This work aims to investigate the effects of dosing regiments on drug delivery in solid tumors and to validate them with experiments on rats. The lumped parameter models of pharmacokinetics and of drug delivery in tumor were developed to simulate time courses of average drug concentration （Ct） of tumor interstitium in two types of dosing regiments （i.e., single-shot and triple-shot ones）. The two regiments were performed via antitumor drug, hydroxycamptothecin （HCPT）, on rats, to measure the drug concentration in the tumor. The simulations of the drug concentration in the tumor of the two dosing regiments were conducted and compared with the experimental data on rats. The coefficients in the models were investigated. It is concluded that the triple-shot method is more effective than that of single-shot injection. The present lumped-parameter model is quantitatively competent for drug delivery in solid tumor.

With the popularisation of intelligent power,power devices have different shapes,numbers and *** means that the power data has distributional variability,the model learning process cannot achieve sufficient extraction of data features,which seriously affects the accuracy and performance of anomaly ***,this paper proposes a deep learning-based anomaly detection model for power data,which integrates a data alignment enhancement technique based on random sampling and an adaptive feature fusion method leveraging dimension *** at the distribution variability of power data,this paper developed a sliding window-based data adjustment method for this model,which solves the problem of high-dimensional feature noise and low-dimensional missing *** address the problem of insufficient feature fusion,an adaptive feature fusion method based on feature dimension reduction and dictionary learning is proposed to improve the anomaly data detection accuracy of the *** order to verify the effectiveness of the proposed method,we conducted effectiveness comparisons through elimination *** experimental results show that compared with the traditional anomaly detection methods,the method proposed in this paper not only has an advantage in model accuracy,but also reduces the amount of parameter calculation of the model in the process of feature matching and improves the detection speed.

Neurotransmitter-initiated signaling pathway were reported to play an important role in regulating the malignant phenotype of tumor *** cells could exhibit a"neural addiction"property and build up local nerve networks to achieve an enhanced neurotransmitter-initiated signaling through nerve growth factor-mediated *** the dysregulated nervous systems might represent a novel strategy for cancer ***,whether intrahepatic cholangiocarcinoma(ICC)could build its own nerve networks and the role of neurotransmitters in the progression ICC remains largely *** staining and Enzyme-linked immunosorbent assay suggested that IcC cells and the infiltrated nerves could generate a tumor microenvironment rich in acetylcholine that promotes IcC metastasis by inducing epithelial-mesenchymal transition(EMT).Acetylcholine promoted iCC metastasis through interacting with its receptor,alpha 5 nicotine acetylcholine receptor subunits(CHRNA5).Furthermore,acetylcholine/CHRNA5 axis activated GSK3β/β-catenin signaling pathway partially through the influx of Ca^(2+)-mediated activation of Ca/calmodulin-dependent protein kinases(CAMKll).In addition,acetylcholine signaling activation also expanded nerve infiltration through increasing the expression of Brain-Derived Neurotrophic Factor(BDNF),which formed a feedforward acetylcholine-BDNF axis to promote ICC ***93,a small-molecule inhibitor of CAMKIll,significantly inhibited the migration and enhanced the sensitivity to gemcitabine of ICC *** all,Acetylcholine/CHRNA5 axis increased the expression ofβ-catenin to promote the metastasis and resistance to gemcitabine of ICC via CAMKIl/GSK3βsignaling,and the CAMKIl inhibitor KN93 may be an effective therapeutic strategy for combating ICC metastasis.

Background:Bitter taste receptors(Tas2rs)are generally considered to sense various bitter compounds to escape the intake of toxic *** taste receptors have been found to widely express in extraoral tissues and have important physiological functions outside the gustatory system in ***:To investigate the physiological functions of the bitter taste receptor cluster Tas2r106/Tas2r104/Tas2r105/Tas2r114 in lingual and extraoral tissues,multiple Tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for Tas2rs and Gnat3 gene was microinjected into the cytoplasm of the ***,T7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder *** real-time polymerase chain reaction(qRT-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste *** to taste substance was also studied using twobottle preference ***:We successfully produced several Tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 *** results showed that the expression of GNAT3 and PLCB2 was not altered in Tas2rs mutant *** qRT-PCR results revealed the changed expression profile of m Tas2rs gene in taste buds of these mutant *** two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of *** addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB ***:These findings implied that these mutant mice would be a valuable means to understand the biological functions of TAS2Rs in extraoral tissues and investigate bitter compound-induced responses mediated by these TAS2Rs in many extraoral tissues.

A Mathematical Model of Renal Blood Distribution Coupling TGF,MR and Tubular System

Objective:To investigate the relationship between renal blood distribution and the physiological activities of the kidney. Methods:A mathematical model is developed based on Hagan-Poiseuille law and mass transport, coupling mechanics of myogenic response (MR), tubuloglomerular feedback (TGF) and the tubular system in the renal medulla. The model parameters, including the permeability coefficients, the vascular lumen radius and the solute concentration at the inlet of the tubes, are derived from the experimental results. Simulations of the blood and water flow in the loop of Henel, the collecting duct and vas rectum, are carried out by the model of the tubular system in the renal medulla, based on conservations of water and solutes for transmural transport. Then the tubular model is coupled with MR and TGF mechanics. Results:The results predict the dynamics of renal autoregulation on its blood pressure and flow,and the distributions are 88.5% in the cortex, 10.3% in the medulla, and 1.2% at papilla,respectively. The fluid flow and solute concentrations along the tubules and vasa recta are obtained. Conclusion:The present model could assess renal functions qualitatively and quantitatively and provide a methodological approach for clinical research.

BACKGROUND Syphilis is an infectious disease caused by Treponema pallidum that can invade the central nervous system,causing *** cases of anti-N-methyl-Daspartate receptor autoimmune encephalitis(AE)secondary to neurosyphilis have been *** report a neurosyphilis patient with anti-γ-aminobutyric acid-B receptor(GABABR)*** SUMMARY A young man in his 30s who presented with acute epileptic status was admitted to a local *** was diagnosed with neurosyphilis,according to serum and cerebrospinal fluid(CSF)tests for *** 14 d of antiepileptic treatment and anti-Treponema pallidum therapy with penicillin,epilepsy was controlled but serious cognitive impairment,behavioral,and serious psychiatric symptoms were *** was then transferred to our *** Mini-Mental State Examination(MMSE)crude test results showed only 2 *** magnetic resonance imaging revealed significant cerebral atrophy and multiple fluidattenuated inversion recovery high signals in the white matter surrounding both lateral ventricles,left amygdala and bilateral ***-GABABR antibodies were discovered in CSF(1:3.2)and serum(1:100).The patient was diagnosed with neurosyphilis complicated by anti-GABABR AE,and received methylprednisolone and *** treatment,his mental symptoms were *** impairment was significantly improved,with a MMSE of 8 *** anti-GABABR antibody titer decreased to 1:*** patient received methylprednisolone and penicillin after *** months later,the patient’s condition was stable,but the serum anti-GABABR antibody titer was 1:*** This patient with neurosyphilis combined with anti-GABABR encephalitis benefited from immunotherapy.
