Background The clinical importance of glycaemic control in patients with diabetes has been well established. This study aimed to explore twice-daily biphasic insulin aspart 30 （BIAsp 30） for insulin initiation in patients with type 2 diabetes mellitus （T2DM） who had poor glycaemic control with human insulins （His）. We use data from a Chinese cohort of the PRESENT *** In the 3-month study, Chinese subjects with T2DM started insulin therapy with BIAsp 30 in routine care. Glycaemic control was measured by glycosylated hemoglobin （HbA1c）, fasting plasma glucose （FPG） and posting plasma glucose （PPG）. The safety assessment included hypoglycaemia and other adverse *** A total of 1989 subjects previously treated with His were switched to BIAsp 30 for 3-month treatment. Mean HbA1c, FPG and PPG were significantly improved after the therapy. The overall rate of hypoglycaemia decreased at the end of the trial except for the patients previously treated with long-acting insulin. Most of the events were minor and diurnal hypoglycaemia. Only one serious adverse drug reaction （SADR）, a local hypersensitivity, was reported. The majority of the patients （296.7%） and physicians （≥84.7%） were either satisfied or very satisfied with the treatment using BIAsp 30 compared with previous HI *** The BIAsp 30 treatment improved both glycaemic control and patients＇ satisfaction without increasing hypoglycaemia in T2DM subjects inadequately controlled by Hls.

In order to study the response law of vortex-induced vibration(VIV)of marine risers under the combined action of roughness and interference effects,and to reveal the coupling mechanism of roughness and interference effects on the riser,a VIV experiment of rough risers in tandem arrangement was conducted in a wave−current combined *** experiment characterized the risers’roughness by arranging different specifications of attachments on the surface of the *** rough risers with different roughness and smooth risers were arranged in tandem arrangement,with the rough risers arranged *** experimental results indicate that the suppression of the attachments on the downstream risers’vibration are more significant both in the CF and IL directions as the reduced velocity *** the downstream riser,the amplitude response of rough riser is more significantly weakened compared with the smooth one at high reduced *** the upstream risers,changes in the roughness and spacing ratio have an impact on their‘lock-in’*** the roughness of downstream risers is relatively large(0.1300)and the spacing between risers is small(S/D=4.0),the reduced velocity range of‘lock-in’region in the CF direction of upstream risers is obviously expanded,and the displacement in the‘lock-in’region is severer.

A vortex-induced vibration(VIV)experiment of rough risers with coupling interference effect under a side-by-side arrangement was carried out in a wave-current combined *** roughness of the riser was characterized by arranging different specifications of surface attachments on the surface of the *** risers with three different roughnesses were arranged side by side with smooth risers to explore the VIV response of the riser under the combined action of roughness and interference effect,and to reveal the coupling mechanism between roughness and interference *** experimental results show that,compared with that of a smooth riser,the VIV of a rough riser under the coupling interference effect has a wider"lock-in"region,and the displacement decreases more significantly at a high reduced velocity,which is more likely to excite higher-order modes and frequency *** addition,the displacement response and frequency response of the smooth riser are not significantly affected by wake interference from the rough riser,which is caused by the decrease of the wake region due to the delay of the boundary layer separation point of the rough riser.

Protein phosphorylation regulates a variety of important cellular and physiological processes in ***-depth profiling of plant phosphoproteomes has been more technically challenging than that of animal *** is largely due to the need to improve protein extraction efficiency from plant cells,which have a dense cell wall,and to minimize sample loss resulting from the stringent sample clean-up steps required for the removal of a large amount of biomolecules interfering with phosphopeptide purification and mass spectrometry *** this end,we developed a method with a streamlined workflow for highly efficient purification of phosphopeptides from tissues of various green organisms including Arabidopsis,rice,tomato,and Chlamydomonas reinhardtii,enabling in-depth identification with high quantitative reproducibility of about 11000 phosphosites,the greatest depth achieved so far with single liquid chromatography-mass spectrometry(LC-MS)runs operated in a data-dependent acquisition(DDA)*** mainstay features of the method are the minimal sample loss achieved through elimination of sample clean-up before protease digestion and of desalting before phosphopeptide enrichment and hence the dramatic increases of time-and *** method,named GreenPhos,combined with single-shot LC-MS,enabled in-depth quantitative identification of Arabidopsis phosphoproteins,including differentially phosphorylated spliceosomal proteins,at multiple time points during salt stress and a number of kinase substrate *** is expected to serve as a universal method for purification of plant phosphopeptides,which,if samples are further fractionated and analyzed by multiple LC-MS runs,could enable measurement of plant phosphoproteomes with an unprecedented depth using a given mass spectrometry technology.

Background Thyroid peroxidase （TPO） is an important autoantigen in Hashimoto＇s thyroiditis （HT）, and almost all epitopes are located in TPO ectodomain. The glycosylation of TPO might contribute to breaking self-tolerance, therefore, pudfied glycosylated recombinant TPO ectodomain is prerequisite of elucidating its role in the pathogenesis of HT. The aim of our study was to investigate whether the glycosylation has influence on the antigenic determinants of recombinant TPO. Methods Bac-to-Bac baculovirus expression system was used to generate recombinant human TPO ectodomain. The antigenicity was analyzed by antigen specific enzyme-linked immunosorbant assays （ELISAs）. The glycosylation of recombinant human TPO ectodomain of High Five insect cell origin was detected by lectin-ELISAs. Results TPO ectodomain was recovered from the culture media as a soluble protein, and it was fused with a hexahistidine tag which allowed purification by nickel-affinity chromatography. The recombinant TPO ectodomain could be recognized by all the 54 HT patients and three TPO monoclonal antibodies. Fucose, sialic acid and galactose were all detected on the recombinant TPO ectodomain. Sera TPOAb binding decreased slightly after non-specific deglycosylation of TPO by periodic acid. Conclusions High Five insect cells derived recombinant human TPO ectodomain had N-glycosylation sites, which might have little effect on recognition by serum TPOAb.
