Long-term dexamethasone treatment diminishes store-operated Ca^(2+) entry in salivary acinar cells

Corticosteroids are used in the treatment of many diseases; however, they also induce various side effects. Dexamethasone is one of the most potent corticosteroids, and it has been reported to induce the side effect of impaired salivary gland function. This study aimed to evaluate the effects of dexamethasone on mouse submandibular gland function to gain insight into the mechanism of dexamethasone-induced salivary hypofunction. The muscarinic agonist carbachol(CCh) induced salivary secretion and was not affected by short-term dexamethasone treatment but was decreased following long-term dexamethasone administration. The expression levels of the membrane proteins Na^+-K^+-2 Cl-cotransporter, transmembrane member 16 A, and aquaporin 5 were comparable between the control and long-term dexamethasone treatment groups. The CCh-induced increase in calcium concentration was significantly lower in the presence of extracellular Ca^(2+) in the long-term dexamethasone treatment group compared to that in the control group. Furthermore, CCh-induced salivation in the absence of extracellular Ca^(2+) and Ca^(2+) ionophore A23187-induced salivation was comparable between the control and long-term dexamethasone treatment ***, salivation induced by the Ca^(2+) -ATPase inhibitor thapsigargin was diminished in the long-term dexamethasone treatment group. In summary, these results demonstrate that short-term dexamethasone treatment did not impair salivary gland function, whereas long-term dexamethasone treatment diminished store-operated Ca^(2+) entry, resulting in hyposalivation in mouse submandibular glands.

Conjecture with water and rheological control for subducting slab in the mantle transition zone

Seismic observations have shown structural variation near the base of the mantle transition zone （MTZ） where subducted cold slabs, as visualized with high seismic speed anomalies （HSSAs）, flatten to form stagnant slabs or sink further into the lower mantle. The different slab behaviors were also accompanied by variation of the ＂660 kin＂ discontinuity depths and low viscosity layers （LVLs） beneath the MTZ that are suggested by geoid inversion studies. We address that deep water transport by subducted slabs and dehydration from hydrous slabs could affect the physical properties of mantle minerals and govern slab dynamics. A systematic series of three-dimensional numerical simulation has been conducted to examine the effects of viscosity reduction or contrast between slab materials on slab behaviors near the base of the MTZ. We found that the viscosity reduction of subducted crustal material leads to a sepa- ration of crustal material from the slab main body and its transient stagnation in the MTZ. The once trapped crustal materials in the MTZ eventually sink into the lower mantle within 20 30 My from the start of the plate subduction. The results suggest crustal material recycle in the whole mantle that is consistent with evidence from mantle geochemistry as opposed to a two-layer mantle convection model. Because of the smaller capacity of water content in lower mantle minerals than in MTZ minerals, dehydration should occur at the phase transformation depth, ~660 kin. The variation of the disconti- nuity depths and highly localized low seismic speed anomaly （LSSA） zones observed from seismic P waveforms in a relatively high frequency band （~ 1 Hz） support the hypothesis of dehydration from hydrous slabs at the phase boundary. The LSSAs which correspond to dehydration induced fluids are likely to be very local, given very small hydrogen （H＋） diffusivity associated with subducted slabs. Changes in microbial community composition in the leaf litter of successional communities after volcanic eruptions of Mount Usu, northern Japan

Changes in the fungal and bacterial biomass and community structure in litter after the volcanic eruptions of Mount Usu, northern Japan were investigated using a chronosequence approach, which is widely used for analyzing vegetation succession. The vegetation changed from bare ground （10 years after the eruptions） with little plant cover and poor soil to monotonic grassland dominated by Polygonum sachalinense with undeveloped soil （33 years） and then to deciduous broad-leaved forest dominated by Populus maximowiczii with diverse species composition and well-developed soil （100 years）. At three chronosequential sites, we evaluated the compositions of phospholipid fatty acids （PLFAs）, carbon （C） and nitrogen （N） contents and the isotope ratios of C （δ13C） and N （δ15N） in the litter of two dominant species, Polygonum sachalinense and Populus maximowiezii. The C/N ratio, δ13C and δ15N in the litter of these two species were higher in the forest than that in the bare ground and grassland. The PLFAs gradually increased from the bare ground to the forest, showing that microbial biomass increased with the development of the soil and/or vegetation. The fungi-to-bacteria ratio of PLFA was constant at 5.3 ± 1.4 in all three sites, suggesting that fungi were predominant. A canonical correspondence analysis suggested that the PLFA comoosition was related to the successional ages and the developing soil properties （P 〈 0.05, ANOSIM）. The chrono- sequential analysis effectively detected the successional changes in both microbial and plant communities.

