Recent evidences suggested that cyclic guanosine monophosphate-specific phosphodiesterase 5 （PDE5） inhibitor represents an important therapeutic target for cardiovascular diseases. Whether and how it ameliorates cardiac fibrosis, a major cause of diastolic dysfunction and heart failure, is unknown. The purpose of this study was to investigate the effects of PDE5 inhibitor on cardiac fibrosis. We assessed cardiac fibrosis and pathology in mice subjected to transverse aortic constriction （TAC）. Oral sildenafil, a PDE5 inhibitor, was administered in the therapy group. In control mice, 4 weeks of TAC induced significant cardiac dysfunction, cardiac fibrosis, and cardiac fibroblast activation （proliferation and transformation to myofibroblasts）. Sildenafil treatment markedly prevented TAC-induced cardiac dysfunction, cardiac fibrosis and cardiac fibroblast activation but did not block TAC-induced transforming growth factor-β0 （TGF-β1） production and phosphorylation of Smad2/3. In isolated cardiac fibroblasts, sildenafil blocked TGF-β1-induced cardiac fibroblast transformation, proliferation and collagen synthesis. Furthermore, we found that sildenafil induced phosphorylated cAMP response element binding protein （CREB） and reduced CREB-binding protein 1 （CBP1） recruitment to Smad transcriptional complexes. PDE5 inhibition prevents cardiac fibrosis by reducing CBP1 recruitment to Smad transcriptional complexes through CREB activation in cardiac fibroblasts.

Application of Computer-Aided Tongue Inspection for Preliminary Screening of Esophageal Cancer

Objective： To differentiate patients with esophageal cancer or premalignant lesions from the high-risk population for preliminary screening of esophageal cancer using a feature index determined by a computer-aided tongue information acquisition and processing system （DS01-B）. Methods： Totally, 213 patients diagnosed with esophageal cancer or premalignant lesions and 2,840 normal subjects were collected including primarily screened and reexamined, all of them were confirmed with histological examinations. Their tongue color space values and manifestation features were extracted by DS01-B and analyzed. Firstly, the analysis of variance was performed to differentiate normal subjects from patients with esophageal cancer and premalignant lesions. Secondly, the logistic regression was conducted using 10 features and gender, age to get a predictive equation of the possibility of esophageal cancer or premalignant lesions. Lastly, the equation was tested by subjects undergoing primary screening. Results： Saturation （S） values in the HSV color space showed significant differences between patients with esophageal cancer and normal subjects or those with mild atypical hyperplasia （P〈0.05）; blue-to-yellow （b） values in the Lab color space showed significant differences between patients with esophageal cancer or premalignant lesions and normal subjects （P〈0.05）. Logistic regression analysis showed that the computer-aided tongue inspection approach had an accuracy of 72.3% （2008/2776） in identifying patients with esophageal cancer or premalignant lesions for preliminary screening in high-risk population. Conclusion： Computer-aided tongue inspection, with descriptive and quantitative profile as described in this study, could be applied as a cost- and time- efficient, non-invasive approach for preliminary screening of esophageal cancer in high-risk population.

Background Estrogen deficiency contributes to postmenopausal osteoporosis. Periosteum might be a potential target of estrogen, but the underlying mechanism at gene level is far from being elucidated. The objective of this study was to investigate the correlation between estrogen and fatty acid synthase （FAS） expression in periosteum. Methods Human periosteum cells were cultured in vitro. Expressed genes in the substrated cDNA library were verified using semi-quantitative PCR and real-time PCR. The expression of FAS in periosteum of ovarectomized （OVX） SD rats was investigated. Results FAS gene was most significantly expressed in the subtracted cDNA library of periosteal cells screened by semi-quantitative PCR. Low FAS expression was verified by real-time PCR in the estrogen exposed human periosteum rather than in the control. The estradiol levels were （20.81±12.62） pg/ml, （19.64±4.35） pg/ml and （13.47±1.84） pg/ml in the sham group, the control, and the OVX group, respectively. The estradiol levels in the OVX group was significantly lower （P=-0.0386）. The FAS gene expression in periosteum in the OVX group, sham group, and control group was 3.09±1.97, 1.33±0.47 and 1.51±1.32, respectively. The gene expression in the OVX group was significantly higher （P=0.0372）. Conclusion Estrogen modulates FAS gene expression in in vitro human perisoteum as well as in in vivo rat periosteum.

