AIM: To elucidate risk factors for survival of elderly acute myeloid leukemia(AML) patients in a real-world practice by observational study. METHODS: We conducted a population-based study in 213 adult and elderly AML patients(127 males and 86 females) in Kagawa Prefecture, Japan. To construct this cohort, we gathered all data for patients diagnosed with AML at 7 hospitals in Kagawa between 2006 and 2010. The primary end point was overall survival(OS) after AML diagnosis. Unadjusted Kaplan-Meier survival plots were used to determine OS in the overall cohort. Multivariate analysis was used to determine the independent adverse prognostic factors for OS, with the covariates of interest including age, gender, race/ethnicity, CCI, education, median income, metropolitan statistical area size and history of myelodysplastic syndrome.RESULTS: The average population of Kagawa during the study period was 992489, and the incidence of AML was 4.26 per 100000 person-years. A total of 197 patients with non-acute promyelocytic leukemia(non-APL)(119 males and 78 females) were also included. The median age of non-APL patients was 70 years(average 67, range 24-95). The 5-year OS rate was 21.1%. Subsequent analysis by age group showed that the survival rate declined with age; the 5-year OS rates of non-APL patients younger than 64 years, 65-74 years, and older than 75 years were 41.5%, 14.1%, and 8.9%, respectively. Multivariate analysis revealed that unfavorable risk karyotype, older age, poor performance status(PS)(3-4), lack of induction chemotherapy, and antecedent haematological disease were independent prognostic predictors. In the subgroup analysis, we also found that older patients with non-APL had lower complete remission rates and higher early death rates than younger patients, irrespective of PS. However, intensive chemotherapy was a significant predictor for longer survival not only in the patients < 75 years of age, but also in those over 75 with PS 0-2. Extracellular vesicles(EVs),especially small EVs(sEVs)derived from liver cells,have been the focus of much attention in the normal physiology and pathogenesis of various diseases affecting the liver.sEVs are approximately 100 nm in size,enclosed within lipid bilayers,and are very stable.The lipids,proteins,and nucleic acids,including miRNAs,contained within these vesicles are known to play important roles in intercellular communication.This mini-review summarizes the application of sEVs.First,liver diseases and the related diagnostic markers are described,and the current active status of miRNA research in diagnosis of hepatocellular carcinoma(HCC)is reported.Second,the biodistribution and pharmacokinetics of sEVs are described,and the liver is highlighted as the organ with the highest accumulation of sEVs.Third,the relationship between sEVs and the pathogenesis of liver disorders is described with emphesis on the current active status of miRNA research in HCC recurrence and survival.Finally,the possibility of future therapy using sEVs from mesenchymal stem(stromal)cells for cirrhosis and other diseases is described.

Microbial depolymerization processes of xenobiotic polymers are discussed. A mathematical model is formulated and inverse problems for a time factor and a molecular factor of a degradation rate are described. Experimental outcomes are introduced in inverse analyses. Once the time factor and the molecular factor are obtained, the microbial depolymerization process is simu-lated. Numerical techniques are illustrated and numerical results are presented.

A new assay method for natural killer (NK) cell activity was established using quantitative RT-PCR (RT-qPCR) to determine the gene expression of granzyme B (GzmB) and perforin 1 (Prf1). The RT-qPCR method was compared to a conventional cytotoxic assay. Upregulated expression of GzmB and Prf1 mRNA and enhanced cytotoxic activity were observed in splenocytes from lipopolysaccharide (LPS)-treated rats. A high correlation, R2 = 0.71, was observed between the gene expression of GzmB and the cytotoxic activity of splenocytes from these rats, indicating that GzmB RT-qPCR is a reliable alternative method to assess NK cell activity/activation. Remarkably, 12.6- to 59.7-fold upregulation of GzmB mRNA expression was observed in leukocytes, the spleen, and splenocytes from LPS-injected rats. Its upregulation appeared to be dose-dependent on the LPS concentration in the range of 0.01 - 0.1 mg/kg. Whereas, only 1.3- to 1.9-fold increase of cytotoxic activity was detected in splenocytes from the rats treated with LPS in the same range. From these, it is evident that, to assess NK cell activity/activation, the GzmB RT-qPCR method is highly sensitive compared with the conventional cytological assay. Furthermore, this GzmB RT-qPCR method is advantageous, as it does not require freshly prepared splenocytes and cell culture procedures. The convenience of GzmB RT-qPCR enables the use of whole blood, leukocytes, the spleen, and/or their frozen samples to evaluate NK cell activity/activation.

