Peficitinib suppresses diffuse-type tenosynovial giant cell tumor by targeting TYK2 and JAK/STAT signaling

Diffuse-type tenosynovial giant cell tumor(dTGCT) is a destructive but rare benign proliferative synovial neoplasm. Although surgery is currently the main treatment modality for dTGCT, the recurrence risk is up to 50%. Therefore, there is a great need for effective drugs against dTGCT with minor side effects. The Janus kinase(JAK)/signal transducer and activator of transcription(STAT) signaling plays a central role in rheumatoid arthritis(RA), a disease with similar characteristics as dTGCT, but its function in dTGCT remains *** fibroblast-like synoviocytes(FLS) and macrophages were isolated from 10 synovial tissue samples from dTGCT patients for the screening and validation of the five clinically approved JAK inhibitors to treat RA against dTGCT. Cell viability, cell death, inflammation and the activity of the JAK family members of cultured dTGCT FLS(both 2-D and 3-D) and macrophages were investigated for the efficacy of the JAK inhibitors. Here, we found that similar to RA, JAK/STAT signaling was markedly activated in the dTGCT synovium. Of the 5 JAK inhibitors, peficitinib was shown to have the most potency in addressing some of the pathological responses of dTGCT FLS and *** potency of peficitinib was much higher than pexidartinib, which is the only FDA-approved drug for dTGCT. Mechanistically, peficitinib inhibited tyrosine kinase 2(TYK2), a JAK family member necessary for the pathological progression of dTGCT FLS and macrophages. In summary, we not only revealed JAK/STAT(especially TYK2) signaling as the major mechanism underlying dTGCT, but also identified peficitinib as a promising drug against dTGCT.

型钢混凝土构件是承载能力、耐久性与耐火性良好的结构构件,在桥梁工程中的应用有巨大潜力。在诸多截面形式中,内置十字形型钢可保障桥墩的刚度均匀性,相比其他的截面形式,具有更为广泛的应用。在桥梁服役期间,失控车辆与船舶对桥墩的撞击作用需要加以考虑。桥墩通常采用底部固接而顶部通过橡胶支座等与桥板搭接的边界条件,与常规针对构件层次的相关撞击试验边界有显著差异。基于数值模拟方法,利用验证有效的有限元模型,模拟分析了内置十字形型钢的型钢混凝土桥墩在侧向撞击作用下的动力响应规律,结果表明,在现实边界和撞击作用位置条件下除需考虑桥墩撞击位置的瞬时变形,还需要关注因受撞和其后的惯性回弹而导致的桥墩顶部变形。同时,基于其中的关键响应与指标,开展了系列的参数分析,所涉及的参数包括桥墩的材料相关参数、截面特征参数、宏观几何参数和撞击作用工况参数等。上述研究将有助于内置十字形型钢的型钢混凝土桥墩的抗撞击设计,保障相应桥梁结构安全。

Influence of the rolling-resistance-based shape of coarse particles on the shear responses of granular mixtures

Shear responses of dense granular mixtures of spherical coarse particles with the rolling resistance and spherical fine particles are studied via triaxial compression tests using the discrete element *** macroscale responses(shear strength and dilatancy)are *** the results with those in the literature indicates that granular mixtures with a rolling resistance coefficient of 0.5 have similar macroscale responses to those of gravel-shaped coarse particle *** quantify the microscale responses including the percentage contributions of contact types,partial coordination number,average particle rotation,average degree of interlocking,and local structural properties,A detailed analysis of the force-fabric anisotropy reveals the mechanisms of the variations in the shear strength with the rolling resistance coefficient and the fines *** mechanism of the variation in the shear strength with the fines content for granular mixtures with a rolling resistance coefficient of 0.5 is different from that for gravel-shaped coarse particle ***,we find that a rolling resistance linear model weakens the linear relationship between the stress ratio and the fabric anisotropy of strong and non-sliding contacts when the fines content is 30% and 40%.

Shear behaviours of cohesionless mixed soils using the DEM:The influence of coarse particle shape

The coarse particles in mixed soils can be cobbles or gravels,with the main difference being their roundness(an indicator describing particle shape characteristics at an intermediate scale).The influence of coarse particle shape(i.e.,roundness)on the macroscopic and microscopic shear behaviours of cohesionless mixed soils with various fines contents(FCs)was investigated via the discrete element method in this *** shapes of coarse particles were formed using the rotation-invariant spherical harmonic method proposed by previous *** equation was proposed to predict the initial void ratios of samples in this study.A decrease in the roundness of coarse particles can increase the peak friction angle(FC≤40%)and critical friction angle(FC≤30%).As the roundness of coarse particles decreases,the peak dilatancy angle initially increases and then decreases(FC≤20%).Furthermore,it was found that the roundness of coarse particles hardly affects the classification of cohesionless mixed soils,as determined by probing the percentage contributions of coarse-coarse,coarse-fine,and fine-fine *** cohesionless mixed soils change from an underfilled structure to an interactive-underfilled structure at the critical state,the main forms of coarse-coarse contacts were ***,the force-fabric anisotropy mechanisms of the influences of the roundness and rolling resistance coefficient of coarse particles on the shear strengths of cohesionless mixed soils were found to be different.

