examination was performed to observe inflammatory cellular infiltration. Results： The administration of PM in comparison with the OVA-only treated group significantly attenuated the infiltration of eosinophUs and other inflammatory cells （P〈0.01）. Airway resistance （RI） in the OVA-only induced group was significantly higher than that of the PBS control group （P〈0.01） when methacholine was added. TnF-α, IgE, TGF-β1 and cytokine levels IL-4, IL-5, IL-β in the BALF decreased compared to control mice （P〈0.01 or P〈0.05）. PM treatment also inhibited the production of chemokines, eotaxin and ICAM-1 in BALF （P〈0.01）, which improved lung function. Histopathological examination revealed that the sensitized treated PM groups had significant lower in inflammatory scores similar to dexamethasone treatments and the untreated group. Difficult airway situations are a major cause of morbidity, mortality and malpractice claims in anesthetic practice and other critical care settings. Although infrequent, they are serious and complex situations where many variables interact. Among these variables, quick and accurate rescuer decision-making is a major outcome determinant. Emerging evidence, recent advances in technology and development of new devices have provided new tools to deal with the problem, but still, basic underlying principles of management remain. Based on the available evidence and expert consensus, multiple guidelines and algorithms have been published, but they may have limited applicability, contain confusing and ambiguous recommendations or address very specific clinical situations. This narrative review attempts to summarize, organize and consolidate current trends and supported recommendations of the updated guidelines in one practical, concise and unified guide, proposing a simple, intuitive and novel algorithm.

AIM To evaluate the effects of asymmetric dimethylarginine(ADMA) in renal arteries from portal hypertensive and cirrhotic *** Rat renal arteries from Sham(n = 15), pre-hepatic portal hypertension(PPVL; n = 15) and bile duct ligation and excision-induced cirrhosis(BDL; n = 15) were precontracted with norepinephrine, and additional contractions were induced with ADMA(10-6-10-3 mol/L), an endogenous inhibitor of nitric oxide(nO) synthase. Concentration-response curves to acetylcholine(1 × 10-9^(-3) × 10^(-6) mol/L) were determined in precontractedrenal artery segments with norepinephrine in the absence and in the presence of ADMA. Kidneys were collected to determine the protein expression and activity of dimethylarginine dimethylaminohydrolase(DDAH), an enzyme that catabolizes ADMA. RESULTS In renal arteries precontracted with norepinephrine, ADMA caused endothelium-dependent contractions. The pD 2 values to ADMA were similar in the Sham and PPVL groups(4.20 ± 0.08 and 4.11 ± 0.09, P > 0.05, respectively), but were lower than those of the BDL group(4.79 ± 0.16, P ne-induced endothelium-dependent relaxation that did not differ, in terms of p D2 and maximal relaxation, among the 3 groups studied. Treatment with ADMA(3 × 10^(-4) mol/L) inhibited acetylcholine-induced relaxation in the 3 groups, but the inhibition was higher(P n the BDL group compared with that for the Sham and PPVL groups. The m RnA and protein expression of DDAH-1 were similar in kidneys from the three groups. Conversely, DDAH-2 expression was increased(P n PPVL and further enhanced(P n the BDL group. However, renal DDAH activity was significantly decreased in the BDL group. COnCLUSIOn Cirrhosis increased the inhibitory effect of ADMA on basal- and induced-release of nO in renal arteries, and decreased DDAH activity in the kidney.

Squeezing ground conditions,which can lead to severe loads in tunnels,have historically been associated with the presence of clay minerals in the *** the years,many methodologies have been proposed to predict squeezing in tunnels based on tunnel depth,in situ stress,ground mineralogy,and ground strength and deformation *** paper presents a comprehensive review of methodologies to predict tunnel squeezing in clay-rich rocks.A new methodology is proposed where ground conditions and squeezing potential are assessed based on the Stress History and normalized Soil Engineering Properties(SHAnSEP)approach adapted to clayrich rocks,Peck’s stability number and Hoek&Brown’s(1997)Geological Strength Index(GSI).A squeezing number S is suggested to classify ground conditions based on the level of squeezing that the ground may experience in response to ***,it is demonstrated that by combining the proposed classification system and an existing classification system for ground squeezing condition,an accurate estimate of tunnel strain can also be *** proposed method is applied to four case studies of tunnels in squeezing ground in shale and mudstone.

This paper proposes a prediction engine designed for non-dedicated clusters, which is able to estimate the turnaround time for parallel applications, even in the presence of serial workload of the workstation owner. The prediction engine can be configured to work with three different estimation kernels： a Historical kernel, a Simulation kernel based on analytical models and an integration of both, named Hybrid kernel. These estimation proposals were integrated into a scheduling system, named CISnE, which can be executed in an on-line or off-line mode. The accuracy of the proposed estimation methods was evaluated in relation to different job scheduling policies in a real and a simulated cluster environment. In both environments, we observed that the Hybrid system gives the best results because it combines the ability of a simulation engine to capture the dynamism of a non-dedicated environment together with the accuracy of the historical methods to estimate the application runtime considering the state of the resources.

Fracture systems have strong influence on the overall mechanical behavior of fractured rock masses dueto their relatively lower stiffness and shear strength than those of the rock matrix. Understanding theeffects of fracture geometrical distribution, such as length, spacing, persistence and orientation, isimportant for quantifying the mechanical behavior of fractured rock masses. The relation betweenfracture geometry and the mechanical characteristics of the fractured rock mass is complicated due tothe fact that the fracture geometry and mechanical behaviors of fractured rock mass are stronglydependent on the length scale. In this paper, a comprehensive study was conducted to determine theeffects of fracture distribution on the equivalent continuum elastic compliance of fractured rock massesover a wide range of fracture lengths. To account for the stochastic nature of fracture distributions, threedifferent simulation techniques involving Oda＇s elastic compliance tensor, Monte Carlo simulation （MCS）,and suitable probability density functions （PDFs） were employed to represent the elastic compliance offractured rock masses. To yield geologically realistic results, parameters for defining fracture distributionswere obtained from different geological fields. The influence of the key fracture parameters andtheir relations to the overall elastic behavior of the fractured rock mass were studied and discussed. Adetailed study was also carried out to investigate the validity of the use of a representative elementvolume （REV） in the equivalent continuum representation of fractured rock masses. A criterion was alsoproposed to determine the appropriate REV given the fracture distribution of the rock mass.

In the present work, the recent developed Unsteady Double Wake Model, USDWM, is used to simulate separated flows past a wind turbine airfoil at high angles of attack. The solver is basically an unsteady two-dimensional panel method which uses the unsteady double wake technique to model flow separation and its dynamics. In this paper, the calculated integral forces have been successfully validated against wind tunnel measurements for the FFA-W3-211 airfoil. Furthermore, the computed highly unsteady flow field is analyzed in detail for a set of angles of attack ranging from light to deep stall conditions.