Para-probiotics are ghost probiotics and consist of non-viable microbial cells. Here, a genus of acetic acid bacteria (AAB) was lyophilized to produce a para-probiotic preparation of non-viable (NV) AAB. The NV-AAB orally administered for 49 days ameliorated antigen-induced sneezing and plasma antigen-specific IgE in an ovalbumin-sensitized allergic mouse model. Our findings indicated that NV-AAB, as a para-probiotic functional food, exhibits an anti-allergic effect. The anti-allergic mechanism of NV-AAB was atypical because of the near-absence of an effect on Th1 and Th2 balance in splenocytes. NV-AAB will be a unique para-probiotic food material with an anti-allergic activity.

This study demonstrates mathematical analysis of biodegradation processes of xenobiotic polymers. A model for microbial population is based on the fact that growth rate of microorganisms is proportional to the microbial population and consumption rate of parts of carbon sources. The model is paired with a model for weight distribution. Those models lead to inverse problems for a molecular factor and a time factor of degradation rate. Solution of the inverse problems allows us to simulate the biodegra-dation process.

Purpose/Aim: We aimed to investigate the effect of monocular blur on the binocular visual field. Materials and Methods: A total of 13 healthy young volunteers participated in this study. The mean subjective refractive error of the dominant eye (DE) was -3.33 ± 1.65D, and the non-dominant eye (NDE) was -3.15 ± 2.84D. The DE was determined by using the hole-in-the-card test. The visual field was examined by the Humphrey Visual Field Analyzer using the 30-2 SITA Standard program. The visual field was measured while wearing soft contact lens under three conditions;① both eyes: near vision correction;② DE: near vision correction +3.00D added, NDE: near vision correction;and ③ DE: near vision correction, NDE: near vision correction +3.00D added. The foveal threshold, mean deviation (MD), and pattern standard deviation (PSD) values were investigated. Results: The foveal threshold value (dB) at ①, ②, and ③ was 41.2, 37.8, and 38.1, respectively. The values at ② and ③ were both significantly lower than that at ① (p Conclusion: These results suggest that monocular blur reduced the sensitivity within the binocular visual field.

The crystalline fullerene C60 particles were formed and immobilized on poly(dimethylsiloxane) (PDMS) substrates under the various discharge conditions by an ink-jet method, and investigated for the reactive oxygen species (ROS) generation property under visible light irradiation. The particles were synthesized by discharging a toluene solution dissolved C60 and poly(methyl methacrylate) (PMMA) with the ink-jet spotting system. The ROS generation was evaluated by comparisons of the fluorescence intensities measured for the formed particles under green laser irradiation and in a dark room using fluorescent dyes, 2’,7’-dichlorofluorescein diacetate and dihydroethidium. The results of transmission electron microscope (TEM) observation showed that the formed particles consisted of crystalline C60. The optimal ink-jet discharge conditions for synthesizing the particles to generate more ROS were found. In the case of the optimal conditions, the structure in which the needle-like particles were three-dimensionally formed was confirmed. The surface area of the crystalline C60 particles was calculated using the SEM observation results, and it was suggested that when the needle-like finer particles were three-dimensionally formed under the optimal conditions, increasing the surface area lead to an increase in the ROS generation amount.

Practical Use of Acoustic Rhinometry for Quantitative Assessment of Nasal Obstruction in an Allergic Model Using Macaque Monkeys

Nasal obstruction, a primary symptom of allergic rhinitis, results in breathing difficulties and decreases quality of life in patients. Quantitative methods are required for diagnosis of nasal obstruction to accurately assess drug efficacy, not only in patients but also in animal models. Acoustic rhinometry is a non-invasive technique to measure nasal airway patency using acoustic reflections. To date, little information on its application in non-human primates (NHP) has been accumulated. Macaque monkeys are a practical NHP model because of their high similarity with humans in their biomedical nature, including genomic and immunologic events and morphology. We established an allergic rhinitis model of cynomolgusmacaque (Macaca fascicularis) sensitized with Japanese cedar pollen and applied acoustic rhinometry to measure nasal congestion. The extent of nasal congestion followed by allergen challenge was successfully quantified as the minimal cross-sectional area (MCA) of the nasal passage for the first time in an NHP allergic model. We also applied this acoustic technique to evaluate the efficacy of an antihistamine drug, fexofenadine. The acoustic assessment achieved quantitative evaluation of the efficacy of fexofenadine on nasal congestion in a macaque allergic model.