The Literary Imagination of the White Pagoda and Dynastic Change in Early Ming Hangzhou

This article examines the literary imaginations of the White Pagoda and demonstrates a shift in its representation from a metaphor for the Song court＇s fate to a fantastic site for the subjugation of unworldly beings. In the late thirteenth century, the Yuan-appointed Tibetan Buddhist monk Yang Lianzhenjia exhumed the imperial mausoleums of the defeated Southern Song, built the White Pagoda on the site of the old Southern Song palace in Hangzhou, and interred the exhumed bones under it. Enthusiastic Song loyalists thus considered the White Pagoda to be a symbol of a humiliating past in which the Mongol Yuan dynasty occupied the south. Meanwhile, Qu You, an early-Ming writer from Hangzhou, began to imagine that the White Pagoda served to pacify the innocent, lonely dead who died during the Song-Yuan social disturbance. Investigating the discourse of the early Ming literati in regard to the pagoda site and the supernatural in early Ming Hangzhou leads to the conclusion that the literary imagination of the White Pagoda would have also contributed to the development of the White Snake Legend, where a white serpent spirit was subdued under Thunder Peak Pagoda in Hangzhou.

Acute and delayed psychiatric sequelae among patients hospitalised with COVID-19:a cohort study using LIFE study data

Process of Decision-Making of Anti-Cancer Treatment in Elderly Patients with Advanced Lung Cancer

The aim of this study was to identify the decision-making process of anti-cancer treatment in elderly patients with advanced lung cancer. Semi-structured interviews were conducted with 17 patients aged 70 and above to collect data on their feelings and thoughts from the time of diagnosis till they made treatment decisions. The data was analyzed using the Modified Grounded Theory Approach that was modified by Prof. Yasuhito Kinoshita. We found the process to be composed of six categories. The results showed that elderly patients with advanced lung cancer were panicked over unavoidable death when informed of stage IV lung cancer and offered suggestions about anti-cancer treatment. However, trying to accept the situation where death is imminent, patients reconsidered having cancer in older age and recognized a desire to survive even in older age. This process diverged into two paths: one group of patients changed their ideas from radical to life-prolonging treatment by recognizing a desire to survive even in older age and then made anti-cancer treatment decisions by carefully choosing treatment that would allow to carry on their usual way of life;the other group of patients made treatment decisions by carefully choosing physicians to whom they could entrust their life in older age. These findings suggest that it is important for nurses to: 1) encourage patients to talk in order to have them think about what they have valued and want to value in the rest of their life;2) assess if patients prefer to decide on their own by carefully choosing treatment or entrust treatment decisions to physicians;and 3) offer support according to patients’ preferences.

Screening Method for 23 Alkaloids in Human Serum Using LC/MS/MS with a Pentafluorophenyl Column in Dynamic Multiple Reaction Monitoring Mode