Vapor-liquid equilibria of CH_4, CO_2 and their binary system CH_4 + CO_2: A comparison between the molecular simulation and equation of state

An accurate knowledge about phase behaviors of CH4, CO2 and their binary mixture is crucial in fields of natural gas liquefaction and refrigeration applications. In this work, two all-atom force fields of TraPPE-EH and EMP2 were used for the components CH4 and CO〉 respectively. Then the vapor-liquid equilibria （VLE） of CH4, CO2 and their binary system were calculated via the NVT- and NpT Gibbs Ensemble Monte Carlo Simulations. Meanwhile the traditional method using Equation of State （EoS） to correlate the VLE properties was also investigated. The EoSs considered in this work were three classic cubic RK, SRK, PR and another advanced molecular-based PC-SAFT equations. For pure components, both molecular simulations and the PC-SAFT EoS could obtain satisfactory predictions for all the saturated properties. However, the saturated liquid densities calculated by the cubic EoSs were not so good. It was also observed that the TraPPE-EH force field had a good representation for CH4 molecule, while the EMP2 force field was not enough accurate to represent CO2 molecules. For the mixture CH4 ＋ CO2, SRK and PR showed the best predictions for the saturated pressure-component property, while good results were also obtained via molecular simulations and PC-SAFT EoS. It was suggested that special combining rules or binary interaction parameters were important to obtain enough accurate prediction of the mixed phase behavior. Compared with the cubic EoS, the PC-SAFT and molecular simulation method showed better adaptabilities for both the pure and mixture systems. Besides, the accurate molecular parameters used in the PC-SAFT and molecular simulations could bring about direct and deep understanding about the molecular characteristics.

Probing the Néel Order in Altermagnetic RuO_(2) Films via X-Ray Magnetic Linear Dichroism

The emerging altermagnetic RuO_(2)with both compensated magnetic moments and broken time-reversal symmetry possesses nontrivial magneto-electronic responses and nonrelativistic spin currents that are closely related to the magnetic easy *** M_(3)-edge X-ray magnetic linear dichroism(XMLD)measurements were performed to probe the Néel order in RuO_(2).For epitaxial RuO_(2) films,characteristic XMLD signals were observed for either RuO_(2)(100)or RuO_(2)(110)at normal incidence or RuO_(2)(001)at oblique *** signals disappeared when the test temperature exceeded the Néel *** non-epitaxial RuO_(2) films,the flat lines in the XMLD patterns of RuO_(2)(100)and RuO_(2)(110)demonstrate that there is no in-plane uniaxial alignment of the Néel order in these samples because of the counterbalanced Néel order of the twin crystals,as evidenced by X-ray diffraction phi-scan *** experimental results unambiguously demonstrate antiferromagnetism in RuO_(2) films and reveal the spatial relation of the Néel order parallel to the RuO_(2)[001]crystalline *** findings deepen the understanding of RuO_(2) and other attractive altermagnetic materials used in the field of spintronics.

Block copolymers with different backbone compositions have been prepared by the condensation of dimethylamino terminated poly（dimethylsiloxane）（PDMS） and hydroquinone terminated poly（phthalazinone ether nitrile）（PPEN） in the presence of chlorobenzcne/N-methyl pyrrolidone （NMP） as solvents. The products were characterized by FTIR, ^1H NMR and gel permeation chromatography. Differential scanning calorimetry analysis indicated that the block copolymers showed separated microphase.

Impact of climate change on wheat yield and quality in the Yellow River Basin under RCP8.5 during 2020–2050

Studying the impact of future climate change on the yield and quality of wheat in the Yellow River Basin(YRB)is both essential for rationalizing the utilization of climate resources and ensuring national food *** this study,we employed corrected refined meteorological grid data(30 km×30 km)generated by the Climate-Weather Research and Forecasting model under RCP8.5,coupled with the DSSAT-CERES-Wheat model to predict the impact of climate change on wheat phenology,yield,and grain nitrogen content(GNC)in the YRB from 2020 to *** to the type of wheat variety and planting management in the YRB,we designed five wheat ecological regions(I-V).The results show that from 2020 to 2050,the agro-meteorological resource,involving temperature,solar radiation,and precipitation,would have an overall increasing trend during the wheat growth seasons in the *** growth period of wheat was generally shortened by approximately 8 d,including 6 d less from sowing to anthesis,and 2 d less from anthesis to *** general,the predicted wheat yield showed a decreasing trend in the YRB,with an average reduction of 0.19%over the period from 2020 to 2050 compared with the baseline period from 1975 to 2005,and the spatial variation in wheat yield change was considerably *** the future,the Qing‒Zang Spring Wheat region(V)in the western part of the YRB would be the only region with a yield increase of 35.4%,whereas the Northern Spring Wheat region(I)in the northern part of the YRB would be the region exhibiting the greatest decrease of 9.9%.Moreover,compared with the baseline,the GNC would increase by an average of 2.3%in the YRB,with a significant increase in the southern and central parts during *** GNC increase in the Huang‒Huai Winter Wheat region(III)was the highest with a 12.5%increment,but the Qing‒Zang Spring Wheat region(V)is the only region where GNC would decline,with an average decrease of 18.9%.This study implies that wheat yield and GNC have a
