Key genes and regulatory networks for diabetic retinopathy based on hypoxia-related genes:a bioinformatics analysis

AIM:To prevent neovascularization in diabetic retinopathy(DR)patients and partially control disease ***:Hypoxia-related differentially expressed genes(DEGs)were identified from the GSE60436 and GSE102485 datasets,followed by gene ontology(GO)functional annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment *** candidate drugs were screened using the CMap ***,a protein-protein interaction(PPI)network was constructed to identify hypoxia-related hub genes.A nomogram was generated using the rms R package,and the correlation of hub genes was analyzed using the Hmisc R *** clinical significance of hub genes was validated by comparing their expression levels between disease and normal groups and constructing receiver operating characteristic curve(ROC)***,a hypoxia-related miRNA-transcription factor(TF)-Hub gene network was constructed using the NetworkAnalyst online ***:Totally 48 hypoxia-related DEGs and screened 10 potential candidate drugs with interaction relationships to upregulated hypoxia-related genes were identified,such as ruxolitinib,meprylcaine,and *** addition,8 hub genes were also identified:glycogen phosphorylase muscle associated(PYGM),glyceraldehyde-3-phosphate dehydrogenase spermatogenic(GAPDHS),enolase 3(ENO3),aldolase fructose-bisphosphate C(ALDOC),phosphoglucomutase 2(PGM2),enolase 2(ENO2),phosphoglycerate mutase 2(PGAM2),and 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3).Based on hub gene predictions,the miRNA-TF-Hub gene network revealed complex interactions between 163 miRNAs,77 TFs,and hub *** results of ROC showed that the except for GAPDHS,the area under curve(AUC)values of the other 7 hub genes were greater than 0.758,indicating their favorable diagnostic ***:PYGM,GAPDHS,ENO3,ALDOC,PGM2,ENO2,PGAM2,and PFKFB3 are hub genes in DR,and hypoxia-related hub genes exhibited favorable diagnostic perform

Neuropeptides are crucial in regulation of a rich variety of developmental,physiological,and behavioral functions throughout the life cycle of *** an integrated approach of multiomics,we identified neuropeptide precursors in the greater wax moth Galleria mellonella,which is a harmful pest of honeybee hives with a worldwide ***,a total of 63 and 67 neuropeptide precursors were predicted and annotated in the *** genome and transcriptome,in which 40 neuropeptide precursors were confirmed in the *** ***,we identified 12 neuropeptide precursor genes present in *** but absent in honeybees,which may be potential novel pesticide target *** hives were contaminated with heavy metals such as lead,enabling its bioaccumulation in *** bodies through the food chain,we performed transcriptome sequencing to analyze the effects of Pb stress on the mRNA expression level of *** neuropeptide *** treatment by Pb,the expression of neuropeptide F1 was found to be significantly downregulated,implying that this neuropeptide might be associated with responding to the heavy metal stress in *** study comprehensively identified neuropeptide precursors in ***,and discussed the effects of heavy metals on insect neuropeptides,with the example of *** results are valuable for future elucidation of how neuropeptides regulate physiological functions in *** and contribute to our understanding of the insect's environmental plasticity and identify potential new biomarkers to assess heavy metal toxicity in insects.

Plasma-assisted ammonia synthesis under mild conditions for hydrogen and electricity storage:Mechanisms,pathways,and application prospects

Predicting the strut forces of the steel supporting structure of deep excavation considering various factors by machine learning methods

The application of steel strut force servo systems in deep excavation engineering is not widespread,and there is a notable scarcity of in-situ measured *** presents a significant research gap in the *** this,our study introduces a valuable dataset and application scenarios,serving as a reference point for future *** main objective of this study is to use machine learning(ML)methods for accurately predicting strut forces in steel supporting structures,a crucial aspect for the safety and stability of deep excavation *** employed five different ML methods:radial basis function neural network(RBFNN),back propagation neural network(BPNN),K-Nearest Neighbor(KNN),support vector machine(SVM),and random forest(RF),utilizing a dataset of 2208 measured *** points included one output parameter(strut forces)and seven input parameters(vertical position of strut,plane position of strut,time,temperature,unit weight,cohesion,and internal frictional angle).The effectiveness of these methods was assessed using root mean square error(RMSE),correlation coefficient(R),and mean absolute error(MAE).Our findings indicate that the BPNN method outperforms others,with RMSE,R,and MAE values of 72.1 kN,0.9931,and 57.4 kN,respectively,on the testing *** study underscores the potential of ML methods in precisely predicting strut forces in deep excavation engineering,contributing to enhanced safety measures and project planning.

