AGK2 pre-treatment protects against thioacetamide-induced acute liver failure via regulating the MFN2-PERK axis and ferroptosis signaling pathway

Background:Acute liver failure(ALF)is an unpredictable and life-threatening critical *** pathological characteristic of ALF is massive necrosis of hepatocytes and lots of inflammatory cells infiltration which may lead to multiple organ ***:Animals were divided into 3 groups,normal,thioacetamide(TAA,ALF model)and TAA+*** L02 cells were divided into 5 groups,normal,TAA,TAA+mitofusin 2(MFN2)-siRNA,TAA+AGK2,and TAA+AGK2+MFN2-siRNA *** liver histology was evaluated with hematoxylin and eosin staining,inositol-requiring enzyme 1(IRE1),activating transcription factor 6β(ATF6β),protein kinase R(PKR)-like endoplasmic reticulum kinase(PERK)and phosphorylated-PERK(p-PERK).C/EBP homologous protein(CHOP),reactive oxygen species(ROS),MFN2 and glutathione peroxidase 4(GPX4)were measured with Western blotting,and cell viability and liver chemistry were also *** endoplasmic reticulum membranes(MAMs)were measured by ***:The liver tissue in the ALF group had massive inflammatory cell infiltration and hepatocytes necrosis,which were reduced by AGK2 *** comparison to the normal group,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+in the TAA-induced ALF model group were significantly increased,which were decreased by AGK2 *** levels of MFN2 and GPX4 were decreased in TAA-induced mice compared with the normal group,which were enhanced by AGK2 *** with the TAA-induced L02 cell,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+were further increased and levels of MFN2 and GPX4 were decreased in the MFN2-siRNA ***2 pre-treatment decreased the apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+and enhanced the protein expression of MFN2 and GPX4 in MFN2-siRNA treated L02 *** observation showed that level of MAMs was promoted in the AGK2 pre-treatment group when compared with the TAA-induced gr

Cell-free expression systems have emerged as a versatile and powerful platform for metabolic engineering,biosynthesis and synthetic biology ***,successful examples of the synthesis of complex natural products using this system are still ***,a structurally unique and complex diketopiperazine alkaloid,is a clinically promising antibiotic that selectively inhibits the transcription termination factor ***,we established a modular cell-free expression system with cascade catalysis for the biosynthesis of bicyclomycin from a chemically synthesized *** six cell-free expressed biosynthetic enzymes,including five iron-andα-ketoglutarate-dependent dioxygenases and one cytochrome P450 monooxygenase,were active in converting their substrates to the corresponding *** co-expressed enzymes in the cell-free module were able to complete the related partial *** vitro biosynthesis of bicyclomycin was also achieved by reconstituting the entire biosynthetic pathways(i.e.,six enzymes)using the modular cell-free expression *** study demonstrates that the modular cell-free expression system can be used as a robust and promising platformforthe biosynthesis of complex antibiotics.

Interaction between circulating cancer cells and platelets:clinical implication

Metastasis is the main cause of cancer-associated mortality. During this complicated process, some cancer cells, also called circulating tumor cells （CTCs）, detach from primary sites, enter bloodstream and extravasate at metastatic site. Thrombocytosis is frequently observed in patients with metastatic cancers suggesting the important role of platelets in metastasis. Therefore this review focuses on how platelets facilitate the generation of CTCs, protect them from various host attacks, such as immune assaults, apoptosis and shear stress, and regulate CTCs intravasation/extravasation. Platelet-derived cytokines and receptors are involved in this cascade. Identification the mechanisms underlie platelet-CTCs interactions could lead to the development of new platelet-targeted therapeutic strategy to reduce metastasis.

Multifunctional cellular carbon foams derived from chitosan toward self-cleaning, thermal insulation, and highly efficient microwave absorption properties

To adapt the practical demand,designing and constructing the multifunctional microwave absorbers(MAs)is the key future direction of research and ***,effective integrating the multiple functions into a single material remains a huge ***,cellular carbon foams(CCFs)with different porous structures were elaborately designed and fabricated in high efficiency through a facile continuous freeze-drying and carbonization processes using a sustainable biomass chitosan as the *** obtained results revealed that the thermal treated temperature and g-C_(3)N_(4) amount played a great impact on the carbonization degrees,pore sizes,and morphologies of CCFs,which led to their tunable electromagnetic(EM)parameters,improved conduction loss,and polarization loss *** to the special cellular structure,the designed CCFs samples simultaneously displayed the strong absorption capabilities,broad absorption bandwidths,and thin matching ***,the as-prepared CCFs exhibited the strong hydrophobicity and good thermal insulation,endowing its attractive functions of self-cleaning and thermal ***,our findings not only presented a facile approach to produce different porous structures of CCFs,but also provided an effective strategy to develop multifunctional high-performance MAs on basis of three-dimensional CCFs.

