In marker-assisted breeding for bacterial blight(BB) resistance in rice, three major resistance genes, viz., Xa21, xa13 and xa5, are routinely deployed either singly or in combinations. As efficient and functional markers are yet to be developed for xa13 and xa5, we have developed simple PCR-based functional markers for both the genes. For xa13, we designed a functional PCR-based marker, xa13-prom targeting the In Del polymorphism in the promoter of candidate gene Os8N3 located on chromosome 8 of rice. With respect to xa5, a multiplex-PCR based functional marker system, named xa5 FM, consisting of two sets of primer pairs targeting the 2-bp functional nucleotide polymorphism in the exon II of the gene TFIIA5(candidate for xa5), has been developed. Both xa13-prom and xa5 FM can differentiate the resistant and susceptible alleles for xa13 and xa5, respectively, in a co-dominant fashion. Using these two functional markers along with the already reported functional PCR-based marker for Xa21(p TA248), we designed a single-tube multiplex PCR based assay for simultaneous detection of all the three major resistance genes and demonstrated the utility of the multiplex marker system in a segregating population.

Objective:To evaluate antimicrobial and antimycobacterial activity of three seaweeds[gelidiella acerosa(***),Turbinaria conoides(***)and Sargassum wightii(***)]from gulf of ***:Three seaweeds ***,*** and *** were collected from gulf of Mannar,South-East Coast of *** extraction of the selected seaweeds was done using hexane and *** extracts were tested for antibacterial activity against four bacterial strains(Escherichia coli,Pseudomonas aeruginosa,Bacillus subtilis and Staphylococcus aureus)by disc diffusion *** active extracts of *** were tested for antimycobacterial activity by luciferase reporter phage assay at two different concentrations(100μg/mL and 500μg/mL).Results:Among the tested seaweeds,ethanol extract of *** showed maximum antibacterial activity against Bacillus subtilis and Staphylococcus aureus followed by hexane extract at concentration of 1 mg/*** remaining two seaweeds(*** and ***)did not show any *** ethanol extract of *** exhibited maximum antimycobacterial activity(87.33%)followed by hexane extract(74.68%)against Mycobacterium tuberculosis H37Rv at 500μg/***:Ethanol extract of *** showed both antibacterial and antimycobacterial *** photochemical studies are needed to identify the active antimycobacterial agents.

BACKgROUND There is increasing interest in transplanting patients with hepatocellularcarcinoma (HCC) with tumors greater than 5 cm (Milan criteria).AIM To investigate possible prognostically-useful factors for liver transplantation inHCC patients with large *** In this clinical study, 50 patients with HCC who were transplanted at our LiverTransplant Center between April 2006 and August 2019 and had tumors greaterthan 6 cm maximum diameter were retrospectively analyzed. Their survival andfull clinical characteristics were examined, with respect to serum alphafetoprotein(AFP) and gamma glutamyl transpeptidase (ggT) levels. Kaplan-Meier survival estimates were used to determine overall survival and disease-freesurvival in these patients. The inclusion criterion was evidence of HCC. Exclusioncriteria were the presence of macroscopic portal vein thrombosis or metastasisand a follow-up period of less than 90 *** Using receiver operating characteristic curve (ROC) analysis, cutoff values of AFP200 ng/mL and ggT 104 IU/L were identified and used in this *** longer overall survival (OS) and disease-free-survival (DFS) were found in patients who had lower values of either parameter, compared withhigher values. Even greater differences in survival were found when the 2parameters were combined. Two tumor size bands were identified, in searchingfor the limits of this approach with larger tumors, namely 6-10 cm and > 10 *** parameters in the 6-10 cm band reflected 5-year OS of 76.2% inpatients with low AFP plus low ggT vs 0% for all other groups. BACKgROUND Polymorphisms of human leukocyte antigen(HLA)genes are suggested to increase the risk of gastric cancer(gC).AIM To investigate the HLA allele frequencies of patients with gC relative to a control group in terms of CagA+multiple(≥2)EPIYA-C *** The patient group comprised 94 patients[44 gC and 50 duodenal ulcer(DU)patients],and the control group comprised 86 individuals[(50 non-ulcer dyspepsia patients and 36 people with asymptomatic Helicobacter pylori(***)].Polymerase chain reaction was performed for the amplification of the *** cagA gene and typing of EPIYA *** sequence-specific oligonucleotide(SSO)typing was performed using Lifecodes SSO typing kits(HLA-A,HLA-B HLA-C,HLA-DRB1,and HLA-DQA1-B1 kits).RESULTS The comparison of gC cases in terms of CagA+multiple(≥2)EPIYA-C repeats showed that only the HLA-DQB1*06 allele[odds ratio(OR):0.37,P=0.036]was significantly lower,but significance was lost after correction(Pc=0.1845).The HLA-DQA1*01 allele had a high ratio in gC cases with multiple EPIYA-C repeats,but this was not significant in the univariate *** compared allele frequencies in the DU cases alone and in gC and DU cases together using the same criterion,and none of the HLA alleles were significantly associated with gC or ***,none of the alleles were detected as independent risk factors after the multivariate *** the other hand,in a multivariate logistic regression with no discriminative criterion,HLA-DQA1*01(OR=1.848),HLA-DQB1*06(OR=1.821)and HLA-A*02(OR=1.579)alleles were detected as independent risk factors for gC and *** None of the HLA alleles were detected as independent risk factors in terms of CagA+multiple EPIYA-C ***,HLA-DQA1*01,HLA-DQB1*0601,and HLA-A*2 were independent risk factors with no criterion in the multivariate *** suggest that the association of these alleles with gastric malignancies is not specifically related to cagA and multiple EPIYA C repeats.
