Targeting-induced local lesions in genomes(TILliNG)is a powerful reverse-genetics tool that enables high-throughput screening of genomic variations in *** TILliNG has been developed for many diploid plants,the technology has been used in very few polyploid species due to their genomic ***,we established an efficient capillary electrophoresis-based TILliNG platform for allotetraploid cultivated tobacco(Nicotiana tabacum L.)using an ethyl methanesulfonate(EMS)-mutagenized population of 1,536 *** optimized the procedures for endonuclease preparation,leaf tissue sampling,DNA extraction,normalization,pooling,PCR amplification,heteroduplex formation,and capillary *** a test screen using seven target genes with eight PCR fragments,we obtained 118 *** mutation density was estimated to be approximately one mutation per 106kb on *** analyses showed that mutations in two heavy metal transporter genes,HMA2S and HMA4T,led to reduced accumulation of cadmium and zinc,which was confirmed independently using CRISPR/Cas9 to generate knockout *** results demonstrate that this powerful TILliNG platform(available at http://***)can be used in tobacco to facilitate functional genomics applications.

BACKGROUND Congenital infantile fibrosarcoma(CIF)and congenital hemangioma(CH)have similarities on prenatal ultrasound and are *** SUMMARY We report 3 cases of fetuses with superficial hypervascular tumors,confirmed by postnatal pathology as CIF(1 case)and CH(2 cases,including 1 in a twin fetus).In Case 1,a mass with a rich blood supply in the fetal axilla was discovered by prenatal ultrasound at 28+0 wk of *** postpartum pathological diagnosis was CIF,the mass was surgically removed,and the prognosis of the child was *** Case 2,at 23+1 wk of gestation,a mass was discovered at the base of the fetus’s thigh on prenatal *** postpartum pathological diagnosis was *** conservative treatment,the mass shrank *** 3 occurred in a twin *** 30+0 wk of gestation,prenatal ultrasound revealed a bulging mass with a rich blood supply on the abdominal wall of one of the *** weeks later,the affected fetus died,and the unaffected baby was successfully delivered by emergency cesarean *** affected fetus was pathologically diagnosed with *** Prenatal ultrasound can provide accurate information,such as the location,size and blood supply of a surface mass in a *** found similarities between CIF and CH in prenatal ultrasound *** it is difficult to distinguish these conditions by prenatal ultrasound alone,for superficial hypervascular tumors that rapidly increase in size in a short period,close ultrasound monitoring of the fetus is required to quickly address possible adverse outcomes.

Chromosome band 11q23 deletion predicts poor prognosis in bone marrow metastatic neuroblastoma patients without MYCN amplification

LSCHL4 from Japonica Cultivar, Which Is Allelic to NAL 1, Increases Yield of Indica Super Rice 93-11

The basic premise of high yield in rice is to improve leaf photosynthetic efficiency and coordinate the sourcesink relationship in rice plants. Quantitative trait loci （QTLs） related to morphological traits and chlorophyll content of rice leaves were detected at the stages of heading to maturity, and a major QTL （qLSCHL4） related to flag leaf shape and chlorophyll content was detected at both stages in recombinant inbred lines constructed using the indica rice cultivar 93-11 and the japonica rice cultivar Nipponbare. Map-based cloning and expression analysis showed that LSCHL4 is allelic to NAL1, a gene previously reported in narrow leaf mutant of rice. Overexpression lines transformed with vector carrying LSCHL4 from Nipponbare and a near-isogenic line of 93-11 （NIL-9311） had significantly increased leaf chlorophyll content, enlarged flag leaf size, and improved panicle type. The average yield of NIL-9311 was 18.70% higher than that of 93-11. These results indicate that LSCHL4 had a pleiotropic function. Exploring and pyramiding more high-yield alleles resem- bling LSCHL4 for super rice breeding provides an effective way to achieve new breakthroughs in raising rice yield and generate new ideas for solving the problem of global food safety.

Ground-penetrating radar study of beach-ridge deposits in Huangqihai Lake, North China： the imprint of washover processes

Determining the origin of beach ridges in lacustrine basins can often be problematic. The sedimen- tary processes responsible for formation of beach ridges on the north shore of Huangqihai Lake were investigated by using ground penetrating radar （GPR）. A 400 MHz GPR antenna was used to achieve a high vertical resolution of 0.044）.08 m. The radar stratigraphy was then determined using principles of seismic stratigraphy. The radar facies （RF） were determined by analyzing internal configuration and continuity of reflections, as well as reflection termination patterns. The identified RF fall into three groups （inclined, horizontal and irregular）. The inclined group consists of RF that display inclined reflections. The horizontal group consists of RF that exhibit predominantly horizontal reflections. In the irregular group, the reflections are typically weak. RF with reflections with gently landward dips in the shore-normal profile are interpreted as washover sheet deposits. RF with steeply landward-dipping and imbricated reflections are interpreted as washover lobes. Washover sheets develop when overwash fails to enter a significant body of water and sedimentation takes place entirely on the relatively flattened topography. Washover lobe development occurs when overwash enters a region in which topography dips steeply landward, and sedimentation takes place on the surface of washover sheets or previous washover lobes. The beach-ridge deposits are interpreted as being formed entirely from vertically and laterally stacked washover sheets and washover lobes. They were formed by wave-dominated processes and secondary overwash processes supplemented by longshore currents.

A GPU based single-pulse search pipeline(GSP)with database and its application to the Commensal Radio Astronomy FAST Survey(CRAFTS)

We developed a GPU based single-pulse search pipeline(GSP)with a candidate-archiving *** based upon the infrastructure of the open source PulsaR Exploration and Search Toolkit(PRESTO),GSP implements GPU acceleration of the de-dispersion and integrates a candidate-archiving *** applied GSP to the data streams from the Commensal Radio Astronomy FAST Survey(CRAFTS),which resulted in quasi-real-time *** integrated candidate database facilitates synergistic usage of multiple machine-learning tools and thus improves efficient identification of radio pulsars such as rotating radio transients(RRATs)and fast radio bursts(FRBs).We first tested GSP on pilot CRAFTS observations with the FAST Ultra-Wide Band(UWB)*** detected all pulsars known from the the Parkes multibeam pulsar survey in the corresponding sky area covered by the *** also discovered 13 new *** measured the computational efficiency of GSP to be~120 times faster than the original PRESTO and~60 times faster than an MPI-parallelized version of PRESTO.

Influence of nitrogen wet deposition on nitrogen output in a typical watershed in the Three Gorges Reservoir Area

In order to explore the influence of wet nitrogen(N)deposition on N output in watersheds,this study selected a typical small watershed(Chenjiagou,CJG)in the Three Gorges Reservoir(TGR)area based on one-year observation of rainfallrunoff N *** and sources of N output were clarified by chemical statistics and isotopic abundance,and the contribution of N deposition to the N output was quantified by the output coefficient *** N flux of wet deposition was estimated at 18.53 kg N ha^(-1) yr^(-1),and originated mostly from agricultural *** N was significantly accumulated from upper to lower reaches due to frequent human *** changes of riverine N were strongly affected by chemical fertilizer,with the highest concentration in spring and the lowest in *** was mainly derived from chemical fertilizer,accounting for 38.83% of all sources.N concentration in processes of different rainfall events had a hysteresis effect corresponding to the flow *** rainfall events greatly changed DTN(Dissolved total N)outputs:rainstorm caused 91.26 kg DTN output,which was 11 times that of moderate rain(8.46 kg)and 4 times that of heavy rain(20.80 kg).N deposition contributed 19.89% of riverine N output in the *** results can provide theoretical support for the control of N pollution in the TGR area.
