The SeqCode,formally called the Code of Nomenclature of Prokaryotes Described from Sequence Data,is a new code of nomenclature in which genome sequences are the nomenclatural types for the names of prokaryotic *** similar to the International Code of Nomenclature of Prokaryotes(ICNP)in structure and rules of priority,it does not require the deposition of type strains in international culture ***,it allows for the formation of permanent names for uncultured prokaryotes whose nearly complete genome sequences have been obtained directly from environmental dna as well as other prokaryotes that cannot be deposited in culture *** the diversity of uncultured prokaryotes greatly exceeds that of readily culturable prokaryotes,the SeqCode is the only code suitable for naming the majority of prokaryotic *** start date of the SeqCode was January 1,2022,and the online Registry(https://***)was created to ensure valid publication of *** SeqCode recognizes all names validly published under the ICNP before *** that date,names validly published under the SeqCode compete with ICNP names for *** a result,species can have only one name,either from the SeqCode or ICNP,enabling effective communication and the creation of unified taxonomies of uncultured and cultured *** SeqCode is administered by the SeqCode Committee,which is comprised of the SeqCode Community and elected administrative *** with an interest in the systematics of prokaryotes is encouraged to join the SeqCode Community and participate in the development of this resource.

Hepatitis b virus （HbV） infection is a global public health problem that concerns 350 million people worldwide. Individuals with chronic hepatitis b （Crib） are at increased risk of developing liver cirrhosis, hepatic de-compensation and hepatocellular carcinoma. To maintain undetectable viral load reduces chronic infection complications. There is no treatment that eradicates HbV infection. Current drugs are expensive, are associated with adverse events, and are of limited efficacy. Current guidelines try to standardize the clinical practice. Nevertheless, controversy remains about management of asymptomatic patients with Crib who are hepatitis b e antigen （HbeAg）-positive with normal alanine aminotransferase, and what is the cut-off value of viral load to distinguish HbeAg- negative Crib patients and inactive carriers. We discuss in detail why dna level alone is not sufficient to begin treatment of Crib.

In this editorial,we comment on the article by Wang et *** manuscript explores the potential synergistic effects of combining zanubrutinib,a novel oral inhibitor of bruton’s tyrosine kinase,with high-dose methotrexate(HD-MTX)as a therapeutic intervention for primary central nervous system lymphoma(PCNSL).The study involves a retrospective analysis of 19 PCNSL patients,highlighting clinicopathological characteristics,treatment outcomes,and genomic *** results indicate the combination’s good tolerance and strong antitumor activity,with an 84.2%overall response *** authors emphasize the potential of zanubrutinib to modulate key genomic features of PCNSL,particularly mutations in myeloid differentiation primary response 88 and cluster of differentiation ***,the study investigates the role of circulating tumor dna in cerebrospinal fluid for disease surveillance and treatment response *** essence,the study provides valuable insights into the potential of combining zanubrutinib with HD-MTX as a frontline therapeutic regimen for *** findings underscore the importance of exploring alternative treatment modalities and monitoring genomic and liquid biopsy markers to optimize patient *** the findings suggest promise,the study’s limitations should be considered,and further research is needed to establish the clinical relevance of this therapeutic approach for PCNSL.

The stimulator of interferon genes(STINg),an integral adaptor protein in the dna-sensing pathway,plays a pivotal role in the innate immune response against ***,it presents a valuable therapeutic target for infectious diseases and *** observed that fangchinoline(Fan),a bis-benzylisoquinoline alkaloid(bbA),effectively impedes the replication of vesicular stomatitis virus(VSV),encephalomyocarditis virus(EMCV),influenza A virus(H1N1),and herpes simplex virus-1(HSV-1)in *** treatment significantly reduced the viral load,attenuated tissue inflammation,and improved survival in a viral sepsis mouse ***,Fan activates the antiviral response in a STINg-dependent manner,leading to increased expression of interferon(IFN)and interferon-stimulated genes(ISgs)for potent antiviral effects in vivo and in ***,Fan interacts with STINg,preventing its degradation and thereby extending the activation of IFN-based antiviral ***,our findings highlight the potential of Fan,which elicits antiviral immunity by suppressing STINg degradation,as a promising candidate for antiviral therapy.

