Three-dimensional reconstruction and comparison of vacuolar membranes in response to viral infection

The vacuole is a unique plant organelle that plays an important role in maintaining cellular homeostasis under various environmental stress conditions. However, the effects of biotic stress on vacuole structure has not been examined using three-dimensional(3D) visualization. Here, we performed 3D electron tomography to compare the ultrastructural changes in the vacuole during infection with different viruses. The 3D models revealed that vacuoles are remodeled in cells infected with cucumber mosaic virus(CMV) or tobacco necrosis virus A Chinese isolate(TNV-AC), resulting in the formation of spherules at the periphery of the vacuole. These spherules contain neck-like channels that connect their interior with the cytosol. Confocal microscopy of CMV replication proteins 1 a and 2 a and TNV-AC auxiliary replication protein p23 showed that all of these proteins localize to the *** microscopy revealed that the expression of these replication proteins alone is sufficient to induce spherule formation on the tonoplast, suggesting that these proteins play prominent roles in inducing vacuolar membrane remodeling. This is the first report of the 3D structures of viral replication factories built on the tonoplasts. These findings contribute to our understanding of vacuole biogenesis under normal conditions and during assembly of plant(+) RNA virus replication complexes.

Effect of herpesvirus infection on pancreatic duct cell secretion

AIM： To examine the effect of acute infection caused by herpesvirus （pseudorabies virus, pRV） on pancreatic ductal secretion. METHODS： The virulent Ba-Dupgreen （BDg） and non- virulent ka-RREp01acgfp （KEg） genetically modified strains of pRV were used in this study and both of them contain the gene for green fluorescent protein （gFp）. Small intra/ interlobular ducts were infected with BDg virus （10^7 pFU/mL for 6 h） or with KEg virus （10^10 pFU/mL for 6 h）, while non-infected ducts were incubated only with the culture media. The ducts were then cultured for a further 18 h. The rate of HCO3 secretion [base efflux -λ（B）] was determined from the buffering capacity of the cells and the initial rate of intracellular acidification （1） after sudden blockage of basolateral base loaders with dihydro-4,4,- diisothiocyanatostilbene-2,2,-disulfonic acid （500 μmol/L） and amiloride （200 μmol/L）, and （2） after alkali loading the ducts by exposure to NH4Cl. All the experiments were performed in HCO3-buffered Ringer solution at 37 ℃ （n = 5 ducts for each experimental condition）. Viral structural proteins were visualized by immunohistochemistry. Virallyencoded gFp and immunofluorescence signals were recorded by a confocal laser scanning microscope.RESULTS： The BDg virus infected the majority of accessible cells of the duct as judged by the appearance of gFp and viral anUgens in the ductal cells. KEg virus caused a similarly high efficiency of infection. After blockage of basolateral base loaders, BDg infection significantly elevated -λB-） 24 h after the infection, compared to the non-infected group. However, KEg infection did not modify -λB-）. After alkali loading the ducts, -λB-） was significantly elevated in the BDg group compared to the control group 24 h after the infection. As we found with the inhibitor stop method, no change was observed in the group KEg compared to the non-infected group. Menstrual cycle characteristics as an indicator of fertility outcomes:evidence from prospective birth cohort study in China

Atrophy of the left dorsolateral prefrontal cortex is associated with poor performance in verbal fluency in elderly poststroke women

This study aimed to investigate the association between atrophy in the prefrontal cortex with executive function and verbal fluency in elderly male and female patients poststroke. Thirty elderly female patients with non-aphasic ischemic stroke aged -〉 60 years and 30 age-matched non-aphasic male patients with ischemic stroke were recruited. Automatic magnetic resonance imaging segmentation was used to assess the volume of the whole prefrontal cortex, along with its subdivisions： anterior cingulate cortex, orbitofrontal cortex and dorsolateral prefrontal cortex. The Semantic Verbal Fluency Test was administered at 3 and 15 months poststroke. At 3 months poststroke, left dorsolateral prefrontal cortex volume was significantly correlated with Verbal Fluency Test score in female patients only （partial coefficient = 0.453, p = 0.045）, after controlling for age, education, diabetes, neurological deficit, white matter lesions volume, as well as the location and volume of infarcts. At 15 months poststroke, there remained a significant association between the left dorsolateral prefrontal cortex volume and Verbal Fluency Test （partial coefficient = 0.661, p = 0.001） and between the left prefrontal cortex volume and Verbal Fluency Test （partial coefficient = 0.573, p = 0.004） in female patients after the same adjustments. These findings indicate that atrophy of the left dorsolateral prefrontal cortex contributes to the impairment of verbal fluency in elderly female patients with stroke. Sex differences may be present in the neuropsychological mechanisms of verbal fluency impairment in patients with stroke.

