Pharmacological targeting cGAS/STING/NF-κB axis by tryptanthrin induces microglia polarization toward M2 phenotype and promotes functional recovery in a mouse model of spinal cord injury

The M1/M2 phenotypic shift of microglia after spinal cord injury plays an important role in the regulation of neuroinflammation during the secondary injury phase of spinal cord *** of shifting microglia polarization from M1(neurotoxic and proinflammatory type)to M2(neuroprotective and anti-inflammatory type)after spinal cord injury appears to be *** possesses an anti-inflammatory biological ***,its roles and the underlying molecular mechanisms in spinal cord injury remain *** this study,we found that tryptanthrin inhibited microglia-derived inflammation by promoting polarization to the M2 phenotype in *** promoted M2 polarization through inactivating the cGAS/STING/NF-κB ***,we found that targeting the cGAS/STING/NF-κB pathway with tryptanthrin shifted microglia from the M1 to M2 phenotype after spinal cord injury,inhibited neuronal loss,and promoted tissue repair and functional recovery in a mouse model of spinal cord ***,using a conditional co-culture system,we found that microglia treated with tryptanthrin suppressed endoplasmic reticulum stress-related neuronal *** together,these results suggest that by targeting the cGAS/STING/NF-κB axis,tryptanthrin attenuates microglia-derived neuroinflammation and promotes functional recovery after spinal cord injury through shifting microglia polarization to the M2 phenotype.

磁刺激下Fe_(3)O_(4)@ZIF-8纳米颗粒影响骨髓间充质干细胞的成骨分化

背景:骨髓间充质干细胞在组织工程和骨再生中具有重要作用,然而如何促进骨髓间充质干细胞的成骨分化仍然是一个挑战。目的:探讨磁刺激下Fe_(3)O_(4)@ZIF-8纳米颗粒对骨髓间充质干细胞成骨分化的影响。方法:采用水热法合成沸石咪唑酯骨架(ZIF-8),采用一锅法合成具有磁性的Fe_(3)O_(4)@ZIF-8纳米颗粒(材料制备中分别加入2.5,5,10,20μg的Fe_(3)O_(4)),通过扫描电镜、X射线光电子能谱、X射线衍射、振动样品磁强计检测等对Fe_(3)O_(4)@ZIF-8纳米颗粒进行表征,筛选出合适的材料进行后续实验。提取4周龄SD大鼠骨髓间充质干细胞,分别与不同质量浓度(25,50,75,100,125μg/mL)的Fe_(3)O_(4)@ZIF-8纳米颗粒溶液共培养,CCK-8法检测细胞增殖,筛选出最佳的材料溶液质量浓度;筛选出材料溶液质量浓度后,施加磁刺激(磁场强度分别为0,50,100,150 MT),CCK-8法检测细胞增殖,筛选出最佳的磁场强度与Fe_(3)O_(4)@ZIF-8纳米颗粒,用于骨髓间充质干细胞诱导分化实验。将SD大鼠骨髓间充质干细胞分别与ZIF-8、Fe_(3)O_(4)@ZIF-8、Fe_(3)O_(4)@ZIF-8(磁场干预)纳米颗粒溶液共培养,以单独培养的细胞为空白对照,成脂诱导后进行油红O染色,成骨诱导后进行碱性磷酸酶、茜素红染色与Runx2蛋白质量浓度检测。结果与结论:(1)扫描电镜下可见Fe_(3)O_(4)@ZIF-8纳米颗粒呈现十二面体结构,随着材料中Fe_(3)O_(4)含量的增加,纳米颗粒的粒径增大,选择粒径约250 nm(该粒径下的纳米颗粒功能性及生物安全性较稳定)的Fe_(3)O_(4)@ZIF-8纳米颗粒(材料制备中分别加入5,10μg的Fe_(3)O_(4))进行后续实验。(2)CCK-8检测结果显示,在100MT磁场作用下,50μg/mL的Fe_(3)O_(4)@ZIF-8纳米颗粒(材料制备中加入10μg的Fe_(3)O_(4))能够显著促进骨髓间充质干细胞的增殖,选择该条件下的纳米颗粒进行骨髓间充质干细胞成骨诱导分化实验。(3)成骨诱导后,Fe_(3)O_(4)@ZIF-8(磁场干预)组骨髓间充质干细胞的碱性磷酸酶活性、细胞外基质矿化程度与Runx2蛋白质量浓度均高于其他3组(P<0.05);成脂诱导后,Fe_(3)O_(4)@ZIF-8(磁场干预)组骨髓间充质干细胞的脂滴形成少于其他3组(P<0.05)。(4)结果表明,在特定的磁场条件下Fe_(3)O_(4)@ZIF-8纳米颗粒可以促进骨髓间充质干细胞的成骨分化。

Factorization method for inverse obstacle scattering problem in three-dimensional planar acoustic waveguides

In this paper, we consider the inverse scattering problem of reconstructing a bounded obstacle in a three-dimensional planar waveguide from the scattered near-field data measured on a finite cylindrical surface containing the obstacle and corresponding to infinitely many incident point sources also placed on the measurement surface. The obstacle is allowed to be an impenetrable scatterer or a penetrable scatterer. We establish the validity of the factorization method with the nearfield data to characterize the obstacle in the planar waveguide by constructing an outgoing-to-incoming operator which is an integral operator defined on the measurement surface with the kernel given in terms of an infinite series.