Antifertility effect of ethanolic extract of Amalakyadi churna in male albino mice

Aim: To evaluate the antifertility activity of the ethanolic extract of Amalakyadi churna by oral administration in male albino mice. Methods: The ethanol extract of Amalakyadi churna at the dose of 250 mg/kg and 400 mg/ kg body weight was administered orally for 30 days to adult male mice. On day 31, the mice were sacrificed and the testis and accessory reproductive organs were removed and weighed. The organs were processed for biochemical estimation and histological work. Results: Treatment with Amalakyadi churna resulted in decrease in the weights of testis and accessory reproductive organs. The diameters of testis, seminiferous tubules and Leydig cell nucleus were decreased. The spermatogenic elements, like spermatogonia, spermatocytes and spermatids in the testis were reduced significantly as well as the sperm count in cauda epididymis. There was a significant reduction in the protein, glycogen, dna and RNA contents and the activity of acid phosphatase in the testis of extract treated mice compared with the control. The cholesterol content and the alkaline phophatase activity were increased significantly in treated mice. Conclusion: Amalakyadi churna extract arrests spermatogenesis in male mice without noticeable side effects.

Leishmaniasis is a set of diseases with a worldwide distribution that affects mainly economically underprivileged populations in developing countries. It has a major impact on public health, with a global cost of billions of dollars per year. The treatment and control of leishmaniasis vary according to the Leishmania species involved, which require reliable methods for species identification. Since most of the currently used methods have limitations, there is a need for assays that allow rapid, precise identification of the offending species. Real-time polymerase chain reactions in conjunction with dissociation curve analysis have been used to detect differences in the dna composition of selected genes of Leishmania spp. Kinetoplast dna is the main molecular target used because of its high copy number per parasite, but other targets have also been studied. As part of an effort to establish melting temperature standards for each target gene, we have reviewed the pertinent literature available in public databases, including Pub Med, Web of Science, Sci ELO and LILACS, using the keywords "Leishmania", "leishmaniasis", "realtime PCR", "melting temperature", and "melting curve", alone or in combination. After applying eligibility criteria, 27 articles were selected for analysis. A considerable variation in the methodologies analyzed was found regarding molecular targets, standardization of the methods, reproducibility and specificity. because of this, statistical analysis was not performed. In most cases, the methods were able to differentiate the parasite at the subgenus level or few species regardless of the target chosen.

AIM To study the role of HbV especially Hbx Open Reading Frame (ORF) in the development of hepatocellular carcinoma (HCC).METHODS HbV 3.2kb fragment was retrieved by digesting recombinant plasmid pbR322-2HbV with EcoR Ⅰ, and Hbx 0.59kb fragments by digesting HbV-dna with bamHⅠ and bglⅡ. These fragments were labelled with digoxigenin to get HbV-dna and Hbx-dna probes. HbV-dna was detected in HCC by dot blot and Southern blot hybridization with HbV-dna probe, so the positive specimens in which HbV-dna were integrated were selected. Hbx-dna was subsequently detected in the selected specimens with Hbx-dna *** HbV-dna was detected in 75% HCC, among which integrated type, integrated + free type covered 63.6% and 36.4%. There was no free type. Hbx-dna was detected in 90.5% specimens of integrated *** Hepatocarcinogenesis was highly related to HbV-dna integration, and HbV-dna mainly integrated into chromosome with incomplete virus dna fragments among which Hbx fragment was the predominant one.

大肠杆菌O157∶H7是一种危害性极大的食源性致病菌,为实现大肠杆菌O157∶H7的快速检测,分别采用碱加热裂解法、TritonX-100加热裂解法、溶菌酶加十二烷基硫酸钠(SDS)裂解法、SDS加蛋白酶K裂解法及热裂解法5种方法提取大肠杆菌O157∶H7基因组dna,考察了环介导等温扩增(LAMP)和实时荧光定量聚合酶链式反应(qPCR)扩增法对所提取dna的检测效果,并建立最佳的核酸快速提取样品预处理方法.结果表明,以碱加热裂解预处理提取的大肠杆菌O157∶H7基因组dna为检测样本时,LAMP扩增及qPCR扩增法的检测性能最好,检出限达到10^(1)CFU/mL,与常规商品化试剂盒提取所得菌液基因组dna的检测性能一致.所建立的碱加热裂解样品预处理方法耗时短、成本低,无须进行复杂的核酸提取,操作简单,可用于大肠杆菌O157∶H7的快速检测.