Serum Mac-2 binding protein glycosylation isomer level predicts hepatocellular carcinoma development in E-negative chronic hepatitis B patients

BACKgROUND Liver cirrhosis is a major risk factor for hepatocellular carcinoma(HCC)development in chronic hepatitis B(CHB). Serum Mac-2 binding protein glycosylation isomer(M2 Bpgi) is a novel serological marker for fibrosis. The role of M2 Bpgi in prediction of HCC is *** To examine the role of serum M2 Bpgi in predicting HCC development in hepatitis B e antigen(HBeAg)-negative *** Treatment-naive CHB patients with documented spontaneous HBeAg seroconversion were recruited. Serum M2 Bpgi was measured at baseline(within3 years from HBeAg seroconversion), at 5 years and 10 years after HBeAg seroconversion and expressed as cut-off index(COI). Multivariate cox regression was performed to identify predictors for HCC development. ROC analysis was used to determine the cut-off value of M2 *** Among 207 patients(57% male, median age at HBeAg seroconversion 40 years old) with median follow-up of 13.1(11.8-15.5) years, the cumulative incidence of HCC at 15 years was 7%. Median M2 Bpgi levels were significantly higher in patients with HCC compared to those without HCC(baseline: 1.39 COI vs 0.38 COI, p p p = 0.001). Multivariate analysis revealed age at HBeAg seroconversion[odds ratio(OR) = 1.196, 95% confidence interval(CI): 1.034-1.382, p = 0.016] and baseline M2 Bpgi(OR = 4.666, 95%CI: 1.296-16.802, p = 0.018) were significant factors predictive of HCC. Using a cut-off value of 0.68 COI, baseline M2 Bpgi yielded AUROC of 0.883 with 91.7% sensitivity and 80.8% *** High serum M2 Bpgi within 3 years after HBeAg seroconversion was a strong predictor for subsequent HCC development in treatment-naive HBeAg-negative CHB patients.

profiles and removal efficiency of polycyclic aromatic hydrocarbons by two different types of sewage treatment plants in Hong Kong

Sewage discharge could be a major source of polycyclic aromatic hydrocarbons （pAHs） in the coastal waters. Stonecutters Island and Shatin Sewage Treatment Works （SCISTW and STSTW） in Hong Kong, adopted chemically enhanced primary treatment and biological treatment, respectively. This study aimed at （1） determining the removal efficiencies of pAHs, （2） comparing the capabilities in removing pAHs, and （3） characterizing the profile of each individual pAHs, in the two sewage treatment plants （STps）. Quantification of 16 pAHs was conducted by a gas Chromatography. The concentrations of total pAHs decreased gradually along the treatment processes （from 301 ± 255 and 307 ± 217 ns/L to 14.9 ± 12.1 and 63.3 ± 54.1 ng/L in STSTW and SCISTW, respectively）. It was noted that STSTW was more capable in removing total pAHs than SCISTW with average total removal effidency 94.4% ± 4.12% us. 79.2% ± 7.48% （p 〈 0.05）. The removal of pAHs was probably due to sorption in particular matter, confirmed by the higher distribution coefficient of individual and total pAHs in solid samples （dewatered sludge contained 92.5% and 74.7% of total pAHs in SCISTW and STSTW, respectively） than liquid samples （final efffluent-total contained 7.53% and 25.3% of total pAHs in STSTW and SCISTW, respectively）. Despite the impressive capability of STSTW and SCISTW in removing pAHs, there was still a considerable amount of total pAHs （1.85 and 39.3 ks/year, respectively for the two STps） being discharged into Hone Kon coastal waters, which would be an environmental concern.
