Diabetes mellitus (DM) is a metabolic disease caused by the absence or dysfunction of insulin;a hormone secreted by the pancreatic beta cell (β-cell) whenever blood glucose exceeds the normal physiological value. The long-term effects of the disease on the body’s organs are one of the leading causes of death in the world. To alleviate this global burden of DM, a number of studies have been conducted to lower blood glucose levels in patients. For genetic and ethical reasons, humans are far from being appropriate subjects in such investigations and the use of animal models has therefore been the way forward. Streptozotocin (STZ) is a glucosamine-nitrosourea compound that selectively destroys β-cells and has been widely used to induce Type I diabetes in several animal species. Recent literature has shown that a non-diabetic dose of STZ, combined with a high-fat diet (HFD), can mimic Type II diabetes. Yet, researchers seldom provide data to corroborate the high sensitivity of STZ on these animal models. In addition, there are few reports of potentially fatal effects of the use of STZ as a supplement in obese HFD animals when attempting to induce Type II diabetes. The present review article highlights the parameters that could be at the origin of the extreme sensitivity and vulnerability of obese animals to STZ.

The prevalence of type 2 diabetes mellitus(T2DM)is increasing rapidly *** of the limited success of generic interventions,the focus of the disease study has shifted toward personalized strategies,particularly in the early stages of the *** Chinese medicine(TCM)is based on a systems view combined with personalized strategies and has improved our knowledge of personalized *** a systems biology perspective,the understanding of personalized diagnostics can be improved to yield a biochemical basis for such strategies;for example,metabolomics can be used in combination with other system-based diagnostic methods such as ultra-weak photon emission(UPE).In this study,we investigated the feasibility of using plasma metabolomics obtained from 44 pre-T2DM subjects to stratify the following TCM-based subtypes:Qi-Yin deficiency,Qi-Yin deficiency with dampness,and Qi-Yin deficiency with *** studied the relationship between plasma metabolomics and UPE with respect to TCM-based subtyping in order to obtain biochemical information for further interpreting disease *** component analysis of plasma metabolites revealed differences among the TCM-based pre-T2DM *** high levels of lipids(e.g.,cholesterol esters and triglycerides)were important discriminators of two of the three subtypes and may be associated with a higher risk of cardiovascular *** metabolomics data indicate that the lipid profile is an essential component captured by UPE with respect to stratifying subtypes of *** results suggest that metabolic differences exist among different TCM-based subtypes of pre-T2DM,and profiling plasma metabolites can be used to discriminate among these *** metabolomics thus provides biochemical insights into system-based UPE measurements.

Evaluating File Formats for Long-term Preservation

National and international publishers have been depositing digital publications at the National Library of the Netherlands (KB) since 2003. Until recently, most of these publications were deposited in the Portable Document Format. New projects, for example the Web archiving project, force the KB to handle more heterogeneous material. Therefore, the KB has developed a quantifiable file format risk assessment method. This method can be used to define digital preservation strategies for specific file formats. The choice for a specific file format at creation time or later in the life cycle of a digital object influences the long-term access to the digital object. The evaluation method contains seven sustainability criteria for file formats that are weighed for importance. There seems to be consensus on the sustainability criteria. However, as the weighing of these criteria is connected to an institution's policy, the KB wonders whether agreement on the relative importance of the criteria can be reached at all. With this paper, the KB hopes to inspire other cultural heritage institutions to define their own quantifiable file format evaluation method.

Background: There have been concerns regarding the mental wellbeing of emergency medical service staff, particularly when faced with multiple trauma exposures. This study aimed to describe a cross-sectional view of the mental wellbeing status of emergency medical service workers, as well as to comment on the usefulness of an electronic survey tool to reach staff that is distributed across large geographical areas. Methods: The data presented here are drawn from an anonymous, voluntary, electronic survey, made available via the internet, to emergency medical service workers in the Western Cape province of South Africa. This paper reports on four domain expressions of mental wellbeing, namely disordered mood, problematic substance use, post-traumatic stress disorder, and domestic discord. Results: Elevated rates were reported by this sample, across all four distress domains, i.e. mood disorder = 12.3%;problematic substance use = 14.9%;post-traumatic stress disorder = 11.9%;and domestic discord = 11.9%. These rates were generally similar to data from comparable emergency worker groups, and much higher than local population estimates. Conclusions: The data support earlier findings that emergency medical work is associated with increased risk for symptoms of psychological distress, which present a strong case for regular organizational monitoring of employees’ mental wellbeing. In this regard, a brief, internet delivered, survey-type tool showed promise for screening employees, in order to guide further streaming of distressed individuals towards appropriate support services. Further work is required to develop clear mechanisms for referral and intervention in order to optimize a confidential and supportive monitoring program.

Photodynamic therapy （PDT） is a tumor treatment modality in which a tumor- localized photosensitizer is excited with light, which results in local production of reactive oxygen species, destruction of tumor vasculature, tumor hypoxia, tumor cell death, and induction of an anti-tumor immune response. However, pre-existing tumor hypoxia may desensitize tumors to PDT by activating the hypoxia-inducible factor 1 （HIF-1） survival pathway. Therefore, we hypothesized that inhibition of HIF-1 with acriflavine （ACF） would exacerbate cell death in human epidermoid carcinoma （A431） cells. PDT of A431 tumor cells was per- formed using newly developed and optimized PEgylated cationic liposomes containing the photosensitizer zinc phthalocyanine （ZnPC）. Molecular docking revealed that ACF binds to the dimerization domain of HIF-la, and confocal microscopy confirmed translocation of ACF from the cytosol to the nucleus under hypoxia. HIF-1 was stabilized in hypoxic, but not normoxic, A431 cells following PDT. Inhibition of HIF-1 with ACF increased the extent of PDT-induced cell death under hypoxic conditions and reduced the expression of the HIF-1 target genes VEgF, PTgS2, and EDN1. Moreover, co-encapsulation of ACF in the aqueous core of ZnPC-containing liposomes yielded an adjuvant effect on PDT efficacy that was comparable to non-encapsulated ACF. In conclusion, HIF-1 contributes to A431 tumor cell survival following PDT with liposomal ZnPC. Inhibition of HIF-1 with free or liposomal ACF improves PDT efficacy.

With the availability of distributed generation (Dg), clusters that can autonomously manage their energy profile are emerging in the power grid. These autonomous clusters manage their load profiles by orchestrating their energy resources, such as Dg, storage, flexible energy consuming appliances, etc. The performance of such an autonomous cluster depends on the composition of its energy resources. In this paper, we study how the performance of a cluster is affected by adding energy resources such as generating units, storage systems or consuming appliances. First, we characterize the energy resources by parameters that describe their relevant properties. Afterwards, we describe a comprehensive set of performance indicators of a cluster that capture the economical, environmental, and social aspects. We present a model that shows how the energy resources influence the performance indicators of the cluster. We have tested our model with a case study, revealing its effectiveness to evaluate the value added by an energy resource to a cluster.

当前,学术界对组织知识转移的研究方兴未艾,但我们对其前因后果的理解依然相当含糊。尽管已经有人对组织知识转移的文献进行了概念性的定性回顾,但目前还没有人对已有的定量研究结果进行总结。为了实现这个目标,本文首先利用元分析方法考察了知识、组织和网络等不同前因变量是如何影响组织知识转移的;然后,对现有的关于知识转移及其后果之间关系的研究进行了整合;最后,论证了组织间和组织内的情境因素、知识转移的方向性以及测量特性对所研究关系的调节作用。通过收集和归纳现有研究结论,本文不仅对组织知识转移的途径和结果提出了新见解,还指明了未来的研究方向。