Proteome changes in the plasma of Papilio xuthus (Lepidoptera:Papilionidae):effect of parasitization by the endoparasitic wasp Pteromalus puparum (Hymenoptera:Pteromalidae)

Although the biochemical dissection of parasitoid-host interactions is becoming well characterized, the molecular knowledge concerning them is minimal. In order to understand the molecular bases of the host immune response to parasitoid attack, we explored the response of Papilio xuthus parasitized by the endoparasitic wasp Pteromalus puparum using proteomic approach. By examining the differential expression of plasma proteins in the parasitized and unparasitized host pupae by two-dimensional (2D) electrophoresis, 16 proteins were found to vary in relation to parasitization compared with unparasitized control samples. All of them were submitted to identification by mass spectrometry coupled with a database search. The modulated proteins were found to fall into the following functional groups: humoral or cellular immunity, detoxification, energy metabolism, and others. This study contributes insights into the molecular mechanism of the relationships between parasitoids and their host insects.

Applying systematic conservation planning to constitute a protection strategy for broad-leaved Korean pine forests in Changbai Mountains, China

The broad-leaved Korean pine(Pinus koraiensis) forest is one of the most biodiverse zonal communities in the North Temperate Zone and an important habitat for many endangered ***-leaved Korean pine forests(BKPFs) are shrinking quickly due to deforestation and rapid urbanization. Thus, scientific protection strategies are urgently needed to change this status. Changbai Mountains contains one of the largest BKPFs and is considered a priority biodiversity conservation area in China. Guided by systematic conservation planning(SCP) methods and procedures, we chose representative species and communities in BKPFs ecosystem as priority conservation objects, and set quantitative conservation target, which is in the light of the biodiversity characteristic of BKPFs. The watershed area is used as planning unit. We used CPlan software to calculate the irreplaceability(Ir)value of each planning unit and the contribution value(Ti) of each conservation object to(1) assess the conservation efficiency;(2) identify the conservation gap of the existing conservation network. Then wecalculated a human disturbance index(HDI) for planning units in the conservation gaps and combine this with the Ir value to design three conservation scenarios to optimize the conservation *** show that planning units with high conservation value 14.16% of the total area, with3084.36 km2 were covered by the existing conservation network. 79.28% of planning units with high conservation value have not been protected which were concentrated mainly in the eight gap *** 25.3% of protection objects achieved their conservation target with the existing conservation network. Conservation efficiency is low. Three conservation scenarios are constituted, each prioritizing a different aim:(1) ecological value;(2)species rescue; and(3) economical avoidance. Hydrothermal synthesis of polyimide-linked covalent organic frameworks towards ultrafast and stable cathodic sodium storage

Redox-active organic materials are capturing growing attention as cathode materials for sustainable alkaline metal ion ***,the storage of Na+in most organic materials-based cathodes is plagued by low capacity and unsatisfying rate performance due to their low active site densities and limited exposed active ***,two polyimide-linked covalent organic frameworks(COFs),namely HATN-PD-COF and HATN-TAB-COF,were fabricated from hydrothermal synthesis with redoxactive triphenylene-2,3,6,7,10,11-hexacarboxylic acid and aromatic amines as starting *** X-ray diffraction and electron microscopy analysis indicate the high crystalline nature of these COFs with AA stacking configuration and orderly mesoporous tunnel.N_(2) sorption measurement discloses the permanent porosity of these two COFs with a Brunauer-EmmettTeller surface area of 1,065-1,200 m^(2)g^(-1)and a large pore size of 2.0-3.1 *** intermittent titration technique and density functional theory calculations reveal the facile Na+ion diffusion along the mesoporous tunnel of these COFs with a small energy barrier of 0.13-0.40 e *** particular,the as-prepared COFs based-cathodes show ultrafast and stable Na+storage associated with their conjugated electronic structure,highly ordered mesoporous tunnel,robust structure,and redox-active C=N/C=O-rich framework as exemplified by the high reversible capacity of 210 m A h g^(-1)at 200 m A g^(-1),record-high rate performance(195 m A h g^(-1)at a high current density of 10,000 m A g^(-1))among organic electrodes and the capacity retention of nearly 91%at 10,000 m A g^(-1)after 7,000 cycles for HATN-PD-COF.

光频梳提供了光波和微波相干链接的桥梁,它的诞生革命性地提高了人们对于光学频率和时间的测量精度,深刻影响着当今世界科技的发展.最早的光频梳产生于锁模激光器系统,然而基于锁模激光器的光梳,因其系统复杂、体积庞大和价格高昂,一般仅限于实验室应用.近年来一种新型光频梳应运而生,并有望解决上述问题.它是通过连续激光耦合进入高品质光学微腔而激发的,在频域上通过四波混频产生等间距的频率分量,在时域上则利用非线性效应平衡微腔色散而形成锁模,这种新型光频梳被称为“微腔光梳”.相比于传统光梳,微腔光梳有着尺寸小、可集成、功耗低和重频范围大等优势,它的出现标志着产生光梳迈向芯片级尺寸的时代,并引起了科学界和工业界越来越多的关注.本文首先概述了微腔光梳的产生与发展历程,随后介绍微腔光梳在实际应用方面取得的进展,最后对微腔光梳当前的问题进行总结,并对未来发展进行展望.