Experimental investigation on rockburst characteristics of highly stressed D-shape tunnel subjected to impact load

Rockburst has always been a challenge for the safe construction of deep underground *** study investigated the rockburst characteristics in highly-stressed D-shape tunnels under impact loads from rock blasting and other mining-related dynamics *** biaxial Hopkinson pressure bar was utilized to apply varying biaxial prestress and the same impact loads to cube specimens with D-shape ***-speed camera and digital image correlation(DIC)were used to capture the failure process and strain field of *** test results demonstrate that the D-shape hole specimen experience rockburst under coupled static stress and impact *** this circumstance,the rockburst mechanism of the D-shaped hole specimens involves spalling in sidewall induced by impact load,indicating dynamic tensile *** high static prestress provides the initial stress field,while the impact load disrupts the stress equilibrium,result in the stress or strain concentration in the sidewall of the D-shape hole,inducing ***,the rockburst process can be divided into(1)calm stage,(2)crack initiation,propagation,and coalesce stage,(3)spalling stage and(4)rock fragments ejection *** load triggers rockburst occurrence,while vertical stress further determines the rockburst *** influence range and magnitude of strain concentration zone and displacement deformation of the tunnel surrounding rock increases with increasing vertical stress,thus inducing more severe rockburst.

Projections of summer light rain frequency in typical terrain over eastern China under wind speed constraint

The variation in near-surface wind speed is a key dynamic parameter in the orographic effect of precipitation over eastern *** this study,we used the latest high-resolution outputs from six GCMs in CMIP6-HighResMIP to evaluate the performance of high-resolution models in simulating the orographic precipitation characteristics of typical mountainous areas in summer over eastern *** orographic precipitation under warming scenarios was projected and constrained according to observational *** results indicated that during the contemporary climate reference period(1979-2009),although the relationship between model-simulated near-surface wind speed and orographic light rain frequency was consistently stable,the sensitivity of the orographic light rain frequency to surface wind variability was generally underestimated,with a deviation approximately 24.1% lower than the observational *** estimated orographic light rain frequency corrected based on the observed near-surface wind speed under a 1.5℃ warming scenario,was 36.1% lower than that of the contemporary period;this reduction was 8.6 times that without the wind speed constraint(4.2%).The MRI-AGCM3-2-S model,with a longer dataset,demonstrated relatively stable reductions in orographic light rain frequency under different warming scenarios(1.5℃,2℃,3℃,and 4℃)after the application of wind speed *** all cases,the reductions exceeded those for the predictions made without the wind speed constraint.

Lactobacillus plantarum AR113 attenuates liver injury in D-galactose-induced aging mice via the inhibition of oxidative stress and endoplasmic reticulum stress

Probiotics could effectively eliminate excess reactive oxygen species(ROS)generated during aging or lipid metabolism disorders,but their mechanism is *** major purpose of this study was to investigate the mechanism of Lactiplantibacillus plantarun AR113 alleviating oxidative stress injury in the D-galactose induced aging *** result showed that pretreatment with *** AR113 significantly relieving H_(2)O_(2)induced cytotoxicity in HepG2 cells by maintain cell membrane integrity and increasing antioxidant enzyme *** D-galactose induced aging mice,*** AR113 could significantly attenuate liver damage and inflammatory infiltration by promoting endogenous glutathione(GSH)synthesis and activating the Nrf2/Keap1 signaling pathway in mice,and increasing the expression of regulated phaseⅡdetoxification enzymes and antioxidant *** analysis shown that gavage of *** AR113 could significantly reduce the expression of G protein-coupled receptor 78(GPR78)and C/EBP homologous protein(CHOP)proteins,and promote the restoration of endoplasmic reticulum(ER)homeostasis,thereby activating cell anti-apoptotic *** results were also confirmed in H_(2)O_(2)-treated HepG2 *** indicated that *** AR113 could inhibit D-galactose-induced liver injury through dual inhibition of ER stress and oxidative *** AR113 have good application potential in anti-aging and alleviating metabolic disorders.

To the Editor:Acute pancreatitis(AP)is the most common gastrointestinal condition that leads to acute hospitalization.[1]The incidence of AP has increased in recent decades,with a current global incidence of 34/100,000,probably due to a rising trend of obesity and gallstone *** is associated with remarkable pain,reduced quality of life(QoL),and even death,thus posing a heavy socioeconomic *** addition,approximately 22%of AP patients experience recurrent AP(RAP)and 10%of AP patients progress to chronic pancreatitis.[2]Petrov and Yadav[3]have,therefore,argued that AP should not be considered as an acute and self-limiting *** and long-term care after AP have drawn widespread attention,which has improved patients’physical and mental health.