Lead halide perovskite nanocrystals(NCs)exhibit excellent optoelectronic performance and have drawn great interests in the fields of biological imaging and ***,the poor stability of CsPbX_(3)(X=Cl,Br,I)in water is still a challenge to hinder their practical *** this work,a facile strategy has been developed for aqueous synthesis of CsPbX_(3) nanocrystals,in which L-glutamic acid(LGlu)has been used to replace oleic acid in the synthetic *** from the synergic effects of L-Glu and oleylamine(OAm),CsPbBr_(3)nanocrystals(L-Glu/OAm-CsPbBr_(3)NCs)with high water stability have been directly prepared under a mild condition at room temperature in water,facilitated by the process of crystal phase transformation from Cs_(4)PbBr_(6) to CsPbBr_(3).L-Glu/OAm-CsPbBr_(3)NCs exhibit a high quantum yield of 85%and a narrow full width at half maximum of 16 nm,demonstrating their efficient luminescence in ***,L-Glu on the surface have contributed greatly to an acidic environment and passivation of surface defects,improving the water stability and dispersibility of CsPbBr_(3)***,L-Glu/OAm-CsPbBr_(3)NCs exhibit great biocompatibility due to the presence of L-Glu,resulting in their good performance for HeLa cell ***,we propose a facile and effective method to prepare CsPbBr_(3)nanocrystals with excellent water stability by using L-Glu and OAm as cooperated ligands and expand their application in cell imaging.

Study of the material removal mechanism of glass-ceramics based on consecutive incremental loading in ductile-regime grinding

Glass-ceramics have many excellent properties and are widely used in various fields. During the grinding process,the workpiece surface is typically subject to material removal by grit of incremental heights, which has rarely been the focus of research. As such, it is necessary to study the material removal mechanism of glass-ceramics under consecutive incremental loading, which more closely reflects the actual grinding process. In this paper,to analyze the plastic deformation and residual stress of lithium aluminosilicate(LAS) glass-ceramics, a finite element model is established based on the Drucker–Prager yield criterion for ductile regimes. A nano-scratch test was also conducted and the test results show that both the residual depth and residual stress increase with an increase in the number of increments, and that consecutive incremental loading promotes the plastic deformation of glass-ceramics and increases the residual stress of the material in the ductile-regime process. These findings provide guidance for achieving higher dimensional accuracy in the actual grinding of glass-ceramics parts.

Current-carrying friction in carbon coated ball bearing

In this work,we proposed a method for coating the whole surfaces of bearing balls uniformly by carbon film with a rotatable ball *** studied the carbon/carbon friction with a self-designed currentcarrying ball bearing friction test system.A notable and instant friction force drop of 28%and significant carbon film wear alleviation were found when currents were *** using TEM-,SEM-,and EDS-analysis,special carbon stacks with a mixture of large wear particles and oxide were found in the wear areas under current applied *** elucidated the current-carrying friction mechanisms as follows:(1)wear particles formation;(2)wear particles charged by tribomicroplasma;(3)formation of surface passivated carbon stacks under electric force;(4)sliding between passivated carbon *** work may facilitate the development of novel solid-lubricated ball bearings and lay some foundations for current-carrying rolling friction.

Approach for Obtaining Material Mechanical Properties in Local Region of Structure Based on Accurate Analysis of Micro-indentation Test

The hot or cold processing would induce the change and the inhomogeneous of the material mechanical properties in the local processing region of the structure,and it is difficult to obtain the specific mechanical properties in these regions by using the traditional material tensile *** accurately get actual material mechanical properties in the local region of structure,a micro-indentation test system incorporated by an electronic universal material test device has been *** indenter displacement sensor and a group of special micro-indenter assemblies are estab-lished.A numerical indentation inversion analysis method by using ABAQUS software is also proposed in this *** on the above test system and analysis platform,an approach to obtaining material mechanical properties in the local region of structures is proposed and *** ball indentation test is performed and combined with the energy method by using various changed mechanical properties of 316L austenitic stainless steel under differ-ent *** investigated results indicate that the material mechanical properties and the micro-indentation morphological changes have evidently *** with the tensile test results,the deviations of material mechanical parameters,such as hardness H,the hardening exponent n,the yield strength σy and others are within 5%obtained through the indentation test and the finite element *** provides an effective and convenient method for obtaining the actual material mechanical properties in the local processing region of the structure.

Multiple optimal solutions to a sort of nonlinear optimization problem

The optimization problem is considered in which the objective function is pseudolinear（both pseudoconvex and pseudoconcave） and the constraints are linear. The general expression for the optimal solutions to the problem is derived with the representation theorem of polyhedral sets, and the uniqueness condition of the optimal solution and the computational procedures to determine all optimal solutions （if the uniqueness condition is not satisfied ） are provided. Finally, an illustrative example is also given.

A Controlled Phase Gate with Nitrogen-Vacancy Centers in Nanocrystal Coupled to a Silica Microsphere Cavity

We show how a controlled phase gate, induced by adiabatic passage of dark states, can be implemented with the nitrogen-vacancy defects in nanocrystal coupled to the optical whispering gallery mode in a silica microsphere cavity. The gate presented is robust to the fluctuations of the experimental parameters compared to the dynamical and geometric phases gates. The feasibility of this scheme is characterized by exact numerical simulations that incorporate various sources of experiment noise. The results demonstrate the practicality by way of current experimental technologies.