Alkaloids are nitrogen-containing organic compounds, generally basic, and found in plants, fungi, and bacteria. Some alkaloids are used in medicine, but some compounds are highly toxic. Accidental ingestion, homicide, and suicide have occurred due to plants containing alkaloids. The identification of toxic components in biological samples is important for the diagnosis and/or treatment of poisoning cases in forensic and emergency medicine. Alkaloids have a wide variety of structures, such as isoquinoline alkaloid, indole alkaloid, tropane alkaloid, and diterpene alkaloid;therefore, there are few reports of simultaneous analysis methods. We have established a method for the simultaneous analysis of 23 alkaloids in human serum with a liquid chromatograph-tandem mass spectrometer (LC/MS/MS). A liquid-liquid extraction which was modified from the first step of the QuEChERS AOAC method was used for serum pretreatment. The separation of the compounds was performed using a pentafluorophenyl (PFP) column, CAPCELL CORE PFP (2.1 mm I.D. × 100 mm, 2.7 μm) in gradient mode. Mobile phase A consisted of 10 mM ammonium formate and 0.1% formic acid in ultrapure water, and mobile phase B was 10 mM ammonium formate and 0.1% formic acid in methanol. Simultaneous analysis was performed in dynamic multiple reaction monitoring mode. The separation of 23 alkaloids was satisfactory, as PFP columns exhibited different retention behaviors than alkyl phase columns. The PFP column effectively retained polar aromatic compounds;therefore, it was suitable for alkaloid analysis. The validated method was applied to a forensic case of aconite poisoning. The present method was useful in LC/MS/MS screening for 23 alkaloids in human serum.

p53 negativity, CDC25B positivity, and metallothionein negativity are predictors of a response of esophageal squamous cell carcinoma to chemoradiotherapy

AIM： Esophageal squamous cell carcinoma is generally sensitive to chemoradiotherapy （CRT）, but some cases are not. Using a retrospective analysis, we aimed to identify the predictors of the response by esophageal squamous cell carcinoma to definitive CRT. METHODS： The intensities of expression of p53, Ki67, Bci-2, Bax, olclin D1, VEGF, CDC25B, and metallothionein （MT） were evaluated immunohistochemically in the biopsy specimens obtained before CRT, and the intensities of their expression were tested for correlations with the clinical effects of CRT. RESULTS： The esophageal squamous cell carcinomas with negative p53, positive CDC25B, and negative MT expression were found to be significantly more sensitive to CRT. In addition, p53 positivity and CDC25B positivity respomd well to CRT. CONCLUSION： Esophageal squamous cell carcinomas with negative p53,positive CDC25B, and negative MT expressions respond well to CRT. A Study of Indian Government Policy on Production and Processing of Cotton and Its Implications

India is a major player in international cotton markets as it is the world＇s second largest cotton producer, consumer and exporter in 2009-2010. In this context, this paper assesses the competitiveness of Indian cotton producers and potential implications for India as a competitor in the world cotton market. This paper developed an updated estimate of the costs of cotton production in India and developed representative farm models for cotton production in three important cotton production states （Gujarat, Maharashtra and Andhra Pradesh） of India. In this research, Rapid Rural Appraisal （RRA） methodology has been adopted to collect information for developing representative farm models. These models are further subjected to stochastic simulation to understand the impact of government policies like subsidies to various inputs on farm level profitability and ultimately on the competitiveness of Indian cotton in international markets. This paper also analyzed the impact of national fiber policy of the government of India on the cotton processing sector in India. The results demonstrate that the net income of the cotton farmers will decrease considerably without the presence of fertilizer subsidies. The study also concludes that if the objectives of the national fiber policy are fulfilled, India will export more of value added cotton products like textiles and garments rather than raw cotton.

Correlation of Various Foods Intakes and Plasma Levels of Omega Fatty Acids in Healthy Japanese Old Men

Background: Trans fatty acids are considered to impair health and some ω fatty acids are protective against atherosclerosis and diabetes mellitus. Trans fatty acids are said to be formed by the partial hydrogenation of vegetable oils. Some amounts are produced in digestive organs of ruminants and present in dairy products or meat. It is important how much these intaken fatty acids influence their plasma levels. Methods: Plasma levels of fatty acids including transforms of healthy old men are measured by gas chromatography and correlations between various foods intakes and plasma levels of trans fatty acids, and ω fatty acids are examined. Results: Intake of fish resulted in increase in plasma levels of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) but intake of meat such as beef, cow and chicken meat did not increase plasma levels of arachidonic acid (AA). Intakes of oils increased plasma levels of dihomo-g-linolenic acid significantly and AA to some extent. Conclusion: Plasma levels of EPA and DHA increased upon intakes of fish in Japanese old men. Oil intake but not meat intake increased DGLA significantly. These results may explain low incidence of cardiovascular diseases in Japanese people compared with American people whose plasma levels of DHA and EPA are lower.