Herein,a turn-on fluorescent probe for the detection of lead ions was developed using 7-diethylaminocoumarin as the fluorophore and dibenzo-18-crown-6 as the recognition *** response performance to lead ions was systematically *** probe showed specific selectivity and high sensitivity to lead ions,with fluorescence intensity at 496 nm increasing linearly with lead ion concentration(R^(2)=0.995)over the range of 1.0×10^(−7)–1.0×10^(−6)mol/L,and an LOD of 11.4 nmol/***’s plot revealed that the probe forms a 1:1 stoichiometry complex with lead ions during the recognition ***,the sensing mechanism of the probe was confirmed by density functional theory calculations,indicating that the recognition mechanism is based on photoinduced electron transfer(PET).The introduction of lead ions blocks PET,resulting in fluorescence ***,it was applied in the detection of practical water samples and bioimaging,demonstrating high application value in the field of chemosensors.

Carbody sway behaviour of railway vehicles due to track irregularity from rail alternate side wear

Track irregularity from rail alternate side wear is manifested as uneven rail wear waveforms alternating in the left and right rails with equal intervals, which will cause carbody sway behaviour of railway vehicles and greatly influences the passenger comfort. In this work, the carbody sway behaviour and mechanism due to track irregularity from rail alternate side wear and possible solutions to this issue were studied by the field testing and numerical calculation approaches. First, the carbody sway of an urban rail transit train is introduced with full-scale field tests, through which the rail alternate side wear is characterized and the formatted track irregularity are presented. Then, multibody vehicle dynamic models are developed to reproduce the carbody sway behaviour induced by the track irregularity from the rail alternate side wear. The creep forces acting on the wheel and rail are preliminarily discussed to study the influence of the carbody sway on the wear of the wheel flange and the rail corner. Finally, some potential solutions, e.g. improving the damping ratio of carbody rigid mode and rail grinding, are proposed to relieve this issue. It is concluded that an increased damping ratio of the carbody mode can alleviate the carbody sway and wheel-rail interactions, while properly maintaining track conditions can improve the vehicle performance.

Medium–Extended Range Forecasting of Meiyu in 2023: Performance of CMA and ECMWF GEPSs

Meiyu, featuring prolonged periods of rainfall over the yangtze–Huai River basin(YHRB), not only replenishes water resources and sustains ecological balance, but also poses potential disaster risks. Accurate early forecasting of Meiyu is crucial for effectively implementing flood prevention strategies. To help refine numerical models and providing guidance for operational forecasters, this study explores the capabilities of two global ensemble prediction systems(GEPSs) of the China Meteorological Administration(CMA) and the ECMWF in forecasting the Meiyu characteristics in 2023 over the YHRB. Results show that the ECMWF GEPS reasonably forecasts the Meiyu rainfall, while the CMA GEPS presents a notable underestimation. The predictable lead time of the Meiyu onset date is eight days by the ECMWF GEPS and six days by the CMA GEPS, respectively. Regarding the regional rainstorm processes, the two GEPSs generally provide a predictable lead time of 48–168 h for reasonably forecasting the patterns of the heavy rainfall area. To further examine their strengths and weaknesses in Meiyu forecasting, this paper revisits their abilities in forecasting key influence systems. By verifying against their respective analyses, it is demonstrated that the ECMWF GEPS reasonably forecasts the spatial coverages of the northwestern Pacific subtropical high(NWPSH) and the South Asian high(SAH), whereas the CMA GEPS presents substantial underestimation. Both GEPSs show generally southward deviations for the eastern ridge line position(RLP) of the SAH, and exhibit a northward deviation for the western RLP of the NWPSH during early forecast lead times. The less Meiyu rainfall predicted by the CMA GEPS compared to the ECMWF GEPS can be attributed to its weaker low-level convergence belt and weaker upper-level divergence area.
